    Notizen: Two separate exercises on a similar theme are reported. In the first, the moment-method program NEC was extensively modified in order to optimize its performance on a four-processor Cray X-MP computer. In the second exercise, the MININEC moment-method program was completely rewritten into the occam programming language so that it could be executed directly by a transputer system. The NEC exercise exploited judicious use of vectorization and alternative parallelization techniques and achieved a reduction of run times by factors of about five or more. The MININEC exercise has so far achieved only a modest reduction in run-time but it has the potential for a much greater improvement if an optimum strategy for configuring the transputer network is implemented.

    Notizen: This paper describes the proper external potential formulation of a cathodic protection system employed for corrosion prevention of structures in infinite electrolytes. The boundary condition associated with the mathematical formulation is not of the standard type encountered in hydrodynamic, electrostatic or heat transfer applications. The developed mathematical model is discretized by the boundary element technique, which is then solved iteratively by the Newton-Raphson method.

    Notizen: Relations form the theory of characteristics are used to connect boundary and interior points in the numerical analysis of multidimensional, unsteady gas dynamics.Since specification of all variables on the boundaries does not occur, it is necessary to employ at least some of the differential equations for boundary points. The objective is to avoid the use of non-central finite differences and the subsequent generation of a pseudo-boundary layer.Boundaries consisting of a moving shock wave, a solid wall and a surface in a supersonic, downstream region are considered. The numerical procedure for determining variables on boundaries is described in datail for the hypersonic, blunt body problem.

    Notizen: The technique of scanning force microscopy was used to study the nanometer-scale structure of NMP cast films of polyaniline. Noncontact mode images provide direct evidence that polyaniline prepared in this form is a granular conductor. The films were found to consist of micrograins whose size and density were determined by the pH of the acid solution used to protonate the films. At pH 7, the polyaniline films exhibited a mostly disordered structure, with small 2-10 nm particles visible. Protonation at pH 5 to pH 3 resulted in partial agglomeration of the primary particles into larger bundles, with sizes up to 75 nm. Treatment in solution pHs of 2 or less resulted in films consisting of close-packed bundles of dimension 20-30 nm. The conductivity of the films exhibited a sharp rise beginning with protonation at pH 2 or less. Effective medium theory (EMT), was used to model the macroscopic conductivity of these films based on the SPM measured microscopic film structure. Using the size and size distribution of polymer micrograins or bundles in a modified EMT, we are able to obtain predicted conductivities that are close to the measured values for these films. © 1995 John Wiley & Sons, Inc.

    Notizen: Many important layered silicate-polymer nanocomposite materials may be synthesized using an in-situ polymerization process. Using this technique, organic monomers are intercalated into the interlayer regions of the hosts, where subsequent polymerization may then occur. In this paper, we report on the in-situ polymerization of styrene in Cu(II)-exchanged hectorite thin films. Scanning force microscopy (SFM) images of the polymer surface reveal that the surface polystyrene is generally aggregated into groups of elongated strands. SFM imaging of the interclay regions, in conjunction with X-ray diffraction (XRD) and electron spin resonance (ESR) data, indicates that approximately 20-30% of these regions contain polystyrene, with minimal reduction in the majority of Cu2+ sites observed. XRD data shows little or no intercalation of the monomer into the true intergallery regions. Instead, the polymer likely forms in intercrystallite or planar defect regions. In addition, two distinct phases of polymeric material are found within these defect regions, a highly polymerized polystyrene in addition to a polystyrene form exhibiting greater material stiffness. © 1998 John Wiley & Sons, Inc.     Notizen: We have synthesized several potential inhibitors and/or modifiers of the carbohydrate portion of plasma membrane glycoconjugates. These include fluorinated and actylated analogs of D-glucosamine, D-galactosamine, and D-mannosamine. These compounds have been tested to determine their effects on both [14C] glucosamine and [3H] leucine incorporation into glycoconjugate and on cell growth and viability using P-288 murine lymphoma cells maintained in tissue culture. The most cytotoxic agent tested was 2-acetamido-2-deoxy-1,3,4,6-tetra-O-acetyl-β-D-glucopyranose or simply β-pentaacetylglucosamine which prevented cell growth at 10-4-10-3 M. β-Pentaacetylglucosamine cytotoxicity was correlated with its high lipid solubility, having an octanol/water partition coefficient of 0.424 as compared with 0.278 for the β-anomer and 0.017 for N-acetylglucosamine. In vitro metabolism studies with [14C]-and/or [3H]-labeled pentaacetylglucosamine have indicated intracellular de-O-acetylation leading to the biosynthesis of UDP-N-acetylglucosamine, followed by the incorporation of this sugar into cellular glycoprotein. Concomitant with the formation of increased amounts of this nucleotide sugar, intracellular UTP and CTP pools fell to one third normal within 3 h after the administration of 1 mM pentaacetylglucosamine. At present it is unclear whether the cytotoxicity of β-pentaacetylglucosamine or other similar agents is due to alterations in nucleotide and nucleotide-sugar pools causing a decrease in energy charge and polynucleotide biosynthesis or is due to a direct effect on membrane glycoconjugate biosynthesis.

    Notizen: Intact murine L1210 leukemic cells incorporated significant quantities of [3H]-N-acetylneuraminic acid directly from CMP-N-acetylneuraminic acid. When pretreated with Vibrio cholerae neuraminidase, incorporation increased sixfold to tenfold. Biochemical studies comparing incorporation of N-acetyl-neuraminic acid from the nucleotide sugar with that from free sugar demonstrated that the relatively high levels of incorporation from CMP-N-acetyl-neuraminic acid could not be due to the incorporation of free sugar generated by extracellular degradation of the nucleotide sugar. Very little N-acetylneuraminic acid was taken up or incorporated by L 1210 cells from free sugar and this incorporation was not increased by neuraminidase pretreatment. Moreover, extracellular breakdown of CMP-N-acetylneuraminic acid during incubations with L 1210 cells was rather insignificant.Electron microscope autoradiography of cells incubated with CMP-N-acetylneuraminic acid demonstrated that greater than 84% of the incorporated radioactivity was associated with the plasma membrane and less than 1% with the Golgi apparatus. These findings are consistent with the conclusion that incroporation of N-acetylneuraminic acid from CMP-N-acetylneuraminic acid is the consequence of a cell surface sialytransferase system. Pretreatment of cells with the nonpenetrating reagent, diazonium salt of sulfonilic acid, significantly inhibited this ectoenzyme system while only marginally affecting galactose uptake and incorporation at the Golgi apparatus. Interestingly, incorporation from CMP-N-acetylneuraminic acid declined as the viability of the cell population declined. When taken together, the above evidence develops a rigorous argument for the presence of a sialyltransferase enzyme system at the cell surface of L 1210 cells.Studies directed towards the detection of a similar ectogalactosyltransferase system were also undertaken. Cells incubated in the presence of UDP-[3H]-galactose incorporated radioactivity into a macromolecular fraction. The presence of excess unlabeled galactose in the incubation medium significantly reduced this incorporation. Electron microscope autoradiographs of cells incubated with UDP-[3H]-galactose, demonstrated that incorporation occurred primarily at the Golgi apparatus. The grain distribution in these autoradiographs was similar to that for free galactose. Thus, the incorporation observed for L-1210 cells incubated in UDP-[3H]-galactose was due primarily to the intracellular utilization of free galactose generated by extracellular degradation of the nucleotide sugar. Inability t o demonstrate an ectogalacto-syltransferase system on L1210 cells does not rule out the possibility that the enzyme is present but undetectable due t o the absence of appropriate cell surface acceptor molecules.
