    Notizen: The growth and reproduction complex contains recessive genes (grc) which influence body weight and gonadal development. Homozygous males are sterile, and they have an arrest of spermatogenesis at the primary spermatocyte stage. Homozygous females are fertile but have a reduced reproductive capacity. The data presented in this paper show that the latter defect is associated with a decrease in the relative number of secondary ovarian follicles and an increase in the number of atretic follicles. This finding indicates that most of the primary follicles do not mature properly. Thus, the genetic defect in gametogenesis controlled by the grc appears to occur at the same stage of development in both females and males.

    Notizen: The biodegradation of monochlorobenzene, the main contaminant in a quaternary aquifer at Bitterfeld, Central Germany, was studied in microcosm experiments employing either original groundwater or defined mineral media together with the indigenous microbial community from the polluted site. The impact of consecutive aerobic–anaerobic–aerobic incubations on monochlorobenzene biodegradation, microbial diversity, and pH development was examined. The related changes in microbial community composition were analyzed by 16S rRNA gene-based single-strand conformation polymorphism (SSCP) fingerprints and sequencing of dominant bands and by quantitative analysis of bacterial respiratory chain quinones as biomarkers. Under aerobic conditions, the indigenous microbial community of the groundwater degraded monochlorobenzene mainly via the modified ortho-pathway. Respiratory chain quinones and SSCP analysis suggested dominance of the genera Acidovorax and Pseudomonas. A shift to anoxic conditions resulted in monochlorobenzene biotransformation but no dechlorination. The ability to degrade monochlorobenzene aerobically remained preserved throughout a fortnightly anoxic period at sufficiently high buffer capacity. Acidification, caused by monochlorobenzene biodegradation, was alkalinity-controlled. At low initial alkalinity a substantial decrease in pH, monochlorobenzene degradation, and total counts of live cells, accompanied by a change of the microbial community composition, was observed.

    Notizen: SYNOPSIS A lipogenic toxin produced by the amoeba Hartmannella rhysodes, Fernald strain, made mammalian cells in culture round up and fill with fat droplets. From this toxin an enzyme was obtained with lecithinase and lysolecithinase activities. This enzyme is different from any isolated elsewhere. When purified enzyme was added to strain L mouse fibroblasts in culture, the cells rounded up and became fatty. Much of the lipogenic activity of the original toxin can be ascribed to this enzyme.

    Notizen: Recent improvements in instrumentation and field techniques have made time domain electromagnetic methods more acceptable. This acceptance has prompted further theoretical work for use in the interpretation of field data.The asymptotic solutions for the transient electromagnetic field components in the vicinity of a fault zone separating two media of high resistivity contrast are obtained for low frequency or late time. Excitation is by normally incident plane waves at the earth's surface. Because of the slow convergence of the asymptotic time series expansions, a numerical polygonal inversion is performed on the real part, or in-phase term, of the time-harmonic surface expressions for the electric and magnetic field components.For both impulsive and step excitation the transient electric field normal to the contact is more sensitive to changes in the structural attitude of the fault plane than the transient electric field parallel to the contact. The transient anomalous vertical magnetic field for either impulsive or step excitation appears to be most diagnostic of dip angle, although waveform shape does not seem to be significantly dependent on the slope of the fault. For dip angles greater than 90 degrees, as measured on the poorly conducting side of the contact, all field components become more insensitive as indicators of the structural attitude. The results presented here should be useful in obtaining several geologic parameters descriptive of a fault zone or lateral resistivity inhomogeneity from transient electromagnetic soundings; they should also provide an aid to differentiating with available geological information between layering effects on transient electromagnetic responses and effects largely due to lateral changes in resistivity.

    Notizen: The contribution of cytochrome and alternative pathways to respiration during soybean [Glycine max (L.) Merr. cv. Chippewa 64] seed formation and maturation was measured by oxygen consumption of embryonic axis and cotyledon tissues and of mitochondria isolated from immature seeds. Respiration rates (dry weight basis) declined duing the last month of seed maturation. Cyanide sensitive respiration (cytochrome pathway) accounted for 80 to 90% of total oxygen consumption throughout seed formation and maturation. The proportion of the total respiration resistant to 1 mM cyanide and inhibited by 1 mM salicylhydroxamic acid (alternative pathway) did not exceed 6%. A residual oxygen consumption, accounting for 5 to 15% of total respiration, persisted in the presence of both inhibitors. Likewise, virtually no alternative respiration was found in mitochondria prepared from immature seeds. The alternative respiratory pathway seems absent during soybean seed formation and maturation regardless of whether grown in the greenhouse or field.

    Notizen: Besides group A (GAS), Lancefield group C β-haemolytic streptococci (GCS) have been implicated as a causative agent in outbreaks of purulent pharyngitis. In this study we have investigated a class CI M protein of a Streptococcus dysgalactiae human wound isolate designated MC. MC shares similar properties with M proteins of GAS. It contributes to the virulence of the investigated GCS strain as revealed by in vivo phagocytosis in chicken embryos. Further, MC showed multiple binding to the human plasma proteins fibrinogen, albumin, plasminogen, IgA and all subclasses of IgG. Until now, an M protein, especially from a group C strain, with such a multiple binding behaviour has not been described. Immunoblot experiments with 150 patient sera, having a rheumatoid factor titre 〉1:256, revealed that 26% of these sera showed serological cross-reactivity between a 68-kDa cartilage protein and the N-terminal part of MC. Only 8% of the sera of healthy patients showed this property. In additional, MC also cross-reacted with antibodies recognising epidermal keratins. The cross-reacting 68-kDa protein from cartilage was different from human serum albumin, but was recognised with anti-vimentin immune serum. The MC was cloned and the gene sequenced. By using PCR, recombinant gene fragments encoding characteristic peptide fragments of MC were expressed in Escherichia coli. The peptides were used to map the binding sites for plasma proteins and to locate the cross-reacting epitopes on the MC molecule. In consequence, sequence alignments revealed that MC shared homologous regions with vimentin and different keratins. Our data, obtained with MC, suggest that not only infections with GAS but also infections with GCS and possibly GGS (the latter species can also produce class CI M-like proteins) may be responsible for the formation of streptococcal-associated sequel diseases.
