    Notizen: Abstract The role of a transmembrane Ca2+ gradient in anion transport by Band 3 of human resealed erythrocyte ghosts has been studied. The results show that a transmembrane Ca2+ gradient is essential for the conformation of erythrocyte Band 3 with higher anion transport activity. The dissipation of the transmembrane Ca2+ gradient by the ionophore A23187 inhibits the anion transport activity. The extent of this inhibition approaches 90% as the Ca2+ concentration on both sides of the ghost membrane is increased to 1.0 mM and half-maximum inhibition is observed at 0.25 mM Ca2+. Addition of ATP (0.4 mM) to the resealing medium can partly reestablish the transmembrane Ca2+ gradient by activation of Ca2+-ATPase and alleviate the inhibition to some extent. N-ethylmaleimide, an inhibitor of erythrocyte Ca2+-ATPase, prevents such restoration. Electron micrographs reveal that numerous larger intramembranous particles can be observed on the P-faces of freeze-fractured resealed ghosts in the absence of a transmembrane Ca2+ gradient.

    Notizen: A calculation method for the secondary ion mass spectrometry (SIMS) analysis of the reaction of a pure metal with gas mixtures of water and oxygen is introduced. Equation systems are to be solved, each expressed as: Y = A × X, where Y is an amplitude column measured from the SIMS spectrum, A is a matrix constructed from the relative abundances of isotopes of the metal and X is an unknown amplitude column of the relevant species MeaOHb±(a = 1, 2; b = 0, 1, 2 and 3). The applicability of the calculation method has been tested in SIMS analysis of ex situ-grown iron oxides with different and known 18O contents. In in situ oxidation, measurement of the ratio of H216O/H218O in the metal-water-oxygen reaction allows fractions of O in the species MeaOHb+ that originate from water to be calculated. Data from such calculations have been used to study the initial stage of the reaction of Fe and Cr in H216O/H218O/16O2 gas mixtures. The results are interpreted in terms of coupled/uncoupled reaction type, reactivity of metal to water relative to oxygen in the mixed gas and degree of dissociation of water.

    Notizen: Iron oxide films with known 18O and 16O content were prepared by thermal oxidation of iron metal in H218O/H216O gas mixtures at 400°C. The oxide films were analysed in four laboratories with SIMS within a month after they were produced. One oxide sample (with 91% 18O) was also analysed after storage in air for 15 months. It turns out that the oxides are stable over that time period. The yield ratios of the detected ionic species based on negative 18O and 16O ions and metal-oxygen-containing ions were compared with the nominal, known values. The effect of interference of residual 16O and 16OH on the results is discussed and appropriate measures for reducing this effect are proposed. Reasonably good agreement is found between the four laboratories.
