    Notes: In processing full-fat soy flour to an acid-precipitated lipid protein concentrate a byproduct whey fraction results which, because of its high biological oxygen demand, represents a serious disposal problem. Processing of food waste streams by reverse osmosis has received considerable attention because of its low theoretical energy requirement since no phase change is involved. In a previous study at this Center a mathematical model was developed for the diffusive transport of soy lipid protein concentrate whey across cellulose acetate membranes. In this study, pumping energy and power costs combined with membrane life and replacement costs were incorporated into the original model to provide a basis for optimization from an energy cost standpoint. Computer-simulated runs were compared with experimental pilot-plant runs, and the agreement between predicted and actual results was quite good. Water flux rates were in the range of 3 to 7 gal/ft2/day. Computer runs were used to optimize the processing of 100,000 gal/day of soy whey at 8000 ppm biological oxygen demand (BOD). Costs were at a minimum with a six-stage tapered flow primary reverse osmosis treatment over a porous cellulose acetate membrane, followed by a six-stage tapered flow reverse osmosis over a tighter membrane. BOD reduction was 94% at a cost of $5.45/1000 gal.

A series of statistically designed and analyzed experiments were conducted on a pilot-plant reverse osmosis unit to study the effect of the operating parameters on solute and solvent transport in cellulose acetate membranes. Sucrose and sodium chloride solutions were tested in addition to soybean whey to relate the mixed solute system in whey to that of single-solute organic and inorganic feed solutions. Water flux was shown to have an Arrhenius dependency on temperature, and some membrane compaction was observed with the more porous membrane. Concentration polarization for sucrose and sodium chloride solutions increased linearly with water flux. Solute flux for soybean whey solutions decreased with molarity and was independent of pressure, whereas solute rejection increased with temperature and pressure and was independent of molarity. Good agreement was obtained using the derived parameters A, B, and τ for soy whey in the diffusion transport model when compared to the observed experimental values.

    Notes: The kinetics of the acid hydrolysis of Streptococcus salivarius levan were studied by examining the weight-average molecular weight. The molecular weights were obtained in a continuous manner from light scattering. Two first order reactions were observed: an initial rapid reaction in competition with a slower reaction.Activation energies, activation entropies, as well as the rate dependence upon substrate and hydrogen ion concentrations were determined. The data seem to indicate that the fast reaction is related to the breaking of branch-point bonds while the slower reaction is related to the breaking of main-chain bonds. Thus, levan hydrolysis seems to be fundamentally different from the completely random degradation of other branched polysaccharides, i.e., dextran, glycogen, and amylopectin.

    Notes: Theoretical calculations predict that the differential melting curves for random polynucleotide sequences having lengths up to several tens of thousands of base pairs have a clear-cut fine structure. This structure appears in the form of multiple narrow peaks 0.3-0.4°C wide on the bell shaped main curve. The differential melting curves have different shapes for different specific sequences. The theory also predicts the disappearance of the fine structure when the length of the sequence increases and when circular, covalently closed DNA is considered instead of the open structure. The predictions of the theory were confirmed by the measurements of differential melting curves for open and covalently closed circular forms of DNA for PM2 phage (N = 104 base pairs) and also for other phage DNA's of different length: T7 (N = 3.8 × 104); SD (N = 9.2 × 104); T2 (N = 17 × 104). It was shown that the effect of fine structure results mainly from the cooperative melting out of DNA regions 300-500 base pairs long.

    Notes: We consider the problem of making allowance for superhelicity in the statistical-mechanical calculations of fluctuational violations of the DNA double helix. A simple model is discussed, making it possible in the calculations to use an approach based on the theory of helix-coil transition in DNA. The proposed algorithms allow calculating the effect of superhelicity on the base-pair fluctuational opening for any given sequence of nucleotides. An algorithm is also proposed allowing for the hairpin and cruciform structures in the palindromic regions of a sequence, as well as the open and helical states. The theory is used to calculate the melting curve for superhelical DNA at temperatures well below the melting point of the linear or nicked forms. The maps of opening probability are calculated for SV40 and φX174 DNA using their recently published complete nucleotide sequences. The data explain well the experimental results of probing the secondary structure of these DNA by single strand-specific endonucleases.

    Notes: Correlations of solvent solubility parameters with molar attraction constants and with properties like surface tension, dipole moment, and index of refraction have been explored. From relations found to be valid for solvents, it is possible to calculate the solubility parameters for polymers. A relation between the dispersion contribution to the surface energy of polymers (a measurable quantity) and the dispersion solubility parameter of polymers has been found which is similar to a relation established for low molecular weight substances.
