    Notes: Abstract  Three RFLP maps, as well as several RAPD maps have been developed in common bean (Phaseolus vulgaris L.). In order to align these maps, a core linkage map was established in the recombinant inbred population BAT93×Jalo EEP558 (BJ). This map has a total length of 1226 cM and comprises 563 markers, including some 120 RFLP and 430 RAPD markers, in addition to a few isozyme and phenotypic marker loci. Among the RFLPs mapped were markers from the University of California, Davis (established in the F2 of the BJ cross), University of Paris-Orsay, and University of Florida maps. These shared markers allowed us to establish a correspondence between the linkage groups of these three RFLP linkage maps. In total, the general map location (i.e., the linkage group membership and approximate location within linkage groups) has been determined for some 1070 markers. Approaches to align this core map with other current or future maps are discussed.

    Notes: Abstract  Wild plant species are often adapted to more stressful environments than their cultivated relatives. Roots are critical in exploiting soil resources that enable plants to withstand environmental stresses, but they are difficult to study. Cultivated lettuce (Lactuca sativa L.) and wild L. serriola L. differ greatly in both shoot and root characteristics. Approximately 100 F2:3 families derived from an interspecific cross were evaluated in greenhouse and field experiments. In the greenhouse, root traits (taproot length, number of laterals emerging from the taproot, and biomass) and shoot biomass were measured 4 weeks after planting. In the field, plants were grown for 9 weeks (close to harvest maturity of the cultivated parent); mild drought stress was induced by withholding water for 1 week, and gravimetric moisture of soil was then determined for five depth increments between 0–100 cm. The families were genotyped using codominantly scored AFLP markers distributed throughout the genome. Composite interval mapping was used to analyze marker-trait associations. Quantitative trait loci were identified for differences between wild and cultivated lettuce for root architectural traits and water acquisition. Thirteen QTL were detected that each accounted for 28–83% of the phenotypic variation. The loci for taproot length (i.e., cm taproot length g–1 plant biomass) and the ability to extract water from deep in the soil profile co-localized in the genome. These coincident loci were identified in separate experiments. The wild L. serriola is therefore a potential source of agriculturally important alleles to optimize resource acquisition by cultivated lettuce, thereby minimizing water and fertilizer inputs and ultimately enhancing water quality.

    Notes: Abstract Lakes and reservoirs provide water for human needs and habitat for aquatic birds. Managers of such waters may ask whether nutrients added by waterfowl degrade water quality. For lakes and reservoirs where primary productivity is limited by phosphorus (P), we developed a procedure that integrates annual P loads from waterfowl and other external sources, applies a nutrient load-response model, and determines whether waterfowl that used the lake or reservoir degraded water quality. Annual P loading by waterfowl can be derived from a figure in this report, using the days per year that each kind spent on any lake or reservoir. In our example, over 6500 Canada geese (Branta canadensis) and 4200 ducks (mostly mallards, Anas platyrhynchos) added 4462 kg of carbon (C), 280 kg of nitrogen (N), and 88 kg of P y−1 to Wintergreen Lake in southwestern Michigan, mostly during their migration. These amounts were 69% of all C, 27% of all N, and 70% of all P that entered the lake from external sources. Loads from all external sources totaled 840 mg P m−2 y−1. Application of a nutrient load-response model to this concentration, the hydraulic load (0.25 m y−1), and the water residence time (9.7 y) of Wintergreen Lake yielded an average annual concentration of total P in the lake of 818 mg m−3 that classified the lake as hypertrophic. This trophic classification agreed with independent measures of primary productivity, chlorophyll-a, total P, total N, and Secchi disk transparency made in Wintergreen Lake. Our procedure showed that waterfowl caused low water quality in Wintergreen Lake.

    Notes: A particularly simple theory of circular dichroism is described and applied to the polynucleotides. It works surprisingly well and predicts the conservative spectrum of DNA and the more intense nonconservative spectrum of RNA. The anomalously low intensity of the CD of the nucleic acids is shown to be a natural consequence of the cancellation of the rotational strengths of the many CD bands. The theory's many successes and predictions as well as its weaknesses are discussed.

    Notes: Working within the restrictions of a model, we have calculated the circular dichroism of the dinucleoside phosphates ApA, CpC, and CpA for various conformations. Comparing the calculated curves with those measured in aqueous solution we find agreement for (1) ApA as a right-handed helix with both bases either as in B-form DNA, or else rotated 180° around the glycosidic bond, (2) CpC as the right-handed conformation with both bases as in DNA, (3) ApC as either the right-handed conformation with both bases as in DNA, or else as a left-handed helix with both bases rotated 180°, and (4) CpA as either a left-handed helix with both bases in a left-handed DNA, or else in the right-handed conformation with both bases rotated 180°. In addition, we have investigated circular dichroism as a measure of unstacking. We find that opening the bases to a 90° total angle (base planes perpendicular) reduces the intensity of the calculated bands to 20% of their original value. Further, we find that allowing the sliding of one base past the other does not lead to a temperature dependence consistent with experiment.

    Notes: The large change in the electronic absorption spectrum of cytidine on going from an aqueous to an aprotic medium is attributed to the existence of a hydrogen bonded solvent-solute complex in solvents capable of donating a proton. The spectrum of cytidine in a variety of solvents and the spectra of a number of nontautomerising model compounds are presented. The tautomerisin of cytidine and its biological implications are discussed.

    Notes: A method is described which relates the circular dichroism of a polymer to its conformation. The method takes into account near and accidental degeneracies and eliminates the artificial distinction between degenerate and nondegenerate systems. Comparison of this method with perturbation theory indicates that the errors inherent in nondegenerate perturbation theory tend to cancel when a circular dichroism spectrum is calculated. The method is applied to dinucleoside phosphates.
