    Notes: We have reexamined the possible role of the Na+/H+ antiport in the cellular response by PC12 pheochromaytoma cells to nerve growth factor (NGF). In contrast to previous reports, we observe no activation of Na+/H+ exchange in these cells, using a very sensitive assay based on the measurement of cytoplasmic pH with dimethylfluorescein dextran (Rothenberg et al., J. Biol Chem., 258: 4883-4809, 1983). Our measurements indicate that the PC12 pheochromacytoma cells, under all conditions tested, show a high rate of Na+/H+ exchange. The discrepancy between these observations and previous experiments could be due to differences in cells in different laboratories, but also to changes in cell adhesion induced by NGF. We describe conditions where intracellular pH and rates of Na+ uptake can be measured reliably in PC12 cells with adequate controls for cell adhesion. We conclude that activation of Na+/H+ exchange is neither sufficient nor required for the differentiation of PC12 cells induced by NGF.

    Notes: The effect of removal of PC12 cell nuclei on neurite outgrowth was studied. Enucleation (80-90%) was accomplished in the presence of cytochalasin B using a centrifugation technique that exploited the very tight adhesivity of PC12 cells for a substratum composed of an extracellular matrix secreted by bovine corneal endothelial cells in response to epidermal growth factor treatment. Neither nucleated nor enucleated PC 12 cells showed significant neurite outgrowth on this particular matrix in the absence of nerve growth factor. In the presence of nerve growth factor both PC12 cell types initiated neurite outgrowth, but whereas neurites from nucleated cells grew continuously for two days, those from enucleated cells reached a maximum length after one day. The results suggest that neurite initiation but not continued neurite growth or stabilization can occur in the absence of transcription.

    Notes: A significant portion of published literature is dedicated to describing the cloning and the characterization of proteins involved in the progression of the cell cycle, which govern cell growth both in cancer and normal ontogenesis. With this abundance of information, the cascading pathways of molecular events that occur in the cell cycle are proving to be exceedingly complicated. The purpose of this conference was to attract the leading clinical and basic science investigators in the growth control field with a final goal to determine how this current wealth of knowledge can be used to impact upon patient care and management by the design of novel adjuvant therapeutics specifically targeted at tumor cells and the identification of molecular diagnostic and/or prognostic markers in an efficient and cost effective manner.     Notes: A series of nine features of the shoulder girdle, chosen as having functional significance in relation to the movements of the shoulder in arboreal locomotion, have been studied in 1188 specimens of 194 genera of mammals. The features were defined metrically and examined by means of a multivariate statistical technique: viz. canonical analysis. The study has shown that those mammals which are nonarboreal differ considerably among themselves and form the arboreal forms. But the myriad shapes of the shoulder girdle in a wide range of mammals (e.g. some marsupials, edentates, rodents, carnivores and primates) which climb or forage in trees, can be summarized mathematically by a very small number of similar canonical variates. This information correlates well with that of a previous series of studies carried out on the primates alone.The biological information that was postulated as being reflected by the individual canonical variates for the primates is also apparent for the arboreal mammals. The different variates separate the forms in ways which are consonant with what is known about the function of the shoulder in locomotion. Aspects of the shape of the shoulder defined by the analysis appear to be discernible from an examination of the contribution of the original variables to each individual canonical variate. This seems to confirm that the shape of the shoulder girdle within a very wide range of mammals is limited by a very small number of underlying factors of biological significance.One interpretation of the results suggests that the genetic model of the mammalian shoulder may have been sufficiently fixed at an early stage in the evolution of the class as to place considerable constraints upon the subsequent evolution of the shoulder in the different Orders.

    Notes: Recent hypotheses about the mechanisms of adaptation of bone to impressed mechanical forces differ according to the extent to which tensile stresses are thought to be functionally important in bone. A pilot study of three anatomical regions by means of the photoelastic analogy suggests the possibility that net tension rarely exists in significantly large regions of bones during normal function.Thus the examination of two situations (opposite linear attachment of muscles to the lips of bony crests and opposite areal attachment of muscles to either side of thin bony plates) suggests that in rare cases where precise anatomical architecture is such that net tension may be present, then bone is not found; such regions consist of appropriate collagenous structures.In the third situation (opposite attachment of tendons to a sesamoid bone) where it would appear that net tension ought to exist, it is suggested that it is likely that it does not. Net tension probably occurs only in tendons running in tightly curved bony grooves: those very situations where sesamoid bones are not present.

    Notes: Comparison of mesopodial skeletal patterns found in native and regenerated limbs of the salamander Plethodon cinereus reveals variant patterns unique to each group. Variant patterns in native limbs are based on fusions between laterally adjacent elements (i.e., in the anteroposterior axis). Variant patterns in the mesopodia of regenerated limbs usually exhibit fusions among proximodistally adjacent elements. Analysis of regenerates derived from limb amputation at different levels shows that the axis of fusion between regenerated mesopodial elements remains the same (i.e., proximodistal) independent of amputation level. However, the frequency of specific fusion combinations is unexpectedly sensitive to amputation level. Proximal (stylopodial) amputation results in mesopodial patterns with predominantly preaxial fusion combinations; distal amputation produces mesopodial patterns with predominantly postaxial fusion combinations. This finding is discussed in the context of other recent studies in which amputation level influenced limb regeneration patterning.

    Notes: The mechanism by which transforming growth factor beta (TGFβ) exerts growth stimulatory effects was examined in C3H/10T1/2 mouse fibroblasts by study of cell cycle regulation of the retinoblastoma gene product (p110Rb) and transcriptional regulation of the p110Rb-associated transcription factor, E2F. Northern blotting analysis shows that TGFβ and/or epidermal growth factor (EGF) stimulate by three to sixfold the level of Rb mRNA which is also reflected by the increased levels of p110Rb. p110Rb becomes phosphorylated in mid-G1 and further phosphorylated at the G1/S transition. Hyperphosphorylation of p110Rb by TGFβ can be observed when cells are in S phase. TGFβ stimulates by three to fourfold the activity of cdk2 kinase consistent with the observed phosphorylation of p110Rb and also with the possibilit that the kinase is involved in phosphorylating p110Rb close to the G1/S transition. Thus, TGFβ as a growth stimulator induces, as does EGF, the phosphorylation of p110Rb during cell cycle progression. Transient transfection of E2F promoter constructs was used to analyze the effect of TGFβ on the modulation of E2F-mediated transcription. The data revealed that TGFβ can stimulate wild-type adenoviral E2 promoter activity by 12-fold. Taken together, TGFβ-induced phosphorylation of p110Rb in mouse fibroblasts appears to exert a positive regulatory function upon genes that have a pivotal role in the G1/S transition of the cell cycle. © 1994 Wiley-Liss, Inc.
