    Notes: To ascertain if 17β-estradiol (E2)-induced proliferation could be attenuated by blocking the expression of endogenous transforming growth factor α (TGFα), estrogen receptor (ER)-positive, estrogen-responsive MCF-7 or ZR-75-1 cells and ER-negative, estrogen-nonresponsive MDA-MB-468 or HS-578T cells were infected with a recombinant amphotropic, replication-defective retroviral expression vector containing a 435 base pair (bp) Apa1-Eco R1 coding fragment of the human TGFα cDNA oriented in the 3′ to 5′ direction and under the transcriptional control of an internal heavy metal-inducible mouse metallothionein (MT-1) promoter and containing the neomycin (neo) resistance gene. E2-stimulated expression of endogenous TGFα mRNA was inhibited by 4-5-fold, and the production of TGFα protein was inhibited by 50-80% when M-1 mass-infected MCF-7 or MZ-1 mass-infected ZR-75-1 cells were treated with 0.75-1 μM CdCl2, whereas in comparably treated parental MCF-7 or ZR-75-1 cells there was no significant effect upon these parameters. E2-stimulated anchorage-dependent growth (ADG) and anchorage-independent growth (AIG) of the M-1 or MZ-1 cells was inhibited by 60-90% following CdCl2 treatment. In contrast, neither the ADG nor AIG of the parental noninfected MCF-7 or ZR-75-1 cells that were maintained in the absence or presence of E2 was affected by comparable concentrations of CdCl2. The ADG and AIG of TGFα antisense MD-1 mass-infected MDA-MB-468 cells that express high levels of endogenous TGFα mRNA were also inhibited by 1 μM CdCl2, whereas the ADG and AIG of MH-1 mass-infected HS-578T cells, a TGFα-negative cell line, were unaffected by CdCl2 treatment. These results suggest that TGFα may be one important autocrine intermediary in regulating estrogen-induced cell proliferation. © 1993 Wiley-Liss, Inc.

    Notes: The Microstructural Physics group at the Cavendish Laboratory is actively involved in a considerable number of research projects which cover a broad range of materials science. In this paper, we describe briefly several such projects, with particular emphasis given to the application of parallel-detection electron energy loss spectroscopy (PEELS) on a scanning transmission electron microscope (STEM) to the analysis of materials such as stainless steels, catalysts, and high temperature superconductors. In addition, we describe a number of related projects that are currently being carried out in the group, particularly those which utilise and develop novel STEM imaging and analytical techniques. © 1993 Wiley-Liss, Inc.

    Notes: Molecules in conditioned medium from stimulated lymphocyte populations or from certain cell lines are known to stimulate cells committed to various hemopoietic lineages as well as pluripotential cells to form colonies in culture. In this study, the relationship between molecules active on pluripotential cells and early cells committed to granulocyte, macrophage, megakaryocyte, or red cell production was explored using techniques of chemical separation. After separation on the basis of charge, or after sequential purification using methods of high resolving power based on hydrophobicity and size, these activities remained associated with one another. The observations provide support for a model which proposes that pluripotential hemopoietic precursor cells as well as their early committed progeny may all be responsive to a single lineage-indifferent factor. Responsiveness to “lineage-specific” factors such as erythropoietin is proposed to be a feature only of later cells after they have made the appropriate receptors as part of their differentiation program.

    Notes: Leukemia inhibitory factor (LIF) is a member of the cytokine family of growth factors. It has been shown to exert a variety of actions on a diverse range of cell types, including neuronal, bone, and hemopoietic cells (Hilton, 1992, Trends Biochem. Sci., 17:72-76). In many of these cell types, studies have indicated the presence of specific receptors for LIF (Godard et al., 1982, J. Biol. Chem., 267:3214-3222; Hilton et al., 1988, Proc. Natl. Acad. Sci. USA, 85:5971-5975; Hilton and Nicola, 1992, J. Biol. Chem., 267:10238-10247.). The mechanism by which these receptors act is believed to involve tyrosine phosphorylation and the signal transducing receptor component gp130. We have previously shown that LIF is capable of inducing both human and murine myoblasts to proliferate in culture (Austin et al., 1992, J. Neurol. Sci., 112:185-191). We now report that LIF binds specifically to receptors on the surface of myoblasts, with an equilibrium dissociation constant of 400 pM and the number of receptors per cell varies with cell density. Binding competition studies showed that LIF binding to these receptor sites was not competed for by a number of other growth factors which stimulate myoblast proliferation including basic fibroblast growth factor (bFGF), transforming growth factor-α (TGFα), insulin-like growth factor 1 (IGF-1), and interleukin-6 (IL-6). There was a time and concentration-dependent down-regulation of receptor numbers following preincubation of myoblasts with LIF. The processing of these receptors subsequent to binding, involves as a first step, internalization and degradation by the myoblast. LIF appeared to stimulate myoblast proliferation rather than cell survival. © 1995 Wiley-Liss, Inc.

    Notes: In one experiment, Sprague Dawley rats (16-21 days of gestation) and their offspring were exposed to 100-MHz (CW) electromagnetic radiation at 46 mW/cm2 (SAR 2.77 mW/g) for 4 h/day for 97 days. In another experiment, the pregnant rats were irradiated daily from 17 to 21 days of gestation with 2450-MHz (CW) microwaves at 10 mW/cm2 (SAR 2 mW/g) for 21 h/day. In a third experiment, 6-day-old rat pups were irradiated 7 h/day for five days with 2450-MHz radiation at 10 mW/cm2. Equal numbers of animals were sham irradiated in each group. Quantitative studies of Purkinje cells showed a significant and irreversible decrease in rats irradiated during fetal or fetal and early postnatal life. In animals exposed postnatally, and euthanized immediately after irradiation, significant decrease in the relative number of Purkinje cells was apparent. However, restoration apparently occurred after forty days of recovery.

    Notes: Changes of electrokinetic properties of cell nuclei and the quantity of granules of heterochromatin located near the nuclear envelope in nuclei of human buccal epithelium cells were studied under the influence of electromagnetic fields in vitro. Irradiation of cells was realized by means of a semiconductor generator of millimeter radiation (wavelength 7.1 mm, frequency 42.2 GHz), the Yav-1 apparatus for extremely high frequency therapy. It was shown that irradiation of cells induced a decrease in electric charge of native human buccal epithelium cell nuclei and an increase in chromatin condensation in nuclei.     Notes: The effects of super high frequency (SHF) microwaves (34-78 GHz) on rates of spontaneous firing of the slowly adapting, stretch-receptor neurons of crayfish were studied. Initially, irradiation of continuously perfused, fluid-cooled preparations at power densities to 250 mW/cm2 caused a transient decrease in the rate of spontaneous firing (the dynamic response). Subsequently, with extinction of the SHF field, the rate of firing increased, finally stabilizing at pre-exposure levels (stationary phase). Rates of firing also increased when the receptor muscle was stretched, and they were inversely correlated with small, field-induced increases of temperature (∼1.5°C). The response to SHF radiation did not depend on frequency if temperature of the medium was constant. No resonant peaks were found when the millimeter range of frequencies was scanned.

    Notes: A study was carried out in 1990 to guide the development of a protocol for assessing residential exposures of children to time-weighted-average (TWA) power-frequency magnetic fields. The principal goal of this dosimetry study was to determine whether area (i.e., spot and/or 24 h) measurements of power-frequency magnetic fields in the residences and in the schools and daycare centers of 29 children (4 months through 8 years of age) could be used to predict their measured personal 24-h exposures. TWA personal exposures, measured with AMEX-3D meters worn by subjects, were approximately log-normally distributed with both residential and nonresidential geometric means of 0.10 μT (1.0 mG). Between-subjects variability in residential personal exposure levels (geometric standard deviation of 2.4) was substantially greater than that observed for nonresidential personal exposure levels (1.4). The correlation between log-transformed residential and total personal exposure levels was 0.97. Time-weighted averages of the magnetic fields measured in children's bedrooms, family rooms, living rooms, and kitchens were highly correlated with residential personal exposure levels (r = 0.90). In general, magnetic field levels measured in schools and daycare centers attended by subjects were smaller and less variable than measured residential fields and were only weakly correlated with measured nonresidential personal exposures. The final measurement protocol, which will be used in a large US study examining the relationship between childhood leukemia and exposure to magnetic fields, contains the following elements: normal- and low-power spot magnetic field measurements in bedrooms occupied by subjects during the 5 years prior to the date of diagnosis for cases or the corresponding date for controls; spot measurements under normal and low power-usage conditions at the centers of the kitchen and the family room; 24-h magnetic-field recordings near subjects' beds; and wire coding using the Wertheimer-Leeper method. © 1994 Wiley-Liss, Inc.     Notes: Pregnant squirrel monkeys were exposed to 2450-MHz (CW) microwaves at an equivalent power density of 10 mW/cm2 (SAR 3.4 mW/g) for three hours daily in a cavity-cage module. The exposure began when pregnancy was determined by a hormonal method, and continued through the offspring's first 9.5 months. After irradiation, the brains of the offspring were fixed with formaldehyde, and the inferior vermis of each cerebella was removed and processed for histologic observations. Purkinje cell density in the uvula was determined in sagittal serial sections. There was no significant difference between control and experimental animals in the number of Purkinje cells per mm of Purkinje cell line (linear density), as well as in the density of Purkinje cells in the Purkinje cell layer.

    Notes: Identified cells of Helix lucorum L. received 20 min exposures to 23, 120, or 200 mT stationary magnetic field (MFs). Resting potentials and input resistances were measured. Controls were instituted for temperature changes and for mechanical and other sources of artifact. Resting potentials did not change with MF exposure. Input resistances decreased significantly in normally silent cells during MF exposure, but increased significantly in spontaneously active cells. The magnitudes of changes were monotonically related to strength of the MF. Changes in excitatory postsynaptic potentials were observed during MF exposure. Elimination of perineuronal glia by proteolytic enzymes eliminated the MF effects.
