    Beschreibung: A DNA fingerprinting method for the characterization of Legionella pneumophila serogroup I strains was established. This method was based on the DNA extraction using Chelex 100 and subsequent PCR analysis using primers under conditions of low stringency. Sixteen single primers were tested for the typing of the 10 epidemiologically unrelated reference strains of L. pneumophila serogroup I as well as patient isolates and environmental strains isolated from the water system of a hospital where patients with legionellosis were treated. In addition, a combination of two primers (Lpm-1 and Lpm-2) originally established for the specific detection of Legionella strains was tested. The PCR results were compared with two further subtyping methods, i.e. monoclonal antibody analysis and pulsed-field gel electrophoresis. The type strains Philadelphia 1, Knoxville 1, Allentown 1, Benidorm 0303E, Bellingham 1, and France 5811 could be distinguished clearly in experiments using all of the primers. Depending on the primer used, Heysham 1 and Oxford 4032E showed different DNA profiles. The strains Olda and Camperdown I were nearly indistinguishable. In contrast, the analysis by PFGE and MAb subtyping revealed distinct types for all 10 reference strains. The discrimination of the patient isolates from two suspected cases of nosocomial legionellosis and environmental isolates was not possible with the 16 single primers used in the study. However, the PCR assay with the combination of Lpm-1 and Lpm-2 as well as the PFGE and MAb analysis were able to differentiate distinct types. The use of the sequence-specific primers under low-stringency annealing conditions allowed both simultaneous gene detection as well as epidemiological typing of Legionella strains.

    Beschreibung: The (87)Rb/(86)Sr and (87)Sr/(86)Sr ratios of Laptev Sea sediments, of Arctic Ocean sediments and of suspended particulate matter (SPM) from Siberian rivers (Lena and Khatanga) form 'pseudo-isochrons' due to grain-size separation processes which are referred to as 'Lena Mixing Envelope' (LME) and as 'Flood Basalt Envelope' (FBE). At the land-ocean transition the reduction of the particle velocity causes a deposition of coarser grained material and the contact with saline water enhances a precipitation of finer-grained material. The coarse-grained material is enriched in Sr showing less radiogenic (87)Sr/(86)Sr ratios whereas fine grained material is depleted in Sr relative to Rb showing more radiogenic (87)Sr/(86)Sr ratios, The experimentally determined spread of the (87)Rb/(86)Sr and (87)Sr/(86)Sr ratios as a function of grain size in one sediment sample is on the same order as the natural spread of the (87)Sr/(86)Sr ratios observed in all samples from the Arctic Ocean. Chemical Index of Alteration (CIA) for the Lena river SPM tend to confirm previous observations that chemical alteration is negligible in the Arctic environment. Thus, these 'pseudo-isochrons' reflect an average age and the average isotope composition in the river drainage area. Calculated apparent ages from the FBE reflect the age of the Siberian flood basalt of about 220 Ma and the initial ratio of 0.707(1) reflects their mantle origin. The age calculated from the LME of about 125 Ma reflects accidentally the Jurassic and Cretaceous age of the sediments drained by the Lena river and the initial ratio of 0.714(1) reflects the crustal origin of their source rocks. Comparison of geographical locations reveals that all samples from the eastern Laptev Sea (east of 120 degrees E) fall along the LME whereas all samples from the western Laptev Sea (west of 120 degrees E) fall between LME and FBE. Mixing calculations based on (143)Nd/(144)Nd measurements, not influenced by grain size, show that about 75% of the western Laptev Sea sediments originate from the Lena drainage area whereas about 25% of the sediments are delivered from the Siberian flood basalt province. Sediments from the central Arctic Ocean are isotopically related to the Lena drainage area and the Siberian flood basalt province. However, sediments from the Arctic Ocean margins close to Novaya Semlya, Greenland, the Fram Strait and Svalbard originate from sources not yet identified. (C) 1999 Elsevier Science B.V.     Beschreibung: The origin of novel phenotypic characters is a key component in organismal diversification; yet, the mechanisms underlying the emergence of such evolutionary novelties are largely unknown. Here we examine the origin of egg-spots, an evolutionary innovation of the most species-rich group of cichlids, the haplochromines, where these conspicuous male fin colour markings are involved in mating. Applying a combination of RNAseq, comparative genomics and functional experiments, we identify two novel pigmentation genes, fhl2a and fhl2b, and show that especially the more rapidly evolving b-paralog is associated with egg-spot formation. We further find that egg-spot bearing haplochromines, but not other cichlids, feature a transposable element in the cis-regulatory region of fhl2b. Using transgenic zebrafish, we finally demonstrate that this region shows specific enhancer activities in iridophores, a type of pigment cells found in egg-spots, suggesting that a cis-regulatory change is causally linked to the gain of expression in egg-spot bearing haplochromines

Abstract: The species-flocks of cichlid fishes in the East African Great Lakes Victoria, Malawi and Tanganyika constitute the most diverse extant adaptive radiations in vertebrates. Lake Tanganyika, the oldest of the lakes, harbors the morphologically and genetically most diverse assemblage of cichlids and contains the highest number of endemic cichlid genera of all African lakes. Based on morphological grounds, the Tanganyikan cichlid species have been grouped into 12–16 distinct lineages, so-called tribes. While the monophyly of most of the tribes is well established, the phylogenetic relationships among the tribes remain largely elusive. Here, we present a new tribal level phylogenetic hypothesis for the cichlid fishes of Lake Tanganyika that is based on the so far largest set of nuclear markers and a total alignment length of close to 18 kb. Using next-generation amplicon sequencing with the 454 pyrosequencing technology, we compiled a dataset consisting of 42 nuclear loci in 45 East African cichlid species, which we subjected to maximum likelihood and Bayesian inference phylogenetic analyses. We analyzed the entire concatenated dataset and each marker individually, and performed a Bayesian concordance analysis and gene tree discordance tests. Overall, we find strong support for a position of the Oreochromini, Boulengerochromini, Bathybatini and Trematocarini outside of a clade combining the substrate spawning Lamprologini and the mouthbrooding tribes of the ‘H-lineage’, which are both strongly supported to be monophyletic. The Eretmodini are firmly placed within the ‘H-lineage’, as sister-group to the most species-rich tribe of cichlids, the Haplochromini. The phylogenetic relationships at the base of the ‘H-lineage’ received less support, which is likely due to high speciation rates in the early phase of the radiation. Discordance among gene trees and marker sets further suggests the occurrence of past hybridization and/or incomplete lineage sorting in the cichlid fishes of Lake Tanganyika.

    Beschreibung: Since the onset of microbiology in the late 19th century, scientists have been growing microorganisms almost exclusively as pure cultures, resulting in a limited and biased view of the microbial world. Only a paradigm shift in cultivation techniques – from axenic to mixed cultures – can allow a full comprehension of the (chemical) communication of microorganisms, with profound consequences for natural product discovery, microbial ecology, symbiosis, and pathogenesis, to name a few areas. Three main technical advances during the last decade are fueling the realization of this revolution in microbiology: microfluidics, next-generation 3D-bioprinting, and single-cell metabolomics. These technological advances can be implemented for large-scale, systematic cocultivation studies involving three or more microorganisms. In this review, we present recent trends in microbiology tools and discuss how these can be employed to decode the chemical language that microorganisms use to communicate.

    Beschreibung: Microplastic pollution within the marine environment is of pressing concern globally. Accordingly, spatial monitoring of microplastic concentrations, composition and size distribution may help to identify sources and entry pathways, and hence allow initiating focused mitigation. Spatial distribution patterns of microplastics were investigated in two compartments of the southern North Sea by collecting sublittoral sediment and surface water samples from 24 stations. Large microplastics (500−5000 μm) were detected visually and identified using attenuated total reflection (ATR) Fourier transform infrared (FTIR) spectroscopy. The remaining sample was digested enzymatically, concentrated onto filters and analyzed for small microplastics (11−500 μm) using Focal Plane Array (FPA) FTIR imaging. Microplastics were detected in all samples with concentrations ranging between 2.8 and 1188.8 particles kg−1 for sediments and 0.1–245.4 particles m−3 for surface waters. On average 98% of microplastics were 〈100 μm in sediments and 86% in surface waters. The most prevalent polymer types in both compartments were polypropylene, acrylates/polyurethane/varnish, and polyamide. However, polymer composition differed significantly between sediment and surface water samples as well as between the Frisian Islands and the English Channel sites. These results show that microplastics are not evenly distributed, in neither location nor size, which is illuminating regarding the development of monitoring protocols.

    Beschreibung: Importance of target-oriented validation strategies for spatio-temporal prediction models is illustrated using two case studies: (1) modelling of air temperature () in Antarctica, and (2) modelling of volumetric water content (VW) for the R.J. Cook Agronomy Farm, USA. Performance of a random k-fold cross-validation (CV) was compared to three target-oriented strategies: Leave-Location-Out (LLO), Leave-Time-Out (LTO), and Leave-Location-and-Time-Out (LLTO) CV. Results indicate that considerable differences between random k-fold ( = 0.9 for and 0.92 for VW) and target-oriented CV (LLO = 0.24 for and 0.49 for VW) exist, highlighting the need for target-oriented validation to avoid an overoptimistic view on models. Differences between random k-fold and target-oriented CV indicate spatial over-fitting caused by misleading variables. To decrease over-fitting, a forward feature selection in conjunction with target-oriented CV is proposed. It decreased over-fitting and simultaneously improved target-oriented performances (LLO CV = 0.47 for and 0.55 for VW).

    Beschreibung: Atmospheric and dissolved methane (CH4) and nitrous oxide (N2O) were measured in the unique coastal ecosystem of the Bodden waters, including the western Oder estuary, (southern Baltic Sea) during five campaigns between 1994 and 1997. The CH4 saturations, ranging from 105-15 500%, showed great spatial and temporal variability with maximum values in September and minimum values in December. The N2O saturations were in the range of 91-312% with a maximum in March. Enhanced concentrations of both gases were observed only in the western Oder estuary near the mouth of the Peene River. Thus, we conclude that the distributions of CH4 and N2O in the investigated Bodden waters are, directly or indirectly, linked to the Peene River runoff and not to the Oder River. Our estimate of the annual CH4 emissions from the Bodden waters to the atmosphere indicates a significant contribution (c. 17%) to the overall CH4 emissions from the Baltic Sea. In contrast, the Bodden waters represent only a small source for atmospheric N2O. CH4 production rates estimated from sediment slurry experiments revealed a significant spatial variability and indicated that methanogenic activity was related to acetate consumption in the surface sediment layer.     Beschreibung: In this study we aim on a reconstruction of mechanisms and kinematics of slope-failure and mass-movement processes along the northeastern slope of Crete in the Hellenic forearc, eastern Mediterranean. Here, subsidence of the forearc basin and the uplift of the island of Crete cause ongoing steepening of the slope in-between. The high level of neotectonic activity in this region is expected to exert a key role in slope-failure development. Newly acquired reflection seismic data from the upper slope region reveal an intact sediment cover while the lower slope is devoid of both intact strata and mass-transport deposits (MTDs). In a mid-slope position, however, we found evidence for a ∼ 4-km³-sized landslide complex that comprises several MTDs from translational transport of coherent sediment bodies over short distances. Morphometric analysis of these MTDs and their source scars indicates that this part of the northeast Cretan slope can be characterized as a cohesive slope. Furthermore, we reconstruct retrogressive development for this complex and determine a critical slope angle for both pre-conditioning of failure and subsequent landslide deposition near source scars. Consequently, data imply that the investigated shallower slope is stable due to low angles in the order of 3°, whereas 5°-inclined mid-slope portions favour both slope destabilization and landslide deposition. The failed mid-slope parts are dominated by sediment truncations from faults almost correlating with the orientation of head- and sidewalls of scars. We suggest that cohesive landslides and MTDs are generated and preserved, respectively, in such critical slope regions. If once generated, cohesive landslides reach the lower slope further downslope that exceeds the threshold gradient for MTD deposition (∼ 5°), they are transported all the way down to the foot of the slope and disintegrate to mass flows. From these observations we suggest that the mass-wasting history of the investigated Cretan slope area over a longer period of time is characterized by repeated sediment erosion and transport into the deeper Cretan Sea basin. The relocation of the critical slope portion in upslope direction and therefore recurrence of mass-wasting events is thereby likely controlled by the progressive steepening of the slope. This mechanism and restriction of sediment failure to narrow, critically-inclined and relocating slope portions likely explains how such an active margin setting can exhibit only scarce findings of MTDs on the slope despite an expected, extensive and widespread mass wasting.

    Beschreibung: Gaining reliable estimates of how long fish early life stages can survive without feeding and how starvation rate and time until death are influenced by body size, temperature and species is critical to understanding processes controlling mortality in the sea. The present study is an across-species analysis of starvation-induced changes in biochemical condition in early life stages of nine marine and freshwater fishes. Data were compiled on changes in body size (dry weight, DW) and biochemical condition (standardized RNA–DNA ratio, sRD) throughout the course of starvation of yolk-sac and feeding larvae and juveniles in the laboratory. In all cases, the mean biochemical condition of groups decreased exponentially with starvation time, regardless of initial condition and endogenous yolk reserves. A starvation rate for individuals was estimated from discrete 75th percentiles of sampled populations versus time (degree-days, Dd). The 10th percentile of sRD successfully approximated the lowest, life-stage-specific biochemical condition (the edge of death). Temperature could explain 59% of the variability in time to death whereas DW had no effect. Species and life-stage-specific differences in starvation parameters suggest selective adaptation to food deprivation. Previously published, interspecific functions predicting the relationship between growth rate and sRD in feeding fish larvae do not apply to individuals experiencing prolonged food deprivation. Starvation rate, edge of death, and time to death are viable proxies for the physiological processes under food deprivation of individual fish pre-recruits in the laboratory and provide useful metrics for research on the role of starvation in the sea. Highlights ► Biochemical condition (RNA–DNA ratio) decreases exponentially during starvation. ► Starvation parameters of individuals can be derived from data collected on groups. ► Physiological rates of starvation compare well across a broad range of temperatures. ► Species and life stages specific starvation parameters indicate selective adaptation.
