    Notes: Summary Mitochondrial creatine kinase (CK) purified from canine myocardium showed a single protein band on SDS-PAGE and was free of MMCK. Its amino acid composition was different than MMCK or BBCK and did not react to antiserum to MMCK or BBCK. Using purified mitochondrial, MM and BBCK, the velocity of reaction (V) was estimated for creatine phosphate (CP), creatine (C), adenosine triphosphate (ATP) and adenosine diphosphate (ADP) over a wide range of concentrations including those at Vmax. The values for Km (mM/L) derived from Lineweaver-Burke plots are shown: The affinity of mitochondrial CK for C is much greater than MMCK which is compatible with the energy shuttle hypothesis, namely ATP is converted by mitochondrial CK to CP, and then diffuses to the myofibril for conversion to ATP for utilization.

    Notes: Summary It is not known whether loss of enzyme activity from the circulation is due to denaturation, inactivation or removal of intact enzyme molecules. This is in part due to the lack of an assay to measure enzyme protein concentration since available assays measure only enzyme activity. Radioimmunoassays for plasma enzymes and isoenzymes have not been possible because of oxidation in radioactive labelling by conventional methods and the problem of subunit dissociation. In the present study, antibodies specific to the B and M subunits of creatine kinase isoenzymes were obtained by immunization of rabbits with canine BB and MM creatine kinase. Anitgens (MM and BB) were radioactively labelled with 125I by acylation, avoiding the problem of oxidation and subunit stabilized by mercaptoethanol (0.020 m) and Trisbuffer (1.6 m). A radioimmunoassay capable of detecting picogram amounts of CK isoenzymes was developed which measures the concentration of enzyme protein rather than activity. The method was shown to provide a sensitive quantitative method for analysis of plasma CK isoenzymes in dogs after myocardial infarction produced by coronary occlusion. This technique may provide a prototype for the development of radioimmunoassays for other plasma isoenzymes and should help to elucidate the nature of the disappearance of isoenzymes from the circulation.

    Notes: We describe multiple human cardiac and skeletal muscle spectrin isoforms. Cardiac muscle expresses five erythroid α,β spectrin-reactive isoforms with estimated MR's of 280, 274, 270, 255, and 246 kD, respectively At least one nonerythroid α-spectrin of MR 284 kD is expressed in heart. While skeletal muscle shares the 280, 270, and 246 kD erythroid spectrins, it expresses an immunologically distinct 284 kD nonerythroid α-spectrin isoform. The 255 kD erythroid β-spectrin isoform is specific for cardiac tissue. By immunocytochemistry, both erythroid β- and nonerythroid α-spectrins are localized to costameres, the plasma membrane, and the neuromuscular junctional region.

    Notes: Abstract. Flow cytometry was used to study the effect of the bacteriocin leucocin B-TA11a on Listeria (L.) monocytogenes. Mixed proportions of dead and live control populations were analyzed by flow cytometry to determine detection limits of the Dead/Live Baclight Bacterial Viability KitTM. High correlations for flow cytometric detection of defined proportions of live or dead cells in mixtures between 10 and 100% of dead (r2 = 0.97) or live (r2 = 0.99) cells were obtained. However, mixtures containing less than 10% of either live or dead control cells gave correlations below 0.72. The growth of L. monocytogenes in the absence and presence of leucocin B-TA11a was analyzed by flow cytometry with Baclight, plate counts, and optical density measurements. Although leucocin B-TA11a initially inhibited listerial growth, the uptake of both Baclight dyes suggested that cells remained viable but became leaky, possibly indicating bacteriocin-induced pore formation in the target membranes.

    Notes: An H2O2-resistant variant (OC14) of the HA1 Chinese hamster fibroblast cell line, which demonstrates cross resistance to 95% O2 and a 2-fold increase in total glutathione content, was utilized to investigate mechanisms responsible for cellular resistance to H2O2- and O2-toxicity. OC14 and HA1 cells were pretreated with buthionine sulfoximine (BSO) to deplete total cellular glutathione. Following BSO pretreatment, cells were either placed in 250 μM BSO to maintain the glutathione depleted condition and challenged with 95% O2, or challenged with hydroged peroxide in the absence of BSO. Total glutathione and the activities of CuZn superoxide dismutase, Mn superoxide dismutase, catalase, glutathione peroxidase, and glutathione transferase were evaluated immediately following the BSO pretreatment as well as following 39 to 42 hr of exposure to 250 μM BSO. BSO treatment did not cause significant decreases in any cellular antioxidant tested, except total glutathione depletion resulted in significant (P 〈 0.05) sensitization to O2-toxicity and H2O2-toxicity in both cell lines at every time point tested. However, glutathione depletion did not completely abolish the resistance to either O2- or H2O2-toxicity demonstrated by OC14 cells, relative to HA1 cells. Also, glutathione depletion did not effect the ability of OC14 cells to metabolize extracellular H2O2. These data indicate that glutathione dependent processes significantly contribute to cellular resistance to acute H2O2- and O2-toxicity, but are not the only determinants of resistance in cell lines. The contribition of aldehydes formed by lipid peroxidation in mechanisms involved with the sensitization to O2-toxicity in glutathione depleted cells was tested by measuring the lipid peroxidation byproduct, 4-hydroxy-2-nonenal (4HNE), bound in Schiff-base linkages or in its free form in cell homogenates at 49 hr of 95% O2-exposure. No significant increase in 4HNE was detected in glutathione depleted cells relative to glutathione competent cells, indicating that glutathione depletion does not sensitize these cells to O2-toxicity by altering the intracellular accumulation of free or Schiff-base bound 4HNE. © 1995 Wiley-Liss Inc.

    Notes: Solvent-cast metal-filled composites made of polymethyl methacrylate and aluminum or copper exhibited electrical resistance behavior as is predicted by the percolation and tunneling mechanisms. A precipitous change in volume resistivity of about 1010 ohm-cm was found to be thermally dependent at volume concentration of filler in the range from 17 to 19.5 percent. Materials with compositions at the conductive-non-conductive threshold showed semi-conductor behavior and exhibited memory switching.

    Notes: Results are presented from a systematic study of the noise (N) and peak-to-noise ratio (P/N) obtained for pulsed electrochemical detection (PED) at a gold micro-wire electrode applied to a mixture of alditols and monosaccharides separated by capillary electrophoresis (CE). The electrode potentials and time periods for oxidative cleaning and reductive reactivation of the electrode were held constant in the PED waveform at minimum allowable values and the time period for digital integration of electrode current (tint) was varied from 50 to 800 ms by 50-ms intervals. The value of N was virtually independent of tint and P/N increased approximately as a linear function of tint in the range 50-200 ms corresponding to waveform frequencies in the range 4.5-2.7 Hz. A detection limit of 9 fmol glucose was determined for a 4-nL injection using tint = 200 ms.
