    Description: Over the twenty-first century, large changes in climate are projected for Antarctica and the Southern Ocean under scenarios of greenhouse gas increase and stratospheric ozone recovery.These changes would potentially have important environmental and societal implications, affecting, for example, sea level change, global ocean heat and carbon uptake, and ecosystem function. However, our ability to make precise estimates of these impacts is hampered by uncertainties in state-of-the-art climate models. Model evaluation is a key step in reducing this model uncertainty by helping to identify biases and shortcomings common to the current generation of models and highlighting priorities for future model development. The evaluation of climate models’ representation of Antarctic climate from the perspective of long-term twenty-first-century climate change was the subject of a workshop organized by Antarctic Climate 21 (AntClim21), one of six current scientific research programs of the Scientific Committee onAntarctic Research (SCAR; see www.scar.org/srp/antclim21).

    Notes: Deuterium nuclear magnetic resonance (NMR) spectra and spin–lattice relaxation times were determined in order to study the dynamics of t-butyl groups in butylated hydroxytoluene. The results are consistent with a model first proposed by Beckmann et al. [J. Magn. Reson. 36, 199 (1979)], where there is an inequivalence between the methyl groups within each t-butyl group. While two methyl groups reorient rapidly relative to the whole t-butyl rotation, the remaining methyl group is more restricted in its motion, reorienting at a rate comparable to that of the t-butyl group itself. The spin–lattice relaxation data show two T1 minima, the high temperature minimum (40 °C) corresponding to the combined t-butyl and "slow'' methyl rotations, and the low temperature minimum corresponding to "fast'' methyl group rotation. Using an explicitly defined T1 fitting function, the T1 data yield activation energies of 2.2 and 6.0 kcal/mol for the fast methyl and t-butyl rotations, respectively, both in agreement with Beckmann's values obtained from proton T1 experiments. It was also possible to simulate the low temperature deuterium NMR spectra from T=−160 °C to T=−80 °C using the aforementioned dynamical inequivalence between the t-butyl methyl groups. While the fast methyl group rotation was in the motional narrowing region for T〉−160 °C, it was possible, from the simulations, to determine the t-butyl exchange rates to within 10%. The jump rates are remarkably close to the values predicted from the T1 results. Above −80 °C, the spectra could not be simulated, implying that a third motion must be present to further alter the high temperature line shapes. The effective axial asymmetry of the T〉−20° spectra indicates that the additional motion involves a two site exchange.

    Notes: A chromosomal gene library of Leptospira interrogans serovar copenhageni strain Wijnberg was constructed in phage lambda gt11. Plaque immunoassay with RαP64 antiserum identified one clone expressing a putative groEL homologue. DNA sequence analysis of the 2.4kb EcoRI-BamHI cloned fragment from strain Wijnberg revealed two open reading frames encoding polypeptides of 10.5kDa (Hsp10) and 58kDa (Hsp58). Sequence comparison of the deduced amino acid sequences of these ORFs confirmed the operon as the groE equivalent of Leptospira. Transcriptional analysis suggested that this operon is primarily under the control of an Eσ70 promoter element. However, both Hsp10 and Hsp58 were overexpressed under heat-shock conditions as determined by [35S]-methionine pulse labelling experiments. As no functional heat-shock promoter could be identified, a 9bp inverted repeat, located between the transcription and translation start sites, may play a role in the upregulation of this operon under heat-shock conditions, similar to mechanisms described for several Gram-positive organisms.

    Notes: Abstract   A recombinant plasmid capable of restoring UV resistance to an Escherichia coli uvrA mutant was isolated from a genomic library of Neisseria gonorrhoeae. Sequence analysis revealed an open reading frame whose deduced amino acid sequence displayed significant similarity to those of the UvrA proteins of other bacterial species. A second open reading frame (ORF259) was identified upstream from, and in the opposite orientation to the gonococcal uvrA gene. Transcriptional fusions between portions of the gonococcal uvrA upstream region and a reporter gene were used to localise promoter activity in both E. coli and N.␣gonorrhoeae. The transcriptional starting points of uvrA and ORF259 were mapped in E. coli by primer extension analysis, and corresponding σ70 promoters were identified. The arrangement of the uvrA-ORF259 intergenic region is similar to that of the gonococcal recA-aroD intergenic region. Both contain inverted copies of the 10 bp neisserial DNA uptake sequence situated between divergently transcribed genes. However, there is no evidence that either the uptake sequence or the proximity of the promoters influences expression of these genes.

    Notes: Abstract The ability of 15 atmospheric general circulation models (AGCM) to simulate the tropical intraseasonal oscillation has been studied as part of the Atmospheric Model Intercomparison Project (AMIP). Time series of the daily upper tropospheric velocity poential and zonal wind, averaged over the equatorial belt, were provided from each AGCM simulation. These data were analyzed using a variety of techniques such as time filtering and space-time spectral analysis to identify eastward and westward moving waves. The results have been compared with an identical assessment of the European Centre for Medium-range Weather Forecasts (ECMWF) analyses for the period 1982–1991. The models display a wide range of skill in simulating the intraseasonal oscillation. Most models show evidence of an eastward propagating anomaly in the velocity potential field, although in some models there is a greater tendency for a standing oscillation, and in one or two the field is rather chaotic with no preferred direction of propagation. Where a model has a clear eastward propagating signal, typical periodicities seem quite reasonable although there is a tendency for the models to simulate shorter periods than in the ECMWF analyses, where it is near 50 days. The results of the space-time spectral analysis have shown that no model has captured the dominance of the intraseasonal oscillation found in the analyses. Several models have peaks at intraseasonal time scales, but nearly all have relatively more power at higher frequencies (〈 30 days) than the analyses. Most models underestimate the strength of the intraseasonal variability. The observed intraseasonal oscillation shows a marked seasonality in its occurrence with greatest activity during northern winter and spring. Most models failed to capture this seasonality. The interannual variability in the activity of the intraseasonal oscillation has also been assessed, although the AMIP decade is too short to provide any conclusive results. There is a suggestion that the observed oscillation was suppressed during the strong El Niño of 1982/83, and this relationship has also been reproduced by some models. The relationship between a model's intraseasonal activity, its seasonal cycle and characteristics of its basic climate has been examined. It is clear that those models with weak intraseasonal activity tend also to have a weak seasonal cycle. It is becoming increasingly evident that an accurate description of the basic climate may be a prerequisite for producing a realistic intraseasonal oscillation. In particular, models with the most realistic intraseasonal oscillations appear to have precipitation distributions which are better correlated with warm sea surface temperatures. These models predominantly employ convective parameterizations which are closed on buoyancy rather than moisture convergence.
