    Notes: Abstract. The cell cycle is driven by the sequential activation of a family of cyclin-dependent kinases (CDK) in association with cyclins. In mammalian cells the timing of activation of cyclin A-associated kinase activity coincides with the onset of DNA synthesis in S-phase. Using in vitro replication of SV40 origin-containing DNA as a model system, we have analyzed the proteins associated with DNA during initiation of DNA replication in S-phase cell extracts. This analysis reveals that, in addition to replication initiation proteins, cyclin A and cdk2 are also specifically associated with DNA. The association of cyclin A and cdk2 with DNA during initiation is cell cycle regulated and occurs specifically in the presence of SV40 origin-containing plasmid and SV40 T antigen (the viral replication initiator protein). The interactions among proteins involved in initiation play an important role in DNA replication. We therefore investigated the ability of cyclin A and cdk2 to associate with replication initiation proteins. Under replication initiation conditions, cyclin A and cdk2 from S-phase extracts specifically associate with SV40 T antigen. Further, the interaction of cyclin A-cdk2 with SV40 T antigen is mediated via cyclin A, and purified recombinant cyclin A associates directly with SV40 T antigen. Taken together, our results suggest that cyclin A and cdk2 are components of the SV40 replication initiation complex, and that protein-protein interactions between cyclin A-cdk2 and T antigen may facilitate the association of cyclin A-cdk2 with the complex.

    Notes: Abstract The applicability of the tungsten oxide denuder tube technique for the measurement of ammonia in the rural troposphere was investigated. The technique is based on selective chemisorption of NH3 from a gas stream, thermal desorption, conversion to NO, and analysis by NO−O3 chemiluminescence. Nitric acid, which is also collected and desorbed as NO, was distinguished from NH3 by differences in desorption temperature. Substituted amines were also collected, but desorbed at a slightly lower temperature than NH3 in dry air. At high relative humidities, alkylamines may be hydrolyzed to NH3 on the denuder surface and hence detected as NH3. Overheating of the denuder tube during the temperature-programmed desorption was found to cause significant irreversible degradation of system performance. The technique was used to measure NH3 mixing ratios at two rural locations in the United States. At a mountain site in Colorado during the winter of 1984, the average NH3 mixing ratio was 0.20 ppbv (σ=0.08 ppbv). At an isolated coastal site in northern California during the spring of 1985, the average NH3 mixing ratio was 0.36 ppbv (σ=0.17 ppbv). Correlations of the latter measurements with wind direction and NO x level suggest that the NH3 mixing ratio in Pacific marine air at 40°N is 〈-0.25 ppbv.

    Notes: Beckwith-Wiedemann syndrome is a human congenital disorder characterized by a wide variety of growth abnormalities, including developmental defects and predisposition to certain tumors. Genetic evidence has suggested a role for p57KIP2, a member of a family of cell cycle inhibitory genes, in Beckwith-Wiedemann syndrome. Two independent groups(1,2) have reported the generation and characterization of mice lacking functional p57KIP2, These mice demonstrate a number of abnormal phenotypes which overlap with, although do not completely recapitulate, Beckwith-Wiedemann syndrome. These findings advance the molecular characterization of a human disorder, and provide insight into the interplay between regulation of cell division and development.

    Notes: The rate coefficients of thermal decomposition of peroxyacetic nitric anhydride (PAN) and peroxymethacrylic nitric anhydride (MPAN) were measured over the temperature range 302-323 K. The resulting Arrhenius expressions were k = 1017.4±0.4 exp(-28.5 ± 0.5/RT) for PAN, and k = 1016.2±0.7 exp(-26.8 ± 1.0/RT) for MPAN, where the activation energy is in Kcal/mol. These results are in good agreement with previous studies of PAN and other PAN-type compounds, and imply that energies of RC(O)OO—NO2 bonds are relatively independent of the nature of R.
