    Notes: The β-thymosins are a family of small proteins originally isolated from the thymus. Recently, two of the major mammalian isoforms, thymosin β4 (Tβ4) and thymosin β10 (Tβ10), are identified as significant actin monomer sequestering proteins which may be involved in regulating actin filament assembly. To study the cellular function of β-thymosins, we have used isoform-specific antibodies to determine their concentration and intracellular distribution, and examined the effects of inducing overexpression of Tβ4 and Tβ10 on actin filament structures. Immunofluorescence labeling of peritoneal macrophages showed that both β-thymosins are uniformly distributed within the cytoplasm. cDNA-mediated overexpression of β-thymosins in CV1 fibroblasts induced extensive loss of phalloidinstained actin stress fibers. Stress fibers in the cell center were more susceptible than those at the periphery. There was a decrease in the number of focal adhesions, as evidenced by a decrease in discrete vinculin staining and an increase in diffuse vinculin fluorescence. The majority of the transfected cells had normal shape in spite of extensive loss of actin filaments. Occasionally, cells overexpressing β-thymosin were observed to divide. In these cells, β-thymosin was excluded from the midbody which contains an actin filament-rich contractile ring. Our results indicate that Tβ4 and Tβ10 are functionally very similar and both are effective regulators of a large subset of actin filaments in living cells. © 1994 Wiley-Liss, Inc.

    Notes: An analysis scheme is presented that separates the individual effects of complexing agents and surfactants on the deposition and stripping steps in trace metal determination by differential pulse anodic stripping voltammetry (DPASV) at a hanging mercury drop electrode. The method involves DPASV measurements of total and electroactive (ASV-labile) metal in pH 7.0 phosphate buffer before and after medium exchange, with and without acidification of the sample solution. Copper, lead, and cadmium were determined in the presence of Triton X-100, a nonionic detergent, and the complexing agents fulvic and humic acids. Both the complexing agents and the surfactant affected deposition more than stripping, although Triton X-100 had a significant effect on the stripping process. Triton X-100 also acted as a complexing agent towards copper. Application of the analysis scheme to pristine and polluted freshwater samples showed that the decrease in the electroactivity of copper and lead was principally caused by the effect of surfactants on deposition. This study underlined the limitations of high-frequency ASV techniques (e.g., DPASV) at a hanging mercury drop electrode for the determination of the toxic fraction of metals in water samples and raised questions about the usefulness of ASV copper complexing capacity titrations for water toxicity investigations.

    Notes: In C2C12 myoblasts, 12-O-tetradecanoylphorbol-13-acetate (TPA) stimulated a phospholipase D (PLD) to degrade phosphatidylcholine (PC) as measured by the release of choline and an increase in the formation of phosphatidic acid (PA) (or phosphatidylbutanol [PtdBuOH] in the presence of 0.5% butanol). Exogenous PLD also stimulated choline release, PA and PtdBuOH formation. The protein kinase C (PKC) inhibitor, Ro-31-8220, and PKC downregulation significantly inhibited the effects of TPA but Ro-31-8220 had no effect on PLD action. Neither basic Fibroblast Growth Factor (bFGF) or Epidermal Growth Factor (EGF) increased PLD activity. All agonists stimulated protein synthesis during both a 90 min and a 6 hr incubation and increased RNA accretion after 6 hr. The response at 90 min was not inhibited by the transcription inhibitor, actinomycin D. Ro-31-8220 and PKC downregulation significantly inhibited all the effects of TPA. In contrast, Ro-31-8220 significantly inhibited the increase in RNA accretion elicited by PLD but had no effect on the ability of agonists other than TPA to enhance protein synthesis. All agonists also stimulated thymidine incorporation into DNA. The effects of EGF, bFGF, and PLD were rapid and transient whereas that of TPA was delayed and sustained. Ro-31-8220 and PKC downregulation significantly inhibited the response due to TPA. Furthermore, Ro-31-8220 also significantly inhibited the effects elicited by EGF and PLD but not that induced by bFGF. In differentiated myotubes, TPA and PLD, but not bFGF or EGF, again stimulated choline release and PtdBuOH formation. However, all agents failed to stimulate protein synthesis and RNA accretion. The data demonstrate the presence in C2C12 myoblasts, but not differentiated myotubes, of both a PLD-dependent and PLD-independent pathway(s) leading to the stimulation of protein synthesis, RNA accretion, and DNA synthesis. © 1995 Wiley-Liss, Inc.

    Notes: The slow rates of polymerization and the low molecular weights of polymer obtained by the free radical-initiated polymerization of vinyl benzoate is noted. The kinetics have been investigated and indicate that addition of the growing chain to the aromatic nucleus to form a stable, nonpropagating radical is the probable reason. The chain length and extent of branching are consistent with the observed rates and also with the methyl affinities of similar compounds.

    Notes: This paper describes preliminary findings on the influence of 60-Hz (2-mT) magnetic fields on tumor promotion and co-promotion in the skins of mice. The effect of magnetic fields on natural killer (NK) cell activity in spleen and blood was also examined. Groups of 32 juvenile female mice were exposed to the magnetic field as described in part I. The dorsal skin of all animals was treated with a subthreshold dose of the carcinogen 7, 12-dimethylbenz(a)anthracene (DMBA). One week after the treatment, two groups were sham exposed (group A) or field exposed at 2 mT (group B) 6 h/day for 21 weeks, to test whether the field would act as a tumor promoter. No tumors developed in these two groups of mice. To test whether the magnetic field would modify tumor development by directly affecting tumor growth or by suppressing immune surveillance, two additional groups of mice were treated weekly with the tumor promoter 12-0-tetradecanoylphorbol-13-acetate (TPA) and then either sham exposed (group C) or field exposed (group D). The time to appearance of tumors was shorter (but not statistically so) in the group exposed to magnetic fields and TPA. Some differences in NK cell activity and spleen size were observed between the sham- and field-exposed groups.

    Notes: Linear viscoelastic properties are found to be a sensitive measure of flow-induced structural changes in a block copolymer. Styrene-butadiene-styrene block copolymer (SBS) with 26% polystyrene (PS) forms a macrostructure in the quiescent state with grains of the order of 1-10 μm. Within each grain, phase separation gives rise to a regular two-phase microstructure with cylindrical PS domains with radius of the order of 200 Å. Large-amplitude oscillatory shear (γ = 4.5) at temperatures between 139 and 181°C was applied to after the grain structure with the objectives of removing the discontinuities at the grain boundaries and of aligning the domains into a continuous ultrastructure. The SBS behaved like a solid (tan δ 〈 1 at low ω) before and like a liquid (tan δ 〉 1) after shear modification. This change expressed itself in the removal of the long relaxation times from the linear viscoelastic spectrum; the intermediate and low relaxation times were not affected by the shear modification. The viscoelastic spectrum slowly recovered during annealing with recovery times of the order of the longest relaxation time of the quiescent structure. Birefringence studies showed that the SBS did not recover into its original grain structure but into a highly oriented domain structure. The discontinuities at the grain boundaries could not be removed completely.
