    Notes: Summary Indoleamine 2,3-dioxygenase (IDO) is an interferon (IFN)-induced protein that initiates the metabolism of tryptophan along the kynurenine pathway. Although IDO can be induced by IFN-γ in many cell types, only mononuclear phagocytes have been shown to be induced to decyclize tryptophan by all three IFN classes. Since tryptophan is an essential amino acid necessary for a variety of metabolic processes, depletion of available tryptophan may be an important mechanism for control of rapidly-dividing microbial pathogens and tumors. The purpose of this review is to present evidence that documents the effects of IFN-induced IDO on prokaryotic and eukaryotic pathogens, as well as on a variety of tumor cell lines.

    Notes: Abstract Studies in vitro have shown that a respiratorydeficient phenotype is expressed by cells when the proportion of mtDNA with a disease-associated mutation exceeds a threshold level, but analysis of tissues from patients with mitochondrial encephalomyopathy, lactic acidosis, and strokelike episodes (MELAS) have failed to show a consistent relationship between the degree of heteroplasmy and biochemical expression of the defect. One possible explanation for this phenomenon is that there is variation of heteroplasmy between individual cells that is not adequately reflected by the mean heteroplasmy for a tissue. We have confirmed this by study of fibroblast clones from subjects heteroplasmic for the MELAS 3243 (A→ G) mtDNA mutation. Similar observations were made with fibroblast clones derived from two subjects heteroplasmic for the 11778 (G→A) mtDNA mutation of Leber's hereditary optic neuropathy. For the MELAS 3243 mutation, the distribution of mutant mtDNA between different cells was not randomly distributed about the mean, suggesting that selection against cells with high proportions of mutant mtDNA had occurred. To explore the way in which heteroplasmic mtDNA segregates in mitosis we followed the distribution of heteroplasmy between clones over approximately 15 generations. There was either no change or a decrease in the variance of intercellular heteroplasmy for the MELAS 3243 mutation, which is most consistent with segregation of heteroplasmic units of multiple mtDNA molecules in mitosis. After mitochondria from one of the MELAS 3243 fibroblast cultures were transferred to a mitochondrial DNA-free (ρ0) cell line derived from osteosarcoma cells by cytoplast fusion, the mean level and intercellular distribution of heteroplasmy was unchanged. We interpret this as evidence that somatic segregation (rather than nuclear background or cell differentiation state) is the primary determinant of the level of heteroplasmy.

    Notes: Summary The two types of scales of the coccolithophorid Hymenomonas (= Cricosphaera) carterae have been studied with the negative staining technique. Both types of scales, including the non-mineralized circular ones and the larger, mineralized elliptical ones are made up of a highly ordered network of radial and concentric fibrils. Both fibrils are ribbon shaped and have dimensions of 10–22×30–75 Å in cross-sections. The margin of the scales is composed of two closely associated fibrillar ribbons which are observed occasionally in edge view. In the elliptical scales, the periphery of the fibrillar network often is obscured by a deposition of the coccolith calcium carbonate. The substructure of the fibrils, the occurrence of kinking sites, and the fibrillar resistance to alkaline and weak acid treatments strongly resemble the scales of Pleurochrysis scherffelii. In relation to the recent findings of Brown and co-workers (Brown et al., 1969, 1970) on scale formation processes, it is hypothesized that the fibrillar network of the scales of both species is structurally and chemically identical, and that the fibrillar material consists of a cellulose-like polysaccharide.

    Notes: Abstract Selected yeasts classified as Candida sake van Uden et Buckley were examined for their physiological, morphological and immunological properties and their DNA relatedness. Candida maltosa Komagata, Nakase et Katsuya is herein recognized as a species separate from C. sake. Candida maltosa was distinguished from C. sake and from C. tropicalis by insignificant DNA reassociation. In addition, C. maltosa was distinguished from C. sake by its higher maximal growth temperature and lower guanine plus cytosine content of its DNA and from C. tropicalis by its failure to utilize soluble starch for growth and its resistance to cycloheximide. The species C. cloacae and C. subtropicalis are placed in synonymy with C. maltosa.

    Notes: Summary The deoxyribonucleic acids of five algicidal nonfruiting myxobacteria are reported to have base ratios ranging from 69–71 G+C mole % as determined by thermal denaturation temperatures; no unusual nucleic acid bases were detected. These organisms are described as amicrocystogenous, gliding, Gram-negative bacilli capable of degrading gelatin, casein, starch, cellulose, chitin, and alginate. All have been shown previously to be algicidal. Poly-β-hydroxybutyrate in each was indicated by its conversion to crotonic acid. Antibiotic sensitivity of the five was similar to that of known nonfruiting myxobacteria. The fine structure of one, Myxobacter 44, revealed a triple-layered cellular envelope, the middle layer of which is lysozyme sensitive. Ruthenium red-positive slime material adhered to the outer surface.

    Notes: Summary The marine blue green alga, Trichodesmium erythraeum, was studied with electron microscopy in an attempt to elucidate the structural basis for its rapid lysis when removed from its marine environment. In this connection, it was found that a thining of the electron-dense layer of the longitudinal wall at the site adjacent to transverse wall attachment was responsible for lysis. The underlying biochemical basis for this change has not been elucidated because of the extreme difficulties of maintaining and growing the alga in culture under defined conditions. Several other features of considerable interest also were found. Especially interesting is the very regular array of gas vacuoles in the form of a hollow cylinder which shields most of the photosynthetic system. It was suggested that the gas vacuoles might possibly function optically, having adaptive value in protecting the free-floating alga from excessive radiation. In addition, a detailed structure of the cylindrical bodies was presented, and its structure with the photosynthetic lamellae was compared. On the basis of sectoring to form fragments of double lamellar units from the cylindrical body which are identical in structure to the photosynthetic lamellae, it has been postulated that the cylindrical body may be the site of synthesis for the photosynthetic system in Trichodesmium erythraeum.
