Synthetic Analogs of Corticotropine. Changes in the N-Terminal Amino Group of the Corticotropine-(1 - 23)-tricosipeptide-amideFour analogs 1c-f of the corticotropine-(1-23)-tricosipeptide 1b, which has a physiological activity of about 100 i. u./mg, are described. In compound 1c, 1-L-serin is replaced by 1-L-Serin-proline; as a result, the physiological activity is lowered by about one-half, while it rises fivefold when 1-L-serine is exchanged for β-alanine in 1d. Replacement of 1-L-methionine by the nonisosteric 1-L-leucine in 1e lowers the physiological effect compared with 1b. Surprisingly, in the corresponding 1-β-alanine compound 1f the simultaneous exchange of 1-L-serine for β-alanine does not bring about any further substantial increase of the effect. - The peptides of the sequence 1 - 10 are synthesized according to the new scheme 1+(3+6); the effect of various additions during the coupling reaction of these decapeptides with the tridecapeptide-amide 10 is discussed.

Vier Analoge 1c-f des Corticotropin-(1-23)-trikosipeptids (1b), das eine physiologische Aktivität von etwa 100 i. E./mg besitzt, werden beschrieben. In Verbindung 1c ist 1-L-Serin durch L-Prolin ersetzt; die physiologische Aktivität sinkt dadurch auf etwa die Hälfte ab, während sie beim Austausch von 1-L-Serin gegen β-Alanin in 1d auf das Fünffache ansteigt. Ersatz von 4-L-Methionin durch das nicht-isostere L-Leucin in 1e vermindert die physiologische Wirkung gegenüber 1b. In der entsprechenden 1-β-Alanin-Verbindung 1f erbringt der gleichzeitige Austausch von 1-L-Serin gegen β-Alanin überraschenderweise keine wesentliche Wirkungssteigerung. - Die Peptide der Sequenz 1 - 10 werden nach dem neuen Aufbauschema 1+(3+6) hergestellt; der Effekt verschiedener Zusätze bei der Kupplung dieser Decapeptide mit dem Tridecapeptid-amid 10 wird diskutiert.
