    Notizen: The Rb2/p130 protein has been shown to have a high sequence homology with the retinoblastoma gene product (pRb), one of the most well-characterized tumor suppressor genes, and with pRb-related p107, especially in their conserved pocket domains, which display a primary role in the function of these proteins. In this study, we report on the biochemical and immunocytochemical characterization of the Rb2/p130 protein, using a polyclonal antibody developed against its “spacer” region included in the pocket domain of the whole protein. We show that pRb/p130 is a phosphoprotein located at the nuclear level and that its phosphorylation pathway can be dramatically reduced by phosphatase treatment. Moreover pRb/p130, with p107, with p107, is one of the major targets of the E1A viral oncoprotein-associated kinase activity, showing a phosphorylation pattern which is modulated during the cell cycle, reaching a peak of activation at the onset of S-phase. © 1995 Wiley-Liss, Inc.

    Notizen: Measurements were made of mass transfer coefficients with gas and liquid phase controlling resistance using a concurrent flow packed column with a wide range of operating conditions and four types of packing.Mass transfer coefficients were much larger than those obtainable in countercurrent towers.The results were correlated with dissipated energy values. The correlations obtained can be used to determine absorption efficiency. Experimental results show that the use of a high velocity concurrent flow column is particularly suitable when mass transfer is controlled by the liquid phase resistance with chemical reaction in the liquid.

    Notizen: A new approach was used to determine the reaction products of methylcobalamin and ionic mercury: purge-and-trap gas chromatography in line with Fourier transform infrared spectroscopy (PT GC/FTIR). This technique simultaneously and specifically determines the spectrum of dimethylmercury (DMeHg) and methylmercury produced by the reaction. No interference from other known organic mercury species could be detected. The method is different from others because it does not require solvent extraction of the organomercurials from aqueous solution, but relies on immediate volatilization from the reaction vessel by addition of 100 μl of 10 mM NaBH4. The sample was purged with nitrogen for 10 min. The volatile species of mercury were trapped in a column at -120°C, injected into the gas chromatograph and detected by FTIR. The efficiency of DMeHg and MeHg formation depended on different parameters: pH, temperature, reaction time, and the methylcobalamin/ionic mercury ratio. The initial reaction product was MeHg which was further transformed to DMeHg. The first methylation rate was two times faster than the second. MeHg formed first, reaching a maximum at higher temperatures (28°C and 37°C) and later decreasing as DMeHg formed. At lower temperatures (20°C) the rate of MeHg formation was slower, being similar to the formation rate of DMeHg. Different species of inorganic mercury such as HgSO4, Hg(NO3)2, Hg(SCN)2, HgCl2 and Hgl2 were used to study differences in methylation by methylcobalamin under standard conditions of acidity, temperature and cofactor Hg(II) ratio.

    Notizen: The results are presented of an interlaboratory study on methylmercury (MeHg) in sediment carried out by a group of European laboratories within the framework of a project managed by the EC Standards, Measurements and Testing Programme (formerly BCR). The aim of this exercise was to evaluate the performance of current methods used for MeHg determination in sediment in order to improve the state-of-the-art prior to the certification of a candidate reference material. The paper describes the organization of the interlaboratory study, the preparation of the sediment material used, the techniques evaluated and the results obtained by the participating laboratories. The outcome of the collaborative project showed that certification could be contemplated, providing that certain analytical techniques were optimized, especially with regard to extraction methods.

    Notizen: Amorphous films of syndiotactic polystyrene were crystallized in dichloromethane, to obtain samples in the clathrate δ form; successively the samples were either annealed at temperatures below the glass transition temperature (Tg) or extracted with acetone. The samples obtained show, in the X-ray analysis, crystalline structures tending toward the “emptied” δ form, free of solvent molecules, as already described for sPS. The study of the transport properties, diffusion and sorption of dichloromethane vapor, shows that the annealed samples are characterized by a slightly lower diffusion coefficient, but much higher sorption at low activity of the vapour. This high sorption is explained by the penetration of solvent molecules both into the amorphous and the “emptied” crystalline phase, producing again the clathrate form at low vapour activity. The high sorption capability makes these samples promising as sorption media to remove traces of polluting chlorinated solvents both in the liquid and in the vapour phase.
