    Beschreibung: Muscle extracts of Pacific whiting, Merluccius productus, and arrowtooth flounder, Atheresthes stomias, were assayed for proteolytic activity using azocasein as a substrate. Pacific whiting extracts showed maximum activity at pH 5.0-5.2 and a temperature of 50°C, while arrowtooth flounder extracts had maximum activity at pH 5.5 and 55°C. Three sources of inhibitors (potatoes, egg white, beef plasma protein) were evaluated in vitro for inhibition of protease activity. All three were found to be effective inhibitors in crude muscle extracts. Further studies utilizing these inhibitors in surimi showed that potato was equivalent to both egg white and beef plasma protein in preserving the gel forming characteristics ofheated kamaboko in both species.

    Beschreibung: We hypothesize that the impact of PCB desorption from resuspended sediments depends upon the intensity of the resuspension (which scales to bottom stress in the absence of organisms), the rate at which each congener desorbs (which depends on the size and hydrophobicity of the chemical, the relative amount of 'labile' and 'resistant' forms, and the size distribution of the suspended particles), and the residence time of the particles in the water column (which depends on the time-variable water column turbulence regime and the particle settling velocities). In order to accurately quantify the impact of PCB desorption from Hudson River sediments, we are conducting experiments that realistically mimic bottom shear stress and water column turbulence and rapidly measure PCB congener release. The objectives of this study are to measure the kinetics of PCB congener desorption from Hudson River sediments under realistic bottom shear and water column turbulence conditions and to quantify the impact of shear stress and contaminant aging on PCB desorption kinetics.

    Notizen: Oligomers of (-)-epicatechin with 4β → 8 interflavan bonds, and as many as five monomer units, have been studied by conformational analysis and time-resolved fluorescence. The conformational analysis yields 2x-1 conformations for each oligomer with x monomer units. There are two conformations accessible at each interflavan bond. These conformations are denoted by + and -. The dominant conformations for the trimer and higher oligomers have an interaction between the phenolic hydroxyl groups on monomers i and i+2. This interaction involves the hydroxyl group at C(5) on monomer i, and either C(13) or C(8) of monomer i+2, depending on whether the conformation of the two intervening interflavan bonds is + + or + -, respectively. Minor contributions to the ensemble for the tetramer and pentamer are made by conformations that contain the sequence of successive interflavan bonds denoted by -+ or --. In -+ the interaction between monomer units i and i + 2 involves an aliphatic hydroxyl with a phenolic hydroxyl, and there are no hydroxyl-hydroxyl interactions between units iand i + 2 in --. The onset of a different decay law for the fluorescence when x increases from 3 to 4 may be associated with the appearance of the -+ and -- conformations as minor constituents in the ensemble.

    Notizen: Thermal and mechanical studies on many linear polyesters have revealed their behavior in crystalline, liquid crystalline, and amorphous phases. Their phase behavior in binary compositions has also been studied by a range of additional techniques and in combinations including the polycarbonate of bisphenol-A. Regions of amorphous compatibility and incompatibility have been identified and measures made of transition temperature changes with composition. The conditions for transreaction have also been determined and the properties measured for the resultant new copolymers.

    Notizen: Appearance of collagen fibrils in the cuticle was seen by electron microscopy to be preceded by fonnation of a finely filamentous matrix material. At first, the fine filaments of the matrix are unorganized. However, signs of orthogonal ordering soon appear in the most superficial portion of the cuticle, and subsequently appear more basally and closer to the underlying epidermis. Meanwhile, fibrils of different staining properties and identifiable as collagen begin to be deposited in the superficial portion of the cuticle, the same region which first showed organized fine filaments. Then, like the fine filaments before them, the collagen fibrils polymerize more basally. Collagen appears to polymerize on the preformed skeleton of fine filaments as though the fine filaments caused the collagen to assemble. Neither the polymerization nor ordering of collagen fibrils seems to require direct cellular intervention but occur first in that portion of the cuticle which is furthest away from the underlying epidermis. The fine filaments may be self ordering, extracellular macromolecules which in turn determine the polymerization of collagen fibrils.

    Notizen: The mature annelid cuticle contains orthogonally oriented collagen in a matrix capped superficially by a dense epicuticle with external corpuscles. The underlying epidermis is a simple columnar epithelium with two major cell types, mucous-secreting cells which secrete through channels in the cuticle to the exterior of the worm, and “supportive” cells which presumably produce and increase the cuticle by secreting into it.The structures of supportive cells, previously interpreted as specialized for establishing interfibrillar collagen order, are revealed by glutaraldehyde fixation as common cellular components without the qualities deemed useful to align collagen. Cell processes which penetrate and sometimes pass completely through the cuticle are not stable, not in geometric order, and lack cilia-like structure. Cilia, unlike the ubiquitous cellular processes, are highly restricted to regions of the epidermis with specialized functions. Cellular control, or other control, of collagen fibrillogenesis remains unestablished.
