    Notizen: During the IML-2 space shuttle mission, the RAMSES instrument was operated in the Spacelab module. This continuous-flow electrophoresis device performs separation and purification of protein solutions on a preparative scale. Samples containing artificial mixtures of pure proteins were used to test the capabilities of the device, and useful separations were obtained for proteins having a mobility difference of only 3 × 10-9 m2·V-1·s-1. Operating conditions that cannot be applied on earth were explored for two different sample concentrations, one of which is too high to allow treatment on earth. It agrees well with a previously published numerical model in that the main cause of loss in resolution in this process is the electrohydrodynamic spreading of the protein filaments.

    Notizen: The separation of proteins by free-flow zone electrophoresis is generally impaired by a number of secondary effects which cause spreading of the protein streams with resultant loss in resolution. A strategy is outlined, based on experimental observations and numerical modeling, which allows the operating conditions and separation chamber dimensions to be chosen so as to obtain complete separation of two proteins of known mobility. This approach takes into account some dispersive phenomena such as molecular diffusion, electroosmosis and residence time gradients. In some cases, the right operating conditions cannot be achieved on earth and electrophoresis separations in microgravity may then be justified.

    Notizen: To estimate the feasibility of isoelectric focusing in a continuous mode, the conditions for obtaining stable and linear pH gradients were first investigated, using a mixture of carrier ampholytes. Then the operating parameters allowing the isoelectric focusing of bovine serum albumin as model protein were determined. A theoretical approach was used to find conditions minimizing the electrohydrodynamic effect, mainly responsible for protein dispersion, for the separation of bovine serum albumin and α-lactalbumin from bovine milk.

    Notizen: Free-flow zone electrophoresis may be used to purify biological samples, due to differences in electrophoretic mobility, in absence of a matrix - most frequently a gel - thus enabling the biological integrity of even fragile molecules to be preserved. However, the process is more complicated than its principle suggests due to different transport phenomena interfering with electrophoretic migration, with the resultant separation depending both on separation effects and dispersive phenomena. The physical origin of the main effects involved was identified. Mathematical expressions were proposed to estimate the influence of the crescent effect and electrohydrodynamics on the process. In this paper, these equations are used to determine the minimum difference in electrophoretic mobility required for a separation to be achieved with respect to the processing parameters. A methodology is proposed which defines the conditions under which the difference in electrophoretic mobilities equals that calculated when considering the influence of dispersive phenomena. Optimized separations of the whey proteins lactoferrin and albumin, known to interact strongly, and the purification of a monoclonal antibody from a mouse ascitic fluid illustrate the approach.

    Beschreibung: Economic development of transition and developed countries is associated with increasingly unhealthy dietary habits among low-income population segments. Drawing on Ulrich Beck’s sociological theory of risk society, the present research note calls attention to the positive relation between national economic development and food risks that result in the rise of food-related diseases and healthcare costs. On this basis, we argue that the knowledge-intensive agribusiness may translate Cochrane’s technological treadmill into Beck’s risk treadmill that shifts a growing share of food-related healthcare costs from producers toward consumers, state, and the healthcare system. This argument motivates a novel research program dealing with the “food risk treadmill” that emerges in response to modern farming and agribusiness practices. Awareness of the food risk treadmill may help to streamline the development of agricultural science and to prevent it from being excessively dominated by the agricultural and food industry.

    Beschreibung: In consequence of strong changes in general economic conditions, adjustments in the agricultural sector can be expected. To date, however, there are only few policy impact analyses on agricultural investments in a dynamic-stochastic context. The objective of this paper is to develop a real options market model which allows the impact assessment of different political schemes. The model combines genetic algorithms and stochastic simulation. Simulations of the model show that investment subsidies and production ceilings are preferable to price floors because the welfare is less reduced for a given stimulation of the willingness to invest.

    Beschreibung: [1]  Widespread cropland abandonment occurred after the collapse of socialism across the former Soviet Union, but the rates and spatial patterns of abandoned lands are not well known. As a result, the potential of this region to contribute to global food production and estimates of the carbon sink developing on currently idle lands are highly uncertain. We developed a spatial allocation model that distributes yearly and sub-national sown area statistics to the most agriculturally suitable plots. This approach resulted in new, high-resolution (1 km 2 ) annual time series of cropland and abandoned lands in European Russia, Ukraine and Belarus from 1990 to 2009. A quantitative validation of the cropland map confirms the reliability of this dataset, especially for the most important agricultural areas of the study region. Overall, we found a total of 87 Mha of cropland and 31 Mha of abandoned cropland in European Russia, Ukraine and Belarus combined, suggesting that abandonment has been severely underestimated in the past. The abandonment rates were highest in European Russia. Feeding our new map dataset into the dynamic vegetation model LPJmL revealed that cropland abandonment resulted in a net carbon sink of 470 TgC for 1990 to 2009. Carbon sequestration was generally slow in the early years after abandonment, but carbon uptake increased significantly after approximately 10 years. Recultivation of older abandoned lands would be associated with high carbon emissions and lead to substantial amounts of carbon not being sequestered in vegetation formations currently developing on idle croplands. Our spatially and temporally explicit cropland abandonment data improve the estimation of trade-offs involved in reclaiming abandoned croplands and thus in increasing agricultural production in this globally important agricultural region.
