    Notizen: Broccoli is well recognized as a source of glucosinolates and their isothiocyanate breakdown products. Glucoraphanin is one of the most abundant glucosinolates present in broccoli and its cognate isothiocyanate is sulphoraphane, a potent inducer of mammalian detoxication (phase 2) enzyme activity and anti-cancer agent. This study was designed to measure: glucosinolate levels in broccoli florets from an array of genotypes grown in several environments; the elevation of a key phase 2 enzyme, quinone reductase, in mammalian cells exposed to floret extracts; and total broccoli head content. There were significant environmental and genotype-by-environment effects on levels of glucoraphanin and quinone reductase induction potential of broccoli heads; however, the effect of genotype was greater than that of environmental factors. The relative rankings among genotypes for glucoraphanin and quinone reductase induction potential changed, when expressed on a per head basis, rather than on a concentration basis. Correlations of trait means in one environment vs. means from a second were stronger for glucoraphanin and quinone reductase induction potential on a per head basis than on a fresh weight concentration basis. Results of this study indicate that development of a broccoli phenotype with a dense head and a high concentration of glucoraphanin to deliver maximum chemoprotective potential (high enzyme induction potential/glucoraphanin content) is a feasible goal.

    Notizen: Classical Mendelian experiments were conducted to determine the genetics and inheritance of quinclorac and acetolactate synthase (ALS)-inhibitor resistance in a biotype of Galium spurium. Plants were screened with the formulated product of either quinclorac or the ALS-inhibitor, thifensulfuron, at the field dose of 125 or 6 g active ingredient (a.i.) ha−1 respectively. Segregation in the F2 generation indicated that quinclorac resistance was a single, recessive nuclear trait, based on a 1 : 3 segregation ratio [resistant : susceptible (R : S)]. Resistance to ALS inhibitors was due to a single, dominant nuclear trait, segregating in the F2 generation in a 3 : 1 ratio (R : S). The genetic models were confirmed by herbicide screens of F1 and backcrosses between the F1 and the S parent. F2 plants that survived quinclorac treatment set seed and the resulting F3 progeny were screened with either herbicide. Quinclorac-treated F3 plants segregated in a 1 : 0 ratio (R : S), hence F2 progenitors were homozygous for quinclorac resistance. In contrast, F3 progeny segregated into three ratios: 1 : 0, 3 : 1 and 0 : 1 (R : S) in response to ALS-inhibitor treatment. This segregation pattern indicates that their F2 parents were either homozygous or heterozygous for ALS-inhibitor resistance. Therefore, there were clearly two distinct resistance mechanisms encoded by two genes that were not tightly linked as demonstrated by segregation patterns of the F3.

    Beschreibung: Tree growth rate is frequently used to estimate mortality probability. Yet, growth metrics can vary in form, and the justification for using one over another is rarely clear. We tested whether a growth index (GI) that scales the realized diameter growth rate against the potential diameter growth rate (PDGR) would give better estimates of mortality probability than other measures. We also tested whether PDGR, being a function of tree size, might better correlate with the baseline mortality probability than direct measurements of size such as diameter or basal area. Using a long-term dataset from the Sierra Nevada, California, U.S.A., as well as existing species-specific estimates of PDGR, we developed growth–mortality models for four common species. For three of the four species, models that included GI, PDGR, or a combination of GI and PDGR were substantially better than models without them. For the fourth species, the models including GI and PDGR performed roughly as well as a model that included only the diameter growth rate. Our results suggest that using PDGR can improve our ability to estimate tree survival probability. However, in the absence of PDGR estimates, the diameter growth rate was the best empirical predictor of mortality, in contrast to assumptions often made in the literature.

    Beschreibung: Tree mortality is often the result of both long-term and short-term stress. Growth rate, an indicator of long-term stress, is often used to estimate probability of death in unburned stands. In contrast, probability of death in burned stands is modeled as a function of short-term disturbance severity. We sought to narrow this conceptual gap by determining (i) whether growth rate, in addition to crown scorch, is a predictor of mortality in burned stands and (ii) whether a single, simple model could predict tree death in both burned and unburned stands. Observations of 2622 unburned and 688 burned Abies concolor (Gord. & Glend.) Lindl. (white fir) in the Sierra Nevada of California, U.S.A., indicated that growth rate was a significant predictor of mortality in the unburned stands, while both crown scorch and radial growth were significant predictors of mortality in the burned stands. Applying the burned stand model to unburned stands resulted in an overestimation of the unburned stand mortality rate. While failing to create a general model of tree death for A. concolor, our findings underscore the idea that similar processes may affect mortality in disturbed and undisturbed stands.

    Beschreibung: Field studies were conducted over a three-year period (2011, 2012, and 2013) in Louisiana to evaluate the effect of glufosinate rate and timing on glyphosate-resistant (GR) rhizomatous johnsongrass control in glufosinate-resistant soybean. Treatments included glufosinate (0.5, 0.6, or 0.7 kg ai ha−1) applied alone POST1 (46 cm tall johnsongrass) and sequentially 3 (POST2) or 4 (POST3) wk after POST1 at 0.5 or 0.6 kg ha−1. Glufosinate (0.7 kg ha−1) applied POST1 controlled johnsongrass 77% at soybean harvest. Averaged across sequential application rate, delaying the sequential application from POST2 to POST3 increased control from 65 to 78% at harvest. Increasing sequential application rate from 0.5 to 0.6 kg ha−1 reduced johnsongrass heights 15% at harvest. Furthermore, delaying the sequential application from POST2 to POST3 reduced GR rhizomatous johnsongrass heights to 63% of the nontreated at harvest. Soybean yields were maximized following the POST1 application of glufosinate at 0.7 kg ha−1 (2670 kg ha−1) and by applying 0.6 kg ha−1 of glufosinate sequentially (2620 kg ha−1), regardless of sequential application timing. Maximum control and soybean yield were observed following glufosinate POST1 at 0.7 kg ha−1 followed by 0.6 kg ha−1 at POST3. This data indicates that glufosinate is an option for management of GR rhizomatous johnsongrass.

    Beschreibung: In behavioral studies, cattle within the same pasture are not considered as independent experimental units because of the potential confounding effects of the herd’s social interactions. However, evaluating cattle behavior on extensive rangelands is logistically challenging for researchers, and treating individual animals as independent experimental units may be beneficial for answering specific research questions. The objective of this study was to evaluate the association patterns among global positioning system (GPS)-tracked cattle at six different study sites in the western United States. A Half-Weight Index (HWI) association value was calculated for each pair of GPS-tracked cows (i.e., dyad) to determine the proportion of time that cattle were within 75 m and 500 m of each other. Cattle at two study sites exhibited relatively low mean HWI-association values (i.e., less than 0.23 HWI); whereas, cattle at other study sites tended to have greater mean HWI associations (i.e., greater than 0.35 HWI). Distinguishing features between study sites with low and high association values were the management of cattle prior to the study, herd size, pasture size, and the number of watering points. However, at all ranches except one, at least 75% of all dyadic associations had HWI values of less than 0.5 at 500 m, indicating that most of the GPS-tracked cows were greater than 500 m from each other for over 50% of tracking period. While interactions among cattle in the same pasture are often inevitable, our data suggests that under some situations, movement patterns of a sub-set of individual GPS-tracked cows may have levels of independence that are sufficient for analysis as individual experimental units. Understanding the level of independence among GPS-tracked cattle may provide options for analysis of grazing behavior for individual cattle within the same pasture.

    Beschreibung: American eel are likely to encounter and pass through hydropower turbines, particularly during the downstream spawning migration, where exposure to stressors can potentially lead to injuries and mortality. Previous research has recovered dead eels downstream of hydropower facilities and, for some fish, injuries were easily attributed to blade strike; however, others showed no external signs of injury suggesting that other stressors, such as rapid decompression may be a potential source of mortality. For this research, yellow– and silver-phase American eel were held and allowed to acclimate to 172 kPa (absolute pressure) in hyper/hypobaric hydro-chambers for about 1 d. After acclimation, the state of buoyancy was determined prior to exposure to a rapid decompression simulating pressures encountered during hydroturbine passage. Fish were then examined for signs of barotrauma. Eel did not attain a state of neutral buoyancy but rather maintained negative buoyancy suggesting that eels, and possibly other benthic species, likely maintain a state of negative buoyancy to facilitate occupancy on or near the substrate. Additionally, eel were found to be resilient to rapid decompression, displaying no instantaneous mortality and minimal injuries, suggesting that barotrauma is not likely a major concern for American eel passing downstream through hydroturbines.
