    Notizen: Summary The mechanism of electrical coupling between cells of earlyXenopus embryos has been studied by examination of the nonjunctional membrane resistances and capacitances as a function of cleavage stage, the junctional and nonjunctional membrane resistances as functions of time during the first cleavage, and the electrical properties of the primitive blastocoel. The changes in membrane resitances and capacitances during the first two cleavages may be completely explained by the addition of new membrane, identical in specific resistance and capacitance to the original membrane, at a constant rate to furrows which are electrically connected to the perivitelline space. Microelectrode recording from the primitive blastocoel indicates that there is no electrical difference detectable between it and the perivitelline space. These results are discussed in the context of current theories of the mechanism of intercellular electrotonic coupling.

    Notizen: Abstract Expression of human parathyroid hormone, hPTH(-1-84), by Escherichia coli N4830: pEX-PPTH was studied in controlled bioreactors. The hPTH is expressed as a fusion protein under control of the bacteriophage λpR promoter. In batch runs, low biomass concentrations but high specific hPTH productivities were obtained with complex TY (bactotryptone and yeast extract) medium whereas high biomass concentration and low specific productivities were found when fructose was used instead of bactotryptone (YF medium). The preinduction temperature was always 30°C; the temperature shift to induce production of fusion protein was varied from 36 to 42°C. Formation of hPTH passed a pronounced maximum as a function of induction temperature when using YF medium. However, the optimum temperature shift was 38°C for both media used. For this temperature increase both media yielded about the same volumetric hPTH productivity (approx. 30 mg hPTH/l per hour). By applying a fedbatch strategy for the YF medium, the productivity of the recombinant protein could be further increased more than fourfold. Compared to shake-flask experiments, the hPTH yield could be increased by a factor larger than 20.

    Notizen: Summary Production of lipase from recombinant Staphylococcus carnosus pLipPS2 was studied in standard stirred tank bioreactors. Only low lipase activity was obtained under conventional operating conditions, i.e., moderate to high stirring speeds and aeration rates for keeping the dissolved oxygen concentration at high levels. Additional targetted experiments indicated that the reason for the observed low lipase activity is lipase inactivation due to surface forces and shear stress at the gas/liquid interface. Therefore, a cultivation strategy is proposed that minimizes gas/liquid interfacial area and maximizes the driving concentration for O2 mass transfer by controlling the dissolved oxygen to low values by gentle stirring and low aeration rates. Thus, high lipase activities can be obtained even in larger scale standard stirred tank bioreactors.

The kallikrein content is greatly reduced after chronic parasympathetic but not after sympathetic nerve section which suggests that the parasympathetic innervation is required for synthesis or storage of this enzyme.

    Notizen: A new mass spectral method has been developed and used to identify biologically important thiols. This method is based upon the use of C-14 labeled N-para-bromophenylmaleimide (BPM) to selectively derivatize thiols. The isotopic label facilitates the isolation and purification of trace quantities of the maleimide thiol adducts by high performance liquid chromatography (HPLC) with radioactivity detection. The use of the bromine atom in the thiol derivative greatly enhances the detectability of the parent and fragment ions containing the BPM moiety because of the generation of the characteristic N + 2 doublet ions. Several examples of the use of this method to identify non-volatile thiol peptides such as glutathione (GSH) and homoglutathione (hGSH) by fast atom bombardment mass spectrometry (FAB-MS) are described in this report.

    Notizen: Summary The ultrastructure and interrelationships of the Golgi body, endoplasmic reticulum and lipid droplets have been studied in the first cleavage Xenopus embryos. Lipid droplets, usually spherical or sometimes multilobed, did not have a discernible limiting membrane, although some had an incomplete electron dense partition. The Golgi bodies and endoplasmic reticulum were seen continuous with lipid droplets and the profiles indicated a probable formation of these membranes from lipid droplet material. Rough endoplasmic reticulum (ER) mainly consisted of paired tubular cisternae and vesicles containing filamentous material that gave a fringed appearance. The relationships of paired cisternae with the Golgi body suggested a transformation of ER membranes into the Golgi body membranes. In addition, paired ER cisternae showed a close apposition with the limiting membrane of the yolk platelet. Lone ER cisternae that contained moderately electron dense material instead of filaments were also present and showed numerous associated vesicles near the Golgi body. The Golgi body showed several morphological forms including a single fenestrated cisterna, two to four flat or cup-shaped cisternae, or up to seven cisternae, some of which were dilated and similar to fringed ER in appearance. These forms could be different developmental stages of the organelle. Coated vesicles were seen continuous with the cisternae of the Golgi body. A probable route for the assembly of the cell surface material has been proposed.

    Notizen: Summary This communication reports the results of a morphological study of three-day old cultures of epiblast tissue from the early chick embryo. The most striking feature of these cultures was the appearance of “domes” or elevated blister-like structures, composed of a single layer of cells which were morphologically distinct from the remaining cells in the culture. The domes arose in high-density areas of the culture. Their roofs were lined by basal laminae that did not develop in other areas of the culture. In several morphological respects, the cells of the dome roof closely resembled the epiblast in vivo. This was in contrast to the cells spread on the substratum in sparse regions of the culture, which did not. Each dome was surrounded by a dense ring of multilayered ruffling cells which appeared to give rise to both the dome roof and to fibroblast-like cells that spread on the substratum beneath the dome. Fibroblast-like cells also developed in discrete patches in other regions of the culture. In other tissues, dome formation has been attributed to fluid transport by the epithelium; in the present case it is also possible to invoke the capacity of the epiblast to fold, as contributing to the mechanism of dome formation.
