    Beschreibung: A phytochemical investigation of the BuOH-soluble fraction of the EtOH extract from the stems of Glycosmis pentaphylla resulted in the isolation of three new phenolic glycosides, glycopentosides D–F ( 1 – 3 , resp.). Their structures were determined by using spectroscopic analysis including UV, 1 H- and 13 C-NMR, DEPT, COSY, ROESY, HMBC, HSQC, HR-ESI-MS, and acid hydrolysis.

    Beschreibung: Abstract In this study, first‐principles calculations were performed to study the stability, mechanical property, electronic structure and lattice dynamics of β‐Si3(Cx,N1−x)4 silicon carbonitride. The solubility of carbon in β‐Si3(Cx,N1−x)4 having a stable structure is shown to be about 15 at.%. Within the limit of solubility, an increase in carbon concentration in β‐Si3(Cx,N1−x)4 will lead to a decrease in the Young's modulus and density and an increase in the Poisson's ratio. The study of deformation behavior shows that the most likely slip system of is on prismatic plane rather than on basal plane. This feature of β‐Si3(Cx,N1−x)4 is similar to WC. In addition, the ductility and fracture toughness of β‐Si3(Cx,N1−x)4 can be optimized by controlling the carbon concentration. The improvement in ductility and fracture toughness can be attributed to the formation of metallic bonds by the incorporation of carbon atoms. The lattice dynamics study shows that the structural stability of β‐Si3(Cx,N1−x)4 is controlled by energy stability criteria under stress‐free condition. In the stressed state, the structural stability of β‐Si3(Cx,N1−x)4 is controlled by the elastic stability criteria. Subsequently, the β‐Si3(Cx,N1−x)4 solid solution was prepared by self‐propagating high‐temperature synthesis (SHS), and the carbon‐concentration‐dependent mechanical properties were consistent with the first‐principles calculations. The maximum fracture toughness of 10.4 MPa·m0.5 was obtained in β‐Si3(Cx,N1−x)4 at carbon concentration of 5 wt%, which means that the solid solution toughening can be used as a supplement to crack bridging toughening and phase transition toughening for ceramic toughening. The results obtained in this study reveal that the β‐Si3(Cx,N1−x)4 solid solution is a promising candidate for high‐speed ceramic bearings.

    Beschreibung: Three new macrocyclic β -dihydroagarofuran-type sesquiterpene pyridine alkaloids, fortuneines A ( 1 ), B ( 2 ), and C ( 3 ), together with the four known alkaloids wilfornine E ( 4 ), aquifoliunine E-I ( 5 ), euoverrine B ( 6 ), and euojaponine I ( 7 ), were isolated from the aerial parts of Euonymus fortunei. Their structures were elucidated by spectroscopic methods, including HR-ESI-MS, 1 H- and 13 C-NMR, DEPT, 1 H, 1 H-COSY, HSQC, HMBC, and ROESY. This is the first isolation of the above sesquiterpene pyridine alkaloids from this plant, except for compound 6 .

    Beschreibung: ABSTRACT We developed a computational approach to detect and segment cytoplasm in microscopic images of skeletal muscle fibers. The computational approach provides computer-aided analysis of cytoplasm objects in muscle fiber images to facilitate biomedical research. Cytoplasm in muscle fibers plays an important role in maintaining the functioning and health of muscular tissues. Therefore, cytoplasm is often used as a marker in broad applications of musculoskeletal research, including our search on treatment of muscular disorders such as Duchenne muscular dystrophy, a disease that has no available treatment. However, it is often challenging to analyze cytoplasm and quantify it given the large number of images typically generated in experiments and the large number of muscle fibers contained in each image. Manual analysis is not only time consuming but also prone to human errors. In this work we developed a computational approach to detect and segment the longitudinal sections of cytoplasm based on a modified graph cuts technique and iterative splitting method to extract cytoplasm objects from the background. First, cytoplasm objects are extracted from the background using the modified graph cuts technique which is designed to optimize an energy function. Second, an iterative splitting method is designed to separate the touching or adjacent cytoplasm objects from the results of graph cuts. We tested the computational approach on real data from in vitro experiments and found that it can achieve satisfactory performance in terms of precision and recall rates. Microsc. Res.     Beschreibung: A new xanthone derivative, garcinexanthone F ( 1 ), which was found to possess an α , β -unsaturated γ -lactone moiety, and a new bisxanthone, bigarcinenone B ( 2 ), with a terpene bridge providing the xanthone xanthone linkage, were isolated from the bark of Garcinia xanthochymus. Their structures were elucidated by spectroscopic methods, especially 2D-NMR techniques. The antioxidant assay in vitro showed that compounds 1 and 2 exhibited significant scavenging activities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical with IC 50 values of 22.32 and 20.14 μ M , and against HO . radical with IC 50 values of 1.16 and 2.85 μ M , respectively.

    Beschreibung: ABSTRACT Muscle fiber images play an important role in the medical diagnosis and treatment of many muscular diseases. The number of nuclei in skeletal muscle fiber images is a key bio-marker of the diagnosis of muscular dystrophy. In nuclei segmentation one primary challenge is to correctly separate the clustered nuclei. In this article, we developed an image processing pipeline to automatically detect, segment, and analyze nuclei in microscopic image of muscle fibers. The pipeline consists of image pre-processing, identification of isolated nuclei, identification and segmentation of clustered nuclei, and quantitative analysis. Nuclei are initially extracted from background by using local Otsu's threshold. Based on analysis of morphological features of the isolated nuclei, including their areas, compactness, and major axis lengths, a Bayesian network is trained and applied to identify isolated nuclei from clustered nuclei and artifacts in all the images. Then a two-step refined watershed algorithm is applied to segment clustered nuclei. After segmentation, the nuclei can be quantified for statistical analysis. Comparing the segmented results with those of manual analysis and an existing technique, we find that our proposed image processing pipeline achieves good performance with high accuracy and precision. The presented image processing pipeline can therefore help biologists increase their throughput and objectivity in analyzing large numbers of nuclei in muscle fiber images. Microsc. Res.     Beschreibung: Mesenchymal stromal cells (MSCs) are promising candidate donor cells for replacement of cardiomyocyte loss during ischemia and in vitro generation of myocardial tissue. We have successfully isolated MSCs from the discarded neonatal thymus gland during cardiac surgery. The thymus MSCs were characterized by cell-surface antigen expression. These cells have high ability for proliferation and are able to differentiate into osteoblasts and adipocytes in vitro. For cardiac differentiation, the cells were divided into 3 groups: untreated control; 5-azacytidine group and sequential exposure to 5-azacytidine, bone morphogenetic protein 4, and basic fibroblast growth factor. Thymus MSCs showed a fibrolast-like morphology and some differentiated cells increased in size, formed a ball-like appearance over time and spontaneously contracting cells were observed in sequential exposure group. Immunostaining studies, cardiac specific genes/ protein expression confirmed the cardiomyocyte phenotype of the differentiated cells. These results demonstrate that thymus MSCs can be a promising cellular source for cardiac cell therapy and tissue engineering. This article is protected by copyright. All rights reserved
