    Notes: Summary We have analyzed the development of neuronal projections inDrosophila by fusing the gene encodingDrosophila kinesin, a microtubule-associated motor protein, toEscherichia coli lacZ, and employing the resulting chimeric protein as a reporter molecule for labelling cells by the “enhancer-trap” method. Expression of kinesin-β-galactosidase in neurons has afforded a detailed view of the morphologies and projections of neurons. The images of cells provided by this method will facilitate anatomical and genetic investigations of theDrosophila nervous system as well as other cell types.

    Notes: Summary Despite the very considerable difficulties presented by the basic molluscan anatomy and the possession of a blood pigment with an oxygen carrying capacity that never exceeds 4.5 vols%, the cephalopod circulatory system contrives to deliver oxygen at a rate fully comparable with that of an active fish. This is achieved by adding accessory pumps to push blood through the gills, by a multiplicity of pulsatile veins and by raising the systemic blood pressure considerably above the levels found in other molluscs. Detailed control of blood distribution is a necessity in a system where the peripheral resistences may be expected to change dramatically when the animal starts to move and large parts of the central nervous system are apparently dedicated to this task. In this account we have reviewed blood pressure and flow at rest and in exercise. We have further examined the evidence which indicates how the animals modulate the cardiac output, drawing attention to the very different response found in cephalopods and the higher vertebrates.

    Notes: Abstract  Although the validation process necessary to ensure that an analytical method is fit for purpose is universal, the emphasis placed on different aspects of that process will vary according to the end use for which the analytical procedure is designed. It therefore becomes difficult to produce a standard method validation protocol which will be totally applicable to all analytical methods. It is probable that far more than 30% of the methods in routine laboratory use have not been validated to an appropriate level to suit the problem at hand. This situation needs to change and a practical assessment of the degree to which a method requires to be validated is the first step to a reliable and cost effective analytical industry.

    Notes: Abstract During July of 1983, 1986, and 1987, we measured rates of oxygen consumption of 234 individuals of 17 species of midwater crustaceans (orders Decapoda, Mysidacea, and Euphausiacea) off the Hawaiian islands at depths from the surface to greater than 1200 m. The routine metabolic rates declined with increasing depths of the species' occurrence to an extent greater than could be accounted for by depth-related changes in body size or water temperature. Most species appeared able to regulate their oxygen consumption down to the lowest oxygen partial pressures found in their depth range (20 mm Hg O2), but did not regulate to such low oxygen partial pressures as did similar midwater crustaceans off California, where oxygen levels reach as low as 6 mm Hg. Metabolic rates of the shallower-living, but not the deepest-living Hawaiian crustaceans were significantly higher than those of Californian crustaceans. This is interpreted as indicating that the metabolic rates of midwater crustaceans are not adapted specifically to differing levels of primary production and that the decline with depth of metabolic rates in these species is not the result of food limitation at depth. The data are, however, consistent with the hypothesis that lower metabolic rates at depth are due to the relaxation of selection pressures relating to visual predation near the surface.

    Notes: Abstract The effect of feeding the flagellate Isochrysis galbana (Parke; clone T-Iso) of modified lipid composition on the growth and lipid composition of juvenile scallops [Placopecten magellanicus (Gmelin)] was investigated in the spring of 1993. I. galbana grown in 85-liter cage culture turbidostats under conditions of nitrogen limitation had a significantly higher total lipid content than when grown under nutrient-replete conditions. This was due mainly to a doubling in the amount of less unsaturated triacylglycerol in the cells. The concentrations of methyl and ethyl ketones were also greater in nitrogen-limited cells. Diets of nitrogen-limited I. galbana and nutrient-replete I. galbana grown in continuous and semi-continuous cultures were compared. Scallop juveniles were batch fed daily, and measurements of ingestion were determined. Samples of juveniles were removed periodically for determination of organic weight. The juveniles did not grow when fed nitrogen-limited or nutrient-replete I. galbana alone; however, when each diet was supplemented with 20% of the diatom Chaetoceros muelleri (Lemm.), there was a significant increase in growth in the juveniles receiving the nitrogen-limited I. galbana compared with juveniles on other diets. In comparison with I. galbana,  C. muelleri provided a rich source of carbohydrates and the essential fatty acid 20:4ω6. This study shows the importance of providing optimal dietary levels of ω3 and ω6 polyunsaturated fatty acids, as well as less unsaturated fatty acids and carbohydrates.

    Notes: Abstract Euthecosomatous pteropods were collected in successive oblique plankton tows with 1 m diameter nets equipped with No. 0, No. 6, and No. 20 nylon mesh. Comparisons of samples revealed that the same species were collected by all 3 nets, but the No. 0 and No. 6 nets failed to collect veligers and juveniles of Spiratella inflata and Creseis virgula conica. Collections with nets of larger mesh size may erroneously suggest seasonal variations in population densities due, not to actual changes in total density, but to changes in the size of individuals and changes in the relative numbers of the different life stages. A large diameter net equipped with No. 20 mesh is recommended for work that includes the smallest life stages of tropical pteropods.
