    Notizen: A method for the synthesis of well-defined poly(alkyl vinyl ether-2-ethyl-2-oxazoline) diblock copolymers with hydrolytically stable block linkages has been developed. Monofunctional poly(alkyl vinyl ether) oligomers with nearly Poisson molecular weight distributions were prepared via a living cationic polymerization method using chloroethyl vinyl ether together with HI/ZnI2 as the initiating system and lithium borohydride as the termination reagent. Using the resultant chloroethyl ether functional oligomers in combination with sodium iodide as macroinitiators, 2-ethyl-2-oxazoline was polymerized in chlorobenzene/NMP to afford diblock copolymers. A series of poly(methyl vinyl ether-2-ethyl-2-oxazoline) diblock materials were found to have polydispersities of ≈ 1.3-1.4 and are microphase separated as indicated by two Tg's in their DSC thermograms. These copolymers are presently being used as model materials to study fundamental parameters important for steric stabilization of dispersions in polar media. © 1993 John Wiley & Sons, Inc.

    Notizen: In this work, the separation of carbon dioxide from methane by reactive liquid membranes consisting of the secondary amines, diethanolamine, and diisopropanolamine in polyethylene glycol with an average molecular weight of 400 was studied. A mathematical model was developed to describe the transport process. This model employs the zwitterion mechanism for the CO2 amine reaction kinetics. Membrane flux experiments utilizing a flat plate device were carried out to verify the model predictions. It was found that large permeabilities and separations were achieved for low CO2 feed concentrations. The results also show that DEA based solutions give better separation than DIPA based solutions.

    Notizen: Equations introduced here identify measurement biases and process leaks, when gross errors exist in measured process variables and the variance-covariance matrix of the measurements, Σ, is unknown. Σ is estimated by the sample variance, S, using process data.For an unknown Σ, the global test statistic is the well-known Hotelling T2 statistic. Its power function has a noncentral F-distribution. For component tests used for specific identification of measurement biases and nodal leaks, two tests are presented with Σ unknown. The first test is independent of the number of component tests, k, and is given by a statistic with an F-distribution. The second test depends on k and has a student t-distribution. The power functions for both component tests are provided. Process examples and a Monte Carlo simulation study presented demonstrate the use and performance of these statistical equations in identifying biases and process leaks.

    Notizen: The solution of the stream function-vorticity equations is developed using a multigrid method to improve efficiency in a finite-element p-method. The equations are discretized using 2-D Lagrange finite elements (linear, quadratic and cubic) and solved in iteratively decoupled form using successive approximation and continuation methods. The comparative performance of several multigrid and direct matrix solvers is investigated with respect to convergence characteristics and time performance on the Cray Y-MP.

    Notizen: This study reports the effects of Simplex bone cement powder (BC) on the proliferation and production of bone resorbing factors in vitro by human adherent monocytes/macrophages. Adherent peripheral blood cells were isolated from seven healthy individuals and exposed to a dispersion of BC powder (1 mg/mL), phytohemagglutinin (PHA, 40 μg/mL), or medium alone at different periods of cell incubation (days 0-2, 0-7, 5-7, or 10-12). Cell proliferation was quantified by incorporation of 3H-thymidine uptake. Culture supernatants were evaluated for levels of interleukin 1-like activity (IL-1) by murine thymocyte proliferation assay, prostaglandin E2 (PGE2) by radioimmunoassay, lysosomal enzyme activity (N-acetyl-β-D-glucosaminidase and β-glucuronidase) using fluorometry, and collagen and casein degrading activity using radioactive substrates. Human adherent peripheral blood cells showed a proliferative response to PHA that coincided with cell maturation; BC did not inhibit PHA-induced cell proliferation of either adherent or nonadherent blood cells, indicating the non-toxic nature of these particles at the concentrations tested. BC stimulated increased release of the lysosomal enzyme N-acetyl-β-D-glucosaminidase; the levels of PGE2, IL-1, collagenase, and caseinase were unchanged. © 1993 John Wiley & Sons, Inc.

    Notizen: When cultured on a basement membrane substratum, endothelial cells undergo a rapid series of morphological and functional changes which result in the formation of histotypic tube-like structures, a process which mimics in vivo angiogenesis. Since this process is probably dependent on several cell adhesion and cell signaling phenomena, we examined the roles of integrins and protein kinase C in endothelial cell cord formation. Polyclonal antisera directed against the entire vitronectin (αvβ3) and fibronectin (α5β1) receptors inhibited cord formation. Subunit-specific monoclonal antibodies to αv, β3, and β1 integrin subunits inhibited cord formation, while monoclonal antibodies to α3 did not, which implicated the vitronectin receptor, and not the fibronectin receptor, in vascular formation. Protein kinase C inhibitors inhibited cord formation, while phorbol 12-myristate 13-acetate (PMA) caused endothelial cells to form longer cords. Since the vitronectin receptor has been shown to be phosphorylated in an in vitro system by protein kinase C, the possible functional link between the vitronectin receptor and protein kinase C during cellular morphogenesis was examined. The vitronectin receptor was more highly phosphorylated in cord-forming endothelial cells on basement membrane than in monolayer cells on vitronectin. Furthermore, this phosphorylation was inhibited by protein kinase C inhibitors, and PMA was required to induce vitronectin receptor phosphorylation in endothelial cells cultured on vitronectin. Colocalization studies were also performed using antisera to the vitronectin receptor and antibodies to protein kinase C. Although no strict colocalization was found, protein kinase C was localized in the cytoskeleton of endothelial cells initially plated on basement membrane or on vitronectin, and it translocated to the plasma membrane of C-shaped cord-forming cells on basement membrane. Thus, both the vitronectin receptor and protein kinase C play a role in in vitro cord formation. © 1993 Wiley-Liss, Inc.

    Notizen: Retinal pigment epithelial cells, which form one aspect of the blood-retinal barrier, control the access of blood-borne components such as diferric transferrin to the neural retina. It has recently been shown that RPE cells remove iron from diferric transferrin in a low pH compartment and subsequently release it in a low molecular weight form that can be chelated by apo-transferrin (Hunt and Davis: J. Cell Physiol. 152:102-110, 1992). It is now shown that photoreceptor cells can bind diferric transferrin to receptors on their inner segments. Moreover, polymerase chain reaction and in situ hybridization show that cells of the neural retina, particularly photoreceptors, make apo-transferrin. © 1993 Wiley-Liss, Inc.

    Notizen: Oncogenesis is manifested as uncontrolled cellular proliferation and in some situations a failure of normal differentiation in the transformed cell. This has led to speculation that the normal role of proto-oncogenes during development may be to mediate the relationship between proliferation and differentiation. The advent of gene targeting in ES cells allows the role oncogenes in development to be tested directly. Two recent studies have examined the phenotype of N-myc mutant mice generated by gene targeting(1,2). In both reports, the mutation is an embryonic lethal at 11.5 days of gestation confirming a critical role for this proto-oncogene in development and the inability of other members of the myc family to substitute functionally for N-myc. Although the phenotypes are similar in general outline, the two reports differ in the specifics of the morphological and histological abnormalities identified. The disparity may result from the mutation created, the genetic background of the mutant mice or the criteria used to determine abnormalities. Assuredly, there is valuable information to be gained about N-myc function from these mutant mice. However, these reports make it clear that morphological and histological abnormalities in N-myc mutant mice serve as a starting point rather than as an endpoint. The challenge now is to link the defect at the cellular level to the abnormalities at the physiological level.
