    Beschreibung: This book is comprised of important reviews and cutting-edge original research papers concerning electrospun and electrosprayed formulations in drug delivery. Electrospinning and electrospraying have, in recent years, attracted increasing attention in the pharmaceutical sector, with research in the area advancing rapidly. It is now possible to prepare extremely complex systems using multi-fluid processes, and to increase production rates to an industrial scale. Electrospun formulations can be produced under GMP conditions and are in clinical trials. In this volume, we explore a range of topics around electrospinning and electrospraying in controlled drug delivery. Four reviews cover the exciting potential of cyclodextrin-containing fibers and the many potential biomedical applications of electrospun fibers. The use of electrospinning to prepare amorphous systems and improve the dissolution rate and solubility of poorly soluble active ingredients is addressed, and the possibilities of such materials in tissue engineering are comprehensively covered. The six original research papers cover the effect of molecular properties on API release from Eudragit-based electrospun fibers; ferulic acid solid dispersions; electrospun medicines to treat psoriasis; scale up of electrospinning and its use to produce low-dose tablets; transepithelial permeation of drugs released from electrospun fibers, and the possibilities for the synergistic chemophotothermal treatment of cancer.

    Beschreibung: In recent years there has been an explosion of interest in the production of nanoscale fibres for drug delivery and tissue engineering. Nanofibres in Drug Delivery aims to outline to new researchers in the field the utility of nanofibres in drug delivery, and to explain to them how to prepare fibres in the laboratory. The book begins with a brief discussion of the main concepts in pharmaceutical science. The authors then introduce the key techniques that can be used for fibre production and explain briefly the theory behind them. They discuss the experimental implementation of fibre production, starting with the simplest possible set-up and then moving on to consider more complex arrangements. As they do so, they offer advice from their own experience of fibre production, and use examples from current literature to show how each particular type of fibre can be applied to drug delivery. They also consider how fibre production could be moved beyond the research laboratory into industry, discussing regulatory and scale-up aspects.

    Notizen: Abstract  CD20 is a hallmark antigen of B lymphocytes. Its expression is restricted to precursor and mature B cells but it is not expressed on plasma cells. The protein is a membrane-embedded phosphoprotein that appears likely to transverse the membrane four times. Its function is unknown although CD20 has been variously proposed to play a role in B-cell activation, proliferation, and calcium transport. A unique homologue of human CD20 has been described in mouse, which also shows a B-cell-specific pattern of expression. Here we describe the generating of mice carrying a CD20 gene disruption. So far, we have failed to detect any major effect of the gene disruption on the differentiation and function of B lymphocytes as judged by the expression of surface markers, antigen receptor signaling, proliferative responses, or calcium uptake. We did note, however, that the mice homozygous for the gene disruption [generated by intercrossing (129 × C57BL/6)F1 CD20 +/- heterozygotes] showed a substantial depletion of the sub-population of peritoneal B cells that lack expression of the B220 (RA3–6B2) isoform of CD45. The loss of the IgM+ 6B2- peritoneal B cells is not, however, attributable to the CD20 gene disruption itself. Rather, it segregates with a polymorphic difference between the 129 and C57BL/6 strains that is linked to the CD20 locus which, intriguingly, is itself close to the CD5 gene. This demonstrates that caution must be exercised when comparing the phenotypes of F2 litter-mates generated from crosses between 129 embryonic stem-cell-derived chimeras and mice of other strains.

    Notizen: Two methods were adopted of measuring sward output, following the prevention of bracken frond growth, with differing results. Accumulated growth estimation, corresponding to production under a system of open-range grazing, showed that asulam used for frond control had no effect on herbage yield in the year of application and large increases were observed in succeeding years compared with untreated control plots. In measuring production by sward regrowth after cutting, corresponding to intensive pasture utilization systems, asulam reduced yields in the year of treatment and increases were not observed until 2 years later. The inhibition of growth could be overcome by applying ground mineral phosphate at rates of 300 or 500 kg ha-1 before spraying but the 500 kg ha-1 rate showed no advantage over 300 kg ha-1. Applications of ground mineral phosphate after spraying had no effect on sward output.It is concluded that indigenous pasture plants in bracken-infested areas of western Scotland cease growth by mid July unless subjected to major earlier defoliation or severe soil moisture deficits and the removal of the frond canopy does not change this pattern significantly. Obtaining the full benefit of bracken clearance depends on making use of the resulting production of extra herbage and limiting as far as possible frond regrowth in treated areas.
