    Description: Mesenchymal stem cells (MSCs) seeded on specific carrier materials are a promising source for the repair of traumatic cartilage injuries. The best supportive carrier material has not yet been determined. As natural components of cartilage’s extracellular matrix, hyaluronic acid and collagen are the focus of biomaterial research. In order to optimize chondrogenic support, we investigated three different scaffold compositions of a hyaluronic acid (HA)-gelatin based biomaterial. Methods: Human MSCs (hMSCs) were seeded under vacuum on composite scaffolds of three different HA-gelatin ratios and cultured in chondrogenic medium for 21 days. Cell-scaffold constructs were assessed at different time points for cell viability, gene expression patterns, production of cartilage-specific extracellular matrix (ECM) and for (immuno-)histological appearance. The intrinsic transforming growth factor beta (TGF-beta) uptake of empty scaffolds was evaluated by determination of the TGF-beta concentrations in the medium over time. Results: No significant differences were found for cell seeding densities and cell viability. hMSCs seeded on scaffolds with higher ratios of HA showed better cartilage-like differentiation in all evaluated parameters. TGF-beta uptake did not differ between empty scaffolds. Conclusion: Higher ratios of HA support the chondrogenic differentiation of hMSCs seeded on a HA-gelatin composite scaffold.

    Description: A combination of mesenchymal stem cells (MSCs) and scaffolds seems to be a promising approach for meniscus repair. To facilitate the search for an appropriate scaffold material a reliable and objective in vitro testing system is essential. This paper introduces a new scoring for this purpose and analyzes a hyaluronic acid (HA) gelatin composite scaffold and a polyurethane scaffold in combination with MSCs for tissue engineering of meniscus. The pore quality and interconnectivity of pores of a HA gelatin composite scaffold and a polyurethane scaffold were analyzed by surface photography and Berliner-Blau-BSA-solution vacuum filling. Further the two scaffold materials were vacuum-filled with human MSCs and analyzed by histology and immunohistochemistry after 21 days in chondrogenic media to determine cell distribution and cell survival as well as proteoglycan production, collagen type I and II content. The polyurethane scaffold showed better results than the hyaluronic acid gelatin composite scaffold, with signs of central necrosis in the HA gelatin composite scaffolds. The polyurethane scaffold showed good porosity, excellent pore interconnectivity, good cell distribution and cell survival, as well as an extensive content of proteoglycans and collagen type II. The polyurethane scaffold seems to be a promising biomaterial for a mesenchymal stem cell-based tissue engineering approach for meniscal repair. The new score could be applied as a new standard for in vitro scaffold testing.

    Description: A striking feature of the Indian Ocean is a distinct geoid low south of India, pointing to a regionally anomalous mantle density structure. Equally prominent are rapid plate convergence rate variations between India and SE Asia, particularly in Late Cretaceous/Paleocene times. Both observations are linked to the central Neo-Tethys Ocean subduction history, for which competing scenarios have been proposed. Here we evaluate three alternative reconstructions by assimilating their associated time-dependent velocity fields in global high-resolution geodynamic Earth models, allowing us to predict the resulting seismic mantle heterogeneity and geoid signal. Our analysis reveals that a geoid low similar to the one observed develops naturally when a long-lived back-arc basin south of Eurasia's paleo-margin is assumed. A quantitative comparison to seismic tomography further supports this model. In contrast, reconstructions assuming a single northward dipping subduction zone along Eurasia's margin or models incorporating a temporary southward dipping intra-oceanic subduction zone cannot sufficiently reproduce geoid and seismic observations.

    Description: Dealing with the threat of anthropogenic climate change has been a challenge for policy makers for a long time. In recent years, the problems posed by climate change and solutions proposed to mitigate its effects have been framed by lexical ‘carbon compounds’, such as carbon footprint or carbon trading and by one dominant metaphor, the market metaphor. Through a detailed content analysis of industry and press coverage from 1985 to the present, this paper examines the fate of one important lexical compound in this context, namely low carbon , which can be used as an adjective or a noun. Over the last two decades this lexical compound moved across and between three discourses, the steel industry, the car industry and what one might call the climate change industry. Using insights from ecolinguistics and the sociology of expectations, the paper discusses how the lexical compound low carbon in general and the metaphor low carbon future in particular came to prominence in policy discourses, especially in the UK, and how they were used to frame expectations of a prosperous low carbon future , while sidelining deeper social and cultural reflections on climate change mitigation. ©2011 Springer Science+Business Media B.V.

    Notes: Abstract Data are presented on the biochemical findings in several intermal organs from an Egyptian mummy with a 14C-dating of approximately 950 B.C. By use of radio immunoassay systems and gas chromatography/mass spectrometry, significant amounts of various drugs were detected in internal organs (lung, liver, stomach, intestines) as well as in hair, bone, skin/muscle and tendon. These analyses revealed a significant deposition of tetrahydrocannabinol (THC), nicotine (and its metabolite cotinine) and cocaine in the tissue from the mummy. The concentration profiles additionally provide evidence for the preferential ways of consumption: Thus, the highest levels of THC in lung specimens point to an inhalation of this drug — as it has been assumed from known ritual smoking ceremonies —, while nicotine and cocaine containing drugs showed their highest concentrations in the intestines and liver, so that they seem to have been consumed perorally. Furthermore, a histopathological examination of the internal organ tissues revealed some evidence for the underlying disease and the probable cause of death. Thus, a severe and presumably recurrent intravital pulmonary bleeding, most obviously due to a parasitosis affecting the lung, was observed.
