    Notes: We report cloning and sequencing of gene ps1 encoding a versatile peroxidase combining catalytic properties of lignin peroxidase (LiP) and manganese peroxidase (MnP) isolated from lignocellulose cultures of the white-rot fungus Pleurotus eryngii. The gene contains 15 putative introns, and the deduced amino acid sequence consists of a 339-residue mature protein with a 31-residue signal peptide. Several putative response elements were identified in the promoter region. Amino acid residues involved in oxidation of Mn2+ and aromatic substrates by direct electron transfer to heme and long-range electron transfer from superficial residues as predicted by analogy with Phanerochaete chrysosporium MnP and LiP, respectively. A dendrogram is presented illustrating sequence relationships between 29 fungal peroxidases.

    Notes: RNase U2 is an endoribonuclease secreted by the fungus Ustilago sphaerogena. Its genomic DNA (rnu2), containing an intron of 116 bp, has been isolated and cloned. The corresponding cDNA has also been synthesized. The recombinant RNase U2 was successfully produced in Pichia pastoris, fused to the yeast alkaline phosphatase signal peptide. The recombinant RNase U2, purified by affinity chromatography, contains three extra amino acids at its amino-terminal end and retains the enzymatic and spectroscopic properties of the natural fungal protein.

    Notes: Moderate pressure techniques (P=20 kbar) have been used to prepare Tl2−xCdxMn2O7 (0≤x≤0.4) pyrochlore-like materials. The x=0.2 compound has been characterized by neutron powder diffraction, magnetic, magnetotransport, and Hall measurements. This material is ferromagnetic below TC=110 K. Both electrical resistance and magnetoresistance (MR) are enhanced with respect to stoichiometric Tl2Mn2O7, due to the drastic reduction in the number of carriers (electrons) induced by hole doping. MR(0.5 T) is higher than 106% at 120 K, and MR(9 T) is 30% at room temperature. We show that hole doped derivatives of Tl2Mn2O7 are promising candidates in which to search for large bulk magnetoresistance. © 2000 American Institute of Physics.

    Notes: An experiment was carried out with Cichlasoma urophthalmus (Günther) juveniles to determine the phosphorus requirement and its interaction with dietary calcium. Twelve isoenergetic and isoproteic diets were prepared using a basal artificial diet containing vitamin-free casein, dextrin, starch, corn oil, fish oil, vitamin mixture and a mineral mixture free of calcium and phosphorus. Calcium and phosphorus levels were determined in the casein. To the basal diets were added different concentrations of phosphorus as potassium monophosphate (0.5, 1.0, 1.5 and 2.5 g kg–1) and calcium as calcium carbonate (0.5, 0.75, 1.0, 1.5, 2.0, 2.5, 3.0 and 4.0 g kg–1). These concentrations resulted in varying Ca–P ratios (1:1, 1.33:1, 1.5:1, 1.6:1 and 2.0:1). Calcium and phosphorus concentrations in the water were 84 mg kg–1 and 0.003 mg kg–1, respectively. The diet with 0.5 g kg–1 phosphorus resulted in deficiency signs such as reduced growth, high conversion ratio, high fat content and low bone mineralization. Increased levels of dietary calcium and phosphorus both gave improved growth and mineralization. Mineralization continued to increase with dietary phosphorus levels above that required for maximum growth. The optimum level of phosphorus in the diet was 1.5 g kg–1, the optimum calcium level was 1.8 g kg–1 and the optimum Ca–P ratio was 1.3. Carcass lipid levels were inversely related to dietary phosphorus.

    Notes: It has been shown previously that gibberellins (GAs) mediate the phytochrome (Phy) control of cowpea (Vigna sinensis L.) epicotyl elongation induced by end-of-day (EOD)-far-red light (FR). In the present work, the EOD-FR effect on GA metabolism and GA levels in cowpea has been investigated. GA1, GA8, GA19 and GA20 were identified in epicotyls, and GA1, GA19, GA20 and GA29-catabolite in leaves of 6-day-old cowpea seedlings. The content of GA1 in the epicotyl paralleled the decrease of its growth rate, supporting the hypothesis that this is the GA bioactive in controlling cowpea epicotyl elongation. FR enhanced both the amount of [3H]GA1 in the epicotyl produced from applied [3H]GA20, and that of applied [3H]GA1 that remained unmetabolized in epicotyl explants, suggesting that Phy may regulate the inactivation of GA1. In agreement with this effect of light on GA1 metabolism, the contents of GA1 in the epicotyl remained higher in FR-treated than in R-treated explants. Moreover, in intact seedlings EOD-FR treatment increased both epicotyl elongation and GA1 content in the responsive epicotyl, whereas it was not altered in the leaves. These results show, for the first time, that photostable Phys modulate the stem elongation in light-grown plants by locally controlling the GA1 levels through regulation of its inactivation.

    Notes: As water and nutrient uptake should be related in the response of plants to salinity, the aim of this paper is to establish whether or not aquaporin functionality is related to H+-ATPase activity in root cells of pepper (Capsicum annuum L.) plants. Thus, H+-ATPase activity was measured in plasma membrane vesicles isolated from roots and aquaporin functionality was measured using a cell pressure probe in intact roots. Salinity was applied as 60 mM NaCl or 60 mM KCl, to determine which ion (Na+, K+ or Cl−) is producing the effects. We also investigated whether the effects of both salts were ameliorated by Ca2+. Similar results were obtained for cell hydraulic conductivity, Lpc, and H+-ATPase activity, large reductions in the presence at NaCl or KCl and an ameliorative effect of Ca2+. However, fusicoccin (an activator of H+-ATPase) did not alter osmotic water permeability of protoplasts isolated from roots. Addition of Hg2+ inhibited both ATPase and aquaporins, but ATPase also contains Hg-binding sites. Therefore, the results indicate that H+-ATPase and aquaporin activities may not be related in pepper plants.

    Notes: After analysing a sample of 442 of the Spanish biggest manufacturing companies, some evidence about the influence of total quality management (TQM) on the companies' operating performance has been obtained. However, companies applying TQM together with the ISO 9000 standards did not show positive results. This fact leads to the consideration that, despite the beliefs about ISO 9000 as a good first step in the way of implementing TQM, once implemented, some of the ISO 9000 principles are contradictory with TQM philosophy. These not congruent systems applied together would drive the company to obtain less benefits than the use of only one of them. The study concludes that when ISO 9000 and TQM are applied simultaneously, the resultant benefits to the company are not better than those experienced if either system were applied in isolation.

    Notes: Abstract A study of the electrochemical lithium insertion in two polymorphs of a reduced molybdenum oxide, Mo4O11, is presented in this work. When used as active materials in cells discharged down to 1 V vs. Li+/Li, both forms, the orthorhombic γ-Mo4O11 and the monoclinic γ′-Mo4O11, incorporated a similar number of lithium atoms per metal atom (Li/Mo=2.12 and 2.25, respectively). Step potential electrochemical spectroscopy experiments proved that the insertion reaction proceeds in both cases through different mechanisms. In situ X-ray diffraction studies showed an almost complete loss of crystallinity of both compounds after the first discharge, leading to amorphous materials with different electrochemical behaviour on cycling. When discharged to low potentials (0.5 V vs. Li+/Li), the γ′-Mo4O11 polymorph showed very good cycling behaviour for at least five lithium atoms per formula unit, corresponding to a specific capacity of 230 Ah/kg after seven complete charge-discharge cycles.

    Notes: Abstract New swath bathymetric, multichannel seismic and magnetic data reveal the complexity of the intersection between the extinct West Scotia Ridge (WSR) and the Shackleton Fracture Zone (SFZ), a first-order NW-SE trending high-relief ridge cutting across the Drake Passage. The SFZ is composed of shallow, ridge segments and depressions, largely parallel to the fracture zone with an `en echelon' pattern in plan view. These features are bounded by tectonic lineaments, interpreted as faults. The axial valley of the spreading center intersects the fracture zone in a complex area of deformation, where N120° E lineaments and E–W faults anastomose on both sides of the intersection. The fracture zone developed within an extensional regime, which facilitated the formation of oceanic transverse ridges parallel to the fracture zone and depressions attributed to pull-apart basins, bounded by normal and strike-slip faults. On the multichannel seismic (MCS) profiles, the igneous crust is well stratified, with numerous discontinuous high-amplitude reflectors and many irregular diffractions at the top, and a thicker layer below. The latter has sparse and weak reflectors, although it locally contains strong, dipping reflections. A bright, slightly undulating reflector observed below the spreading center axial valley at about 0.75 s (twt) depth in the igneous crust is interpreted as an indication of the relict axial magma chamber. Deep, high-amplitude subhorizontal and slightly dipping reflections are observed between 1.8 and 3.2 s (twt) below sea floor, but are preferentially located at about 2.8–3.0 s (twt) depth. Where these reflections are more continuous they may represent the Mohorovicic seismic discontinuity. More locally, short (2–3 km long), very high-amplitude reflections observed at 3.6 and 4.3 s (twt) depth below sea floor are attributed to an interlayered upper mantle transition zone. The MCS profiles also show a pattern of regularly spaced, steep-inclined reflectors, which cut across layers 2 and 3 of the oceanic crust. These reflectors are attributed to deformation under a transpressional regime that developed along the SFZ, shortly after spreading ceased at the WSR. Magnetic anomalies 5 to 5 E may be confidently identified on the flanks of the WSR. Our spreading model assumes slow rates (ca. 10–20 mm/yr), with slight asymmetries favoring the southeastern flank between 5C and 5, and the northwestern flank between 5 and extinction. The spreading rate asymmetry means that accretion was slower during formation of the steeper, shallower, southeastern flank than of the northwestern flank.
