Notes: Abstract The locus (RP1) for one form of autosomal dominant retinitis pigmentosa (adRP) was mapped on chromosome 8q11-q22 between D8S589 and D8S285, which are about 8 cM apart, by linkage analysis in an extended family ascertained in the USA. We have studied a multigeneration Australian family with adRP and found close linkage without recombination between the disease locus and D8S591, D8S566, and D8S166 (Zmax = 1.137– 4.650 at θ = 0.00), all mapped in the region known to harbor RP1. Assuming that the mutation of the same gene is responsible for the disease in both families, the analysis of multiply informative meioses in the American and Australian families places the adRP locus between D8S601 and D8S285, which reduces the critical region to about 4 cM, corresponding to approximately 4 Mb, which is completely covered by a yeast artificial chromosome contig assembled recently.

Notes: The case documents an eventful year in the history of 3M, a company that until November 1995 had been considered a paragon of corporate consistency. The particular emphasis of this case is on the spin-off or demerger of 3M's data storage and imaging businesses into a new company called Imation. Although the decision to demerge was taken in a global context, the detailed comments and reflections of participants involved in this process are drawn from Europe and, in particular, from the UK. The case makes extensive use of quotes from these managers in order to bring to life decisions and events. This case explores the strategy behind 3M's decision to demerge, and considers the myriad of structural and cultural issues arising from the decision. Finally, the case examines the results of the demerger and assesses the position of the fledgling company.

Notes: Abstract A new viability assay for Cryptosporidium and Eimeria sporozoites is described. It involves the use of both acridine orange and bis-benzimide and is more rapid, easier and less subjective than procedures used previously. The assay has been used to investigate the effects of respiratory inhibitors and pH on the sporozoites of C. parvum, C. muris and E. tenella. Neither cyanide nor azide reduced the viability of C. parvum or E. tenella, whereas they had some effect on C. muris. This latter organism, an intracellular parasite of stomach epithelial cells, also differed from the other two in being able to survive pH 2 for as long as 1 h.

Notes: Abstract The bradyzoite and tachyzoite forms of Toxoplasma gondii, purified from infected animals, were analysed for their activities of phosphofructokinase, pyruvate kinase, lactate dehydrogenase, NAD+- and NADH-linked isocitrate dehydrogenases, and succinic dehydrogenase. Both developmental stages contained high activities of phosphofructokinase (specific for pyrophosphate rather than ATP), pyruvate kinase and lactate dehydrogenase, suggesting that energy metabolism in both forms may centre around a high glycolytic flux linked to lactate production. The markedly higher activity of the latter two enzymes in bradyzoites suggests that lactate production is particularly important in this developmental form. NAD+-specific isocitrate dehydrogenase was not detectable in either stage of the parasite (and proved useful as a measure of the purity of the bradyzoite preparation), whereas both parasite forms contained low activities of NADP+-linked isocitrate dehydrogenase. The results are consistent with the bradyzoites lacking a functional TCA cycle and respiratory chain and are suggestive of a lack of susceptibility of this developmental stage to atovaquone.

Notes: From ancient times, the need for control and the ability to gain it by developing tools, language and ideas has distinguished the human species. More recently, empowerment has been used as a way of ensuring members of an organization have sufficient control. This not only boosts trust and morale but enables people to take risks and respond to competitive challenges from every level of the organization. Essential measures for successful empowerment include passing on information on all aspects of the organization's performance, providing training and using these to encourage either individual leadership or collective leadership as part of a team. Empowerment will not work unless we address the human need for self-control.

Notes: Employee involvement is essential to improving pollution control. Organizations like Dow Chemical have been able to use employee involvement to reduce pollution. It begins by understanding, as Dow does, that "If pollution is a cost to you (my competitor) and a cost to me, I win if my costs are lower". Controlling these costs requires people to think about it - a lot. Everyone must accept ownership for pollution. All the pieces must fit together. Unfortunately, even the most advanced companies are still struggling with these issues. An anonymous survey was sent to some of the top pollution-reducing companies in the world. Responses to the questionnaire that focused on a wide range of issues often proved surprising to disturbing. Executives in charge sometimes felt they had employee involvement but few of the operational conditions for it to exist. Dow Chemical provides a rare example of being able to connect all the pieces, through its three keys.

Notes: Claims that every successful corporate culture has to have a unifying factor which makes the company distinctive. Looks at American Express, which, it suggests, has a central culture of focusing on the customer. Examines how American Express places its focus on the customer and the benefits of doing so. Concludes that by empowering its employees and encouraging them, a firm will not have trouble in developing good employee relations.
