    Notes: Effects of palmitic, stearic, oleic, and linoleic acid on mitogen-induced DNA synthesis, on production of IL-1β, IL-2, IFN-gamma, and TNF-α, and on IL-2R expression were determined in human peripheral lymphocytes. Free fatty acids (FFA) were added over a wide range of concentrations to cells cultured under serum free conditions with fatty acid free albumin. DNA synthesis was stimulated by low and inhibited by high FFA concentrations. Physiologica concentrations were stimulatory, except for linoleic acid. Cytokine production became affected by all FFA tested. Palmitic acid enhanced the release of IFN-gamma at concentrations that diminished TNF-α production. Saturated fatty acids were significantly more potent than unsaturated fatty acids in affecting cytokine production. IFN-gamma secretion was significantly more stimulated or inhibited by the various FFA compared with the other cytokines. IL-2R expression correlated with the production of IL-2. When tested in combination, stimulatory as well as inhibitory effects of the individual FFA became attenuated. It is suggested that palmitic, stearic, oleic, and linoleic acid are physiological regulators of DNA synthesis and cytokine release in human peripheral lymphocytes. Modulation of FFA ratios may be an effective means for the fine tuning of the immune system. As secretory mechanisms of cytokines appear to exhibit substrate specificity for FFA, the release of individual cytokines may be selectively influenced by FFA. © 1994 Wiley-Liss, Inc.

    Notes: The nuclear DNA of eukaryotic organisms is associated with a variety of proteins, which together make up what is called “chromatin.” Chromatin serves to package all genes into higher-order structures such as nucleosomes, solenoids, and loop domains. Tight packing of a particular gene and its regulatory sequences does not allow the approach of RNA I or II polymerase proteins. Before or during the activation of such an inactive gene its chromatin has to adopt a relaxed, more “open” configuration. This altered chromatin can be probed by its higher sensitivity toward nucleases, such as DNAse I or S1 nuclease, and the appearance of DNAse I-hypersensitive sites. These sites may constitutively be present or may be induced, and they can be mapped to specific DNA sequence motifs. In many cases, such sites are delimited by non-B-DNA, notably Z-DNA, which in turn may form part of enhancer elements. The Z-DNA configuration may be induced or maintained by methylation of cytosyl residues within underlying sequences.Two plant gene model systems have been selected to probe their chromatin structure. Constitutively expressed T-DNA genes of Agrobacterium induced tobacco crown gall tumor cells have been shown to be organized in canonical nucleosomes, to be more sensitive to DNAse I than the bulk of host chromatin, and to contain a series of six constitutive DNAse I-hypersensitive sites. Inducible ribulose-1,5-bisphosphate carboxyl-ase/oxygenase small subunit (rbcS) genes of pea are rearranged into a nuclease-sensitive format upon activation by light, especially in their promoter region. The rbcS promoter harbors a series of five constitutive DNAse I-hypersensitive sites and one light-inducible site, which is surrounded by potential regulatory sequences (enhancer cores, inverted repeats). The 3′ region of rbcS genes also contains constitutive sites. Methylation/demeth-ylation of Alu I-, Fnu4H1-, HaeIII-, Sau3AI-, and Sau 96I sequences in rbcS promoters does not play any role in rbcS gene inactivation/activation.

    Notes: We have previously identified a 12 nucleotide long sequence element, the TCE, that was demonstrated to be necessary for translational control of expression in the male germ line of Drosophila melanogaster (Schäfer et al., 1990). It is conserved among all seven members of the Mst(3)CGP gene family, that encode structural proteins of the sperm tail. The TCE is invariably located in the 5′ untranslated region (UTR) at position + 28 relative to the transcription start site. In this paper we analyse the mode of action of this element. We show that protein binding occurs at the TCE after incubation with lestis protein extracts from Drosophila melanogaster. While several proteins are associated with the translational control element in the RNA, only one of these proteins directly crosslinks to the sequence element. The binding activity is exclusively observed with testis protein extracts but can be demonstrated with testis extracts from other Drosophila species as well, indicating that regulatory proteins involved in translational regulation in the male germ line are conserved. Although binding to the TCE can occur independent of its position relative to the transcription start site of the in vitro transcripts, its function in vivo is not exerted when shifted further downstream within the 5′ UTR of a fusion gene. In addition to being a translational control element the TCE also functions as a transcriptional regulator. Consequently, a DNA-protein complex is also formed at the TCE. In contrast to the RNA-protein complexes we find DNA-protein complexes with protein extracts of several tissues of Drosophila melanogaster. © 1993 Wiley-Liss, Inc.

    Notes: Rat brain was exposed to 591-MHz, continuous-wave (CW) microwaves at 13.8 or 5.0 mW/cm2 to determine the effect on nicotinamide adenine dinucleotide, reduced (NADH), adenosine triphosphate (ATP) and creatine phosphate (CP) levels. On initiation of the in vivo microwave exposures, fluorimetrically determined NADH rapidly increased to a maximum of 4.0%-12.5% above pre-exposure control levels at one-half minute, then decreased slowly to 2% above control at three minutes, finally increasing slowly to 5% above control level at five minutes. ATP and CP assays were performed on sham- and microwave-exposed brain at each exposure time. At 13.8 mW/cm2, brain CP level was decreased an average of 39.4%, 41.1%, 18.2%, 13.1%, and 36.4% of control at exposure points one-half, one, two three, and five minutes, respectively, and brain ATP concentration was decreased an average of 25.2%, 15.2%, 17.8%, 7.4%, and 11.2% of control at the corresponding exposure periods. ATP and CP levels of rat brain exposed to 591-MHz cw microwaves at 5 mW/cm2 for one-half and one minute were decreased significantly below control levels at these exposure times, but were not significantly different from the 13.8 mW/cm2 exposures. For all exposures, rectal temperature remained constant. Heat loss through the skull aperture caused brain temperature to decrease during the five-minute exposures. This decrease was the same in magnitude for experimental and control subjects. Changes in NADH, ATP, and CP levels during microwave exposure cannot be attributed to general tissue hyperthermia. The data support the hypothesis that microwave exposure inhibits mitochondrial electron transport chain function, which results in decreased ATP and CP levels in brain.
