    Notizen: Abstract This paper gives a quantitative description of the viscoelastic properties of aqueous solutions of entangled rod-shaped micelles. The experimental data are compared with the theoretical predictions of a special constitutive equation which is based on the concept of deformation-dependent tensorial mobility. In the regime of small deformations, shear stresses or shear rates, the dynamic features of the viscoelastic solutions are characterized by the equations of a simple Maxwell material. These phenomena are linked to the average lifetime of the micellar aggregates and the rheological properties are controlled by kinetic processes. At these conditions one observes simple scaling laws and linear relations between all theological quantities. At elevated values of shear stresses or deformations, however, this simple model fails and non-linear properties as normal stresses, stress overshoots or shear-thinning properties occur. All these phenomena can be described by a constitutive equation which was first proposed by H. Giesekus. The experimental results are in fairly good agreement with the theoretical predictions, and this model holds for a certain, well defined value of the mobility factor α. This parameter describes the anisotropic character of the particle motion. In transient and steady-state flow experiments we always observed α = 0.5. Especially at these conditions, the empirically observed Cox-Merz rule, the Yamamoto relation and both Gleißle mirror relations are automatically derived from the Giesekus model. The phenomena discussed in this paper are of general importance, and can be equally observed in different materials, such as polymers or proteins. The viscoelastic surfactant solutions can, therefore, be used as simple model systems for studies of fundamental principles of flow.

    Notizen: Abstract  Our analysis of IgG rheumatoid factors (RFs) from three patients with rheumatoid arthritis (RA) revealed that most contained significant numbers of skewed mutations per V region, suggesting that these RFs arose from antigen-driven responses. To further study IgG RFs in RA, we used pComb3 vector to construct an IgG1,λ combinatorial antibody library from a synovial fluid sample. After panning against human IgG, Fab fragments from 71/96 phage clones bound to Fc-coated wells. Sequence analysis of 20 randomly chosen Fc-binders showed that 17 (85%) clones had identical heavy (H) and light (L) chain V regions, represented by Humha311 and Humla211, respectively. Of the remaining three clones, two had the same Humla211 L chain, but each with a different H chain V region. All the putative germline V genes for these RFs also encode RF in RA patients. However, none of these RF V regions are similar to those of the two IgG RFs derived by the hybridoma technique from the same synovial fluid. The Humha311 H chain has two frameshifts: a one-base insertion upstream of the JH region and a four-base deletion near the end of the CH1 region, resulting in a mainly unconventional amino acid sequence in the CH1 region. In the future, it will be important to study the presence of IgG molecules with such unconventional CH1 amino acid sequences, and the effects of these amino acid sequences on the structures and immunological properties of the IgG molecules.

    Notizen: Abstract With a view to cloning the root-knot nematode resistance gene Mi in tomato by chromosome walking, we have developed a molecular probe for the tightly linked acid phosphatase-1 (Aps-1) locus. The acid phosphatase-1 allozyme (APS-11), encoded by the Aps-1 1 allele originating from Lycopersicon peruvianum, was purified to apparent homogeneity from tomato roots and suspension cells. Microsequencing of CNBr and tryptic peptides generated from APS-11 provided a partial amino acid sequence, which accounted for approximately 23% of the protein and revealed two stretches of homology with soybean proteins KSH3 and VSP27, comprising 22 matches within 26 amino acid residues. The partial amino acid sequence information enabled us to isolate a 2.4 kb genomic Aps-1 1 sequence by means of the polymerase chain reaction (PCR), primed by degenerate pools of oligodeoxyribonucleotides, synthesized on the basis of the amino acid sequences. Synthesis of the 2.4 kb PCR product was specific for genomic templates carrying the L. peruvianum Aps-1 1 allele. Crucial to the priming specificity and the synthesis of the 2.4 kb genomic sequence was the use of degenerate primer pools in which the number of different primer species was limited by incorporating deoxyinosine phosphate residues at three and four base ambiguities. In using cDNA as a template, a 490 bp sequence was obtained, indicating a high proportion of intron sequences in the 2.4 kb genomic Aps-1 1 sequence. The Aps-1 1 origin of the PCR product was confirmed by RFLP (restriction fragment length polymorphism) analysis, using both a chromosome 6 substitution line and a pair of nearly isogenic lines, differing for a small chromosomal region around the Aps-1/Mi loci.

    Notizen: Summary For the ion-chromatographic separation of structurally diverse phosphonic acids on Dionex HPIC AS7, postcolumn derivatization with Fe(III) and subsequent UV detection is systematically investigated, and optimized postcolumn eluent (PCE) conditions are given. The complexes are detected with highest sensitivity close to the absorption maximum at 300 nm, contrary to the practice in the literature (330 nm). It is further tested whether and to which extent this derivatization and detection methodology may be transferred to post-column complexation with Cu(II) and Ni(II), and also to a specific UV detection of orthophosphate. Direct RI detection, without previous enrichment, is described and compared in its validity and applicability to post-column complexation/UV detection. A photometric method is presented which allows for differentiation between inorganic phosphate, compounds which can be hydrolyzed to, and compounds which can be oxidized to orthophosphate, and thus makes a rapid control possible of reaction mixtures or stock solutions for these different phosphor species.

    Notizen: Abstract Recently we studied time dependent structural changes that are coupled with flow instabilities (Fischer 1998; Wheeler 1998; Fischer 2000). Within a stability analysis, a classification scheme for the feedback circuit of coupled shear-induced structure and flow instabilities was derived by Schmitt et al. (1995) and applied to our samples. Here, inhomogeneous flow layers of different concentration and viscosity are generated by shear-induced diffusion (spinodal demixing) and, as consequence, one no longer observes a homogeneous solution but a type of shear banding that is seen here for the first time. In this paper we present the behaviour of the first normal stress difference observed in the critical shear-rate regime where transient shear-induced structure is coupled with flow instability. Similar to the oscillations of the shear stresses (strain-controlled rheometer) one observes oscillations in the first normal stress difference. This behaviour indicates that elastic structures are built up and destroyed while the shear-induced structures occur and that the induced phase is more elastic than the initial one. Oscillations of shear stress and first normal stress difference are in phase and indicate that both phenomena are caused by the same mechanism.

    Notizen: Abstract Several years ago, Münstedt and Laun reported on the influence of branching on the elongational flow properties of polymer chains (Münstedt and Laun, 1981). They concluded that, in addition to the molecular weight distribution, the degree of branching strongly affects the degree of strain thickening of the elongational viscosity in such a way that the maximum in this material function increases with branching. In a recent paper by Lin, a ternary system of dodecyldimethylamine oxide-sodium laureth sulphate-sodium chloride surfactant solutions was investigated by CryoTEM and rheology (Lin, 1996). He reported a linear relation between the added sodium chloride and the branching of the wormlike micelles. In this paper we present an investigation of these surfactant solutions in elongational flow. Our results indicate that for branched micellar systems the presence of branching enhances the maximum of the elongational viscosity in the same manner as in the case of polymer melts.

    Notizen: Abstract In recent years, bovine Onchocerca species have been used as models for human onchocerciasis in drug screens. They have been suggested for immunology studies and evaluation of vaccine candidates. Therefore, mast cells and their association with other inflammatory cells were studied in five onchocercal species of cattle and deer using immunohistology. Intact mast cells occurred in large numbers in the capsule and septae of nodules, in fibrous tissue adjacent to nonnodular worms, and perivascularly. Inactive and, more frequently, activated and degranulating mast cells were observed within infiltrates in the nodule center or around nonnodular filariae. They were not detected in direct contact with the cuticle of adult worms or of microfilariae or among the macrophages, giant cells, and neutrophils forming the innermost layer around the worms. Eosinophils, but not mast cells, were obviously associated with microfilariae-producing females. The distribution, frequency, and activity of mast cells were similar for all five species and O. volvulus.
