Derivatives of Imidazo[4,5-b]- and of Imidazo[4,5-c]pyridinesCondensation of 2,3-diamino- and of 3,4-diaminopyridine with chloracetonitrile in polyphosphoric acid yields 2-chloromethyl-imidazo[4,5-b]- (1a) and -[4,5-c]pyridine (5), respectively. By reaction with various secondary amines, the corresponding amino derivatives 2 or 6 are obtained. These compounds have been converted into their sodium salts, which react with benzyl chlorides. To elucidate the structure of the reaction products, NMR and UV spectroscopy as well as HMO calculations are used. It is found that benzylation in the imidazo[4,5-b]pyridine series occurs preferentially at N-3 (3) and, to a smaller extent, at N-1 (4). In the case of imidazo-[4,5-c]pyridines, benzylation at N-5 has also been observed (9).

Kondensation von 2.3- bzw. 3.4-Diamino-pyridin mit Chloracetonitril in Polyphosphorsäure ergibt 2-Chlormethyl-imidazo[4.5-b]- (1a) bzw.-[4.5-c]pyridin (5). Durch Umsetzung mit sekundären Aminen werden die entsprechenden Amino-Derivate 2 bzw. 6 erhalten. Diese werden als Na-Salze mit Benzylchloriden umgesetzt; die Konstitution der Reaktionsprodukte wird mit Hilfe von NMR-, UV- und HMO-Untersuchungen aufgeklärt. Es stellt sich heraus, daß in der Reihe der Imidazo[4.5-b]pyridine vorwiegend Benzylierung an N-3 (zu 3) und in geringerem Maße an N-1 (zu 4) stattfindet. In der Reihe der Imidazo[4.5-c]pyridine tritt daneben teilweise Benzylierung an N-5 (zu 9) ein.
