Notizen: Survey data collected during a grassland-recording investigation showed a strong positive correlation between nitrogen input and the output of utilized starch equivalent per acre. At the levels of output covered, sward type was not very important in relation to total yield. Herbage species were important for other reasons and the choice of seeds mixtures should be determined by factors such as earliness and lateness of growth and resistance to disease and drought. The survey showed that well-managed ley and permanent pasture are both capable of producing at a high level. A disturbing feature of the results was the low production of most spring resowings, primarily as a result of poor management. Although the records were obtained from an above-average group of farms, the results have general application. Potential responses to fertilizers may be even greater on less intensive farms, especially with regard to phosphate and potash. However, the level of production of a field will always depend finally on the level of management exercised by the farmer himself.

Notizen: Mesoplodon traversii (Gray, 1874) is shown to be a senior synonym of the recently described beaked whale Mesoplodon hahamondi Reyes et al., 1995 on the basis of a phylogenetic analysis of mitochondrial DNA control region sequences. The mandible and teeth of M. traversii, first reported in 1873 by Hector as Dolichodon layardii. are redescribed. The species can be distinguished by features of the calvaria; including the large jugal, broad rostrum, and small distance between premaxillary foramina. The male teeth, which are large and spade-shaped with a strong terminal denticle, are also diagnostic. M. traversii is known only from Pitt Island and White Island, New Zealand and Robinson Crusoe Island, Juan Fernandez Archipelago, Chile.

Notizen: Mesoplodon perrini, a new species of beaked whale is described on the basis of five animals stranded on the coast of California (between 32°55′N, 117°15′W and 36°37′N, 121°55′W) from May 1975 to September 1997. Four of these animals were initially identified as Hector's beaked whales M. hectori based on cranial morphology (Mead 1981). A fifth specimen was initially identified as a neonate Cuvier's beaked whale Ziphius cavirostris based on external features. These specimens were first recognized as representatives of an undescribed species through phylogenetic analysis of mitochondrial (mt) DNA control region and cytochrome b sequence data. Although similar morphologically, the genetic data do not support a close evolutionary relationship between M. perrini and M. hectori. Instead, these data suggest a possible sister species relationship with the lesser beaked whale M. peruvianus. Sightings of two small beaked whales off California in the 1970s which were tentatively identified as M. hectori are also likely to be M. perrini. We suggest that M. hectori is confined to the Southern Hemisphere, while M. perrini is known to date only from the North Pacific.

Notizen: SUMMARY. Temperature and current discharge regulated phytoplanktonic concentration, chlorophyll-a concentration, the light-saturated rate of photosynthesis (Pmax), and photosynthetic capacity (Pcap) in the Mississippi River at Prairie Island, Minnesota. The chlorophyll-a maximum was 48 mg m−3 in 1975, a wet year with a high current discharge, and 190 mg m−3 in 1976, a relatively dry year. The highest values of Pmax were 0.37 (mgO2 I−1h−1) in 1975 and 1.60 in 1976. Pcap varied from 3 to 21 (gO2 per g chlorophyll-a h−1) both years, and its value was highly correlated with temperature. The temperature optimum shifted from 16°C for Pcap in the spring, to greater than 28°C in the summer. Multiple regression analysis indicated a second-order relationship of Pcap in the spring to temperature. Other independent variables explained only negligible variation of Pcap.

Notizen: SUMMARY. 〈list xml:id="l1" style="custom"〉1Use of a multi-column thin-layer pneumatic sampler and modified analytic procedures has enabled resolutions of chemical and biological strata at 2.5 or 5.0 cm depth intervals. Examination of meromictic Lake Fidler, Tasmania, indicates the presence of a thin, intense stratum of bacteriochlorohyll d in the upper monimolimnion, associated with a discrete stratum of Chlorobium cf. limicola at the microaerobic interface between the oxygenated and sulphide-rich zones.2Algae included small populations of Chlorophyceae, Chrysophyceae, Bacillariophyceae and Cryptophyceae. Bacteria included microaerophils and obligate anaerobes, pigmented and colourless, in well-defined strata in the upper monimolimnion. A population of the microcrustacean Calamoecia tasmanica tasmanica was present in the mixolimnion. Chaoborus larvae were concentrated within the Chlorobium layer.3The chemical profile of Lake Fidler was stable, with a chemocline constant in position relative to the lake bottom. The surface water levels rose and fell through a distance of 1m in conjunction with heavy rainfall in the rainforest, and with river level variation, but had no measurable effect on the absolute position of the chemocline. Marked heterogeneity of dissolved substances at depths in the vicinity of bacteria suggested endogenous influence on pH and gelbstoff (‘gilvin’ in Australia).4Downwelling light attenuation was influenced primarily by gelbstoff (‘gilvin’) in the mixolimnion, with only red light (peak at 700 nm) measurable below 2 m. Light was absorbed mainly by Chlorobium in the monimolimnion, and was unmeasurable deeper than 3 m.5The absorption spectrum of the bacteriochlorophyll d in vivo, with a maximum absorbance at 721 nm, corresponds with the available downwelling light penetrating the mixolimnion to the Chlorobium layer.

Beschreibung: The green mussel Perna viridis (Mytilidae) is a recent invader in the Atlantic basin including the Caribbean region and southeastern USA. In this study we examined the relative abundance of P. viridis across habitats in Tampa Bay, Florida, and density patterns at specific sample sites. Pilings, piers and floating objects were favourable habitats for P. viridis , while mangroves, flats and seawalls were not, although these were occasionally inhabited by P. viridis . Density and size-class distribution of P. viridis were estimated from approximately monthly samples collected between April 2002 and March 2003. Density of P. viridis varied significantly with sample date, reaching a maximum of 4,000 m –2 in September. Perna viridis exhibited the ability to dominate some habitats within the Tampa Bay ecosystem and the potential to compete for space with native species such as the eastern oyster, Crassostrea virginica , but the impacts were neither consistent nor widespread. Crassostrea virginica appears to have some refuges from P. viridis , while a small native mussel, Brachidontes cf. exustus , co-occurs with P. viridis in most sites; its density appears to be independent of P. viridis density.

Beschreibung: Homing endonucleases (HEs) can be used to induce targeted genome modification to reduce the fitness of pathogen vectors such as the malaria-transmitting Anopheles gambiae and to correct deleterious mutations in genetic diseases. We describe the creation of an extensive set of HE variants with novel DNA cleavage specificities using an integrated experimental and computational approach. Using computational modeling and an improved selection strategy, which optimizes specificity in addition to activity, we engineered an endonuclease to cleave in a gene associated with Anopheles sterility and another to cleave near a mutation that causes pyruvate kinase deficiency. In the course of this work we observed unanticipated context-dependence between bases which will need to be mechanistically understood for reprogramming of specificity to succeed more generally.
