    Notes: The combination of two-dimensional polyacrylamide gel electrophoresis (2-DPAGE), computer image analysis and several protein identification techniques allowed the Escherichia coli SWISS-2DPAGE database to be established. This is part of the ExPASy molecular biology server accessible through the WWW at the URL address http://www.expasy.ch/ch2d/ch2d-top.html. Here we report recent progress in the development of the E. coli SWISS-2DPAGE database. Proteins were separated with immobilized pH gradients in the first dimension and sodium dodecyl sulfate-polyacrylamide gel electrophoresis in the second dimension. To increase the resolution of the separation and thus the number of identified proteins, a variety of wide and narrow range immobilized pH gradients were used in the first dimension. Micropreparative gels were electroblotted onto polyvinylidene difluoride membranes and spots were visualized by amido black staining. Protein identification techniques such as amino acid composition analysis, gel comparison and microsequencing were used, as well as a recently described Edman “sequence tag” approach. Some of the above identification techniques were coupled with database searching tools. Currently 231 polypeptides are identified on the E. coli SWISS-2DPAGE map: 64 have been identified by N-terminal microsequencing, 39 by amino acid composition, and 82 by sequence tag. Of 153 proteins putatively identified by gel comparison, 65 have been confirmed. Many proteins have been identified using more than one technique. Faster progress in the E. coli proteome project will now be possible with advances in biochemical methodology and with the completion of the entire E. coli genome.

    Notes: Two-dimensional (2-D) maps of cytosol and enriched-membrane platelet proteins has allowed the identification of more than 25 spots by three different methods: matching of the platelet gels with other 2-D reference maps, immunoblotting with chemiluminescence detection, and N-terminal sequencing. Different G protein (guanosine triphosphate-binding protein) subunits, cytoskeletal proteins, and proteins common to the human liver, red blood cells and plasma were identified. The two platelet protein maps presented here contribute to the project of identification of human cell and body fluid proteins. They may serve as working tools since platelets are popular models for the study of central nervous system neurotransmitter systems and stimulus-response coupling mechanisms.

    Notes: We present solutions for the effective stress induced by gas flow through a porous solid into a borehole resulting from sudden pressure reduction. Tensile effective stress that exceeds the strength of the solid will lead to borehole failure. This has applications to the intentional creation of cavities, relevant to the efficient recovery of coalbed methane, and the avoidance of borehole stability problems in conventional gas production.

    Notes: SUPG methods were originally developed for the scalar advection-diffusion equation and the incompressible Navier-Stokes equations. In the last few years successful extensions have been made to symmetric advective-diffusive systems and, in particular, the compressible Euler and Navier-Stokes equations. New procedures have been introduced to improve resolution of discontinuities and thin layers. In this paper a brief overview is presented of recent progress in the development and understanding of SUPG methods.

    Notes: We examine the solution of a practical engineering problem on a parallel computer. The problem involves the steady laminar viscous flow about an ONERA M6 wing and the computer is a 64-processing-node Connection Machine CM-5E. We show that efficient domain decomposition procedures lead to a balanced load on the processors and low communication times. The net result is that solutions can be attained in roughly 20 min elapsed time for a 48,011-node, 266,566-element unstructured mesh. We conclude that this is sufficiently fast to support the design process.

    Notes: A new finite element procedure is introduced for the analysis of nearly-incompressible media. The approach may be simply implemented by a small change of the standard technique, and is applicable to arbitrary anisotropic and/or nonlinear media.     Notes: A fully discrete stability and accuracy analysis of some algorithms for the one-dimensional heat equation is presented.
