    Notes: Summary In humans, the H (heavy) and L (light) chains of the iron-storage protein ferritin, are derived from multigene families. We have examined the chromosomal distribution of these H and L sequences by Southern analysis of hybrid cell DNA and by chrosomal in situ hybridization. Our results show that human ferritin H genes and related sequences are found on at least seven different chromosomes while L genes and related sequences are on at least three different chromosomes. Further, we have mapped the chromosomal location of expressed genes for human H and L ferritin chains and have found an H sequence which may be a useful marker for idiopathic hemochromatosis.

    Notes: Zusammenfassung Kurz zusammengefasst sind die wichtigsten, hier niedergelegten Tatsachen folgende: Es wurde eine Methode für die totale Entfernung der Leber bei Hunden ausgebildet, welche frei von den meisten Fehlerquellen ist, die früheren Exstirpationsmethoden des Organs bei Säugetieren anhaften. Es wurde gefunden, dass eine geeignete Einführung von Glukose bei derart operierten Hunden das Leben viele Stunden nach Verlust des gesamten Lebergewebes erhält. Von den vielen an hepatektomierten Tieren gemachten Beobachtungen möchten wir hervorheben, 1. dass die Leber zur Erhaltung des Blutzuckerspiegels absolut wesentlich ist; 2. dass die Leber der wichtigste Faktor, wenn nicht der einzige, bei dem Prozess der Desamidierung und der Harnstoffbildung ist, und 3. dass Bilirubin auch ohne Mitwirkung der Leber gebildet werden kann.

    Notes: Summary We have used a full length cDNA clone to determine the chromosomal location ofthegene encoding human ornithine aminotransferase (OAT), a mitochondrial matrix enzyme. Southern blot analysis of ScaI-digested DNA from 34 human-mouse somatic cell hybrids revealed 11 human fragments. Three fragments mapped to chromosome 10q23-10qter, confirming the previous provisional assignment of the functional gene to this autosome by analysis of OAT expression in somatic cell hybrids (O'Donnell et al. 1985). The remaining eight fragments were assigned to the X chromosome, and regionally assigned to Xp21-Xp11 by use of an X-chromosome mapping panel. These X chromosome sequences could represent pseudogenes, or related members of a multigene family. Two of the X chromosome fragments are alternate alleles of a restriction fragment length polymorphism (RFLP) making this OAT-related locus an excellent genetic marker. The RFLP may now be used to determine any possible relationship between this locus and several X-linked eye defects.

    Notes: Abstract Fluence-rate response curves for wavelengths from 640 nm to 730 nm were constructed for the day-extension promotion of flowering in green, light-grown, wheat (Triticum aestivum L., cv. Alexandria), a long-day plant. The resultant action spectrum had action maxima at 660 nm and 716 nm and resembles spectra for the high-irradiance reaction (HIR) seen in etiolated plants. Because, the HIR is thought to be controlled by type I pytochrome (that which is most abundant in etiolated tissue) our results indicate the involvement of type I phytochrome in the photomorphogenesis of a light-grown, green plant.

    Notes: Abstract Using two random DNA markers, and pulsed field gel electrophoresis, a 1.5-Mb physical map surrounding the 11p13 aniridia locus (AN2)has been assembled. The map was constructed using a combination of single- and double-restriction digests on DNA from normal controls and a patient transmitting familial aniridia. The aniridia patient has a chromosome translocation and the two DNA markers flank the breakpoint. This 11p13 breakpoint lies no further than 100 kb from the DNA marker 1104 (D11S95),located on the centromeric side of the breakpoint. Two CpG islands, separated by 550 kb and flanking the translocation, suggest an upper limit to the size of the gene.

    Notes: Abstract A cDNA clone encoding amino acids 809–1451 of the protease inhibitor α2-macroglobulin has been isolated from an adult human liver cDNA library. This cDNA was used to examine DNA samples prepared from a panel of human-mouse somatic cell hybrids with different numbers and combinations of human chromosomes for the presence of the human α2-macroglobulin gene. The cosegregation of this gene and chromosome 12 in the cell hybrid panel indicated that the α2-macroglobulin structural gene (designated A2M) is on human chromosome 12.

    Notes: Abstract In order to map human PGKsequences, DNA was prepared from 55 human-mouse somatic cell lines. The DNA was digested to completion with HindIII and Southern filters prepared. These filters were hybridized at high stringency conditions to a human PGKcDNA. Mouse and human X-linked and autosomal bands were distinguished and, in addition to known X-linked sequences, two autosomal PGKsequences were mapped: a 1-kb band to chromosome 19 and a 5-kb band to chromosome 6. The PGKcDNA probe was also hybridized to flow-sorted chromosomes confirming the presence of PGK sequences on the X chromosome and chromosomes 6 and 19.

    Notes: Synopsis The number of venomous caudal spines and their length and position relative to one another were determined in seven species of South American freshwater rays (Potamotrygonidae) and eight marine or euryhaline species of four families from the Caribbean Coast of South and Central America. Most species have two visible spines at certain stages in the shedding-replacement cycle and only one visible spine at other stages (following shedding). If we include the embryological beginnings of the spines before they erupt and become visible, the spine counts of most rays are actually 2 rather than 1 or 2. Since most species apparently follow this pattern, spine counts are of little use in distinguishing between species except in the relatively few that may have only one, or no spines. Eight captive Potamotrygon specimens maintained in simulated tropical temperature conditions over 12 months showed periodic shedding and replacement of spines. The molts were biannual for a given ray but annual for a given spine. They alternated between two spine loci and their cycles were approximately six months out of phase with each other. Recent studies on Dasyatis sabina by others report only one molt per year, with replacement spines forming always posterior to the primary spine rather than alternating between posterior and anterior. Supernumerary spines (counts of more than two, up to five) are also discussed, as are counts of one and zero.

    Notes: Summary Chalcone synthase (CHS) catalyzes the first and key regulatory step in the branch pathway of phenylpropanoid biosynthesis specific for synthesis of ubiquitous flavonoid pigments and UV protectants. In bean (Phaseolus vulgaris L.) and other members of the Leguminoseae, chalcone synthase is also involved in the synthesis of the isoflavonoid-derived phytoalexin antibiotics characteristic of this family. We have demonstrated that the haploid genome of bean contains a family of about six to eight CHS genes, some of which are tightly clustered. Treatment of bean cells with fungal elicitor activates several of these genes leading to the accumulation of at least five and probably as many as nine distinct CHS transcripts encoding a set of CHS isopolypeptides of Mr 42–43 kDa but with differing pI in the range pH 6–7. In elicited cells specific transcripts and encoded polypeptides are differentially induced with respect to both the extent and kinetics of accumulation. Wounding or infection of hypocotyl tissue also activates several CHS genes with marked differences in the pattern of accumulation of specific transcripts and encoded polypeptides in wounded compared to infected tissue or elicited cells, indicating operation of more than one cue for defense gene activation. Illumination induces accumulation of a different set of CHS transcripts including only one of the set hitherto demonstrated to be induced by biological stress. The organization and differential regulation of the CHS gene family in bean are discussed in relation to the functions of this enzyme in adaptative and protective responses to diverse enviromental stresses.

    Notes: Summary Primary kidney cultures from adult beige-J (bg J/ bg J) mice were selected for epithelial cell growth using D-valine medium. After 2 weeks of attachment and proliferation in vitro, the cells form a confluent or nearly confluent monolayer that retains several phenotypic characteristics of the beige-J mutant. These include large, multilamellar inclusion bodies that are apparently dysmorphic lysosomes, and higher concentrations of neutral glycosphingolipids and dolichols than control cells. β-Glucuronidase activity, used as a lysosomal enzyme marker, is not elevated in beige-J-cultured kidney cells compared with controls, as it is in the intact kidney. The high levels of β-glucuronidase activity in both control and mutant cells may mask expression of this difference in vitro. The action of the beige-J mutation in kidney cells is thought to be due to a block in exocytosis that results in the accumulation of abnormal lysosomes and their components. The maintenance of the beige phenotype in vitro indicates that the mutation is not suppressed in primary kidney cell cultures. The expression of the beige phenotype in vitro should be useful for studies concerning the primary lesion of this mutation.
