    Notes: Cytological changes following transection of the proximal root of the trigeminal ganglion in adult rats were assessed by light and electron microscopy. Radices were transected about 3-5 mm from the ganglia and animals were killed from 1 to 60 days after the operation. Light microscopically, it was found that all Nissl granules became uniformly stained and evenly distributed throughout the cytoplasm within 3 days. Three types of cell alteration involving Nissl granules occurred within 3 to 12 days after the operation: (1) chromatolysis, (2) dark staining of the cytoplasm accompanied by an increase of Nissl granules, and (3) faint staining of the cytoplasm accompanied by dispersion of Nissl granules. Electron microscopically, the chromatolysis pattern was characterized by peripheral concentration of the granular endoplasmic reticulum (gER) and ribosomes in the cytoplasm. Neurons of the darkstaining type showed an increased number of polysomal complexes throughout the cytoplasm, whereas those of the faint-staining type had diffusely dispersed cisternae of the gER which were shortened and bore reduced numbers of attached ribosomes. Perinuclear localization of profiles of Golgi complexes disappeared temporarily 1-3 days after the operation, but the normal perinuclear pattern appeared to return after 1 week. Enzyme histochemistry of acid phosphatase activity revealed an increase in the number of very fine reaction products in the cytoplasm up to 14 days following the operation. Cells recovered the normal pattern of Nissl staining by 48 days. Myelin figures, which are rarely observed in normal ganglia, were still observed in dense lysosomal bodies after 30 days. Nuclear size in affected neurons steadily increased up to about 2 weeks postoperation but returned to normal by 48 days.

    Notes: The neurons of the trigeminal ganglia of the rat and chicken were characterized by means of light microscopic, electron microscopic, and histochemical methods. Light microscopy disclosed four types of neurons, based on the characteristics of Nissl granules: (1) large neurons with diffusely distributed and very fine granules, (2) neurons containing coarse and sparsely distributed Nissl granules, (3) neurons containing dense Nissl granules of varying size, and (4) small neurons with granules concentrated peripherally. Electron microscopy allowed further definition of these four types of neurons by the length and arrangement of flattened cisterns of granular endoplasmic reticulum (gER) and the number of neurofilaments. Type 1 cells were largest, with a mean nuclear area of 139.8 ± 28.3 μm2. Type 4 cells were smallest, with a mean nuclear area of 74.6 ± 20.9 μm2. The mean nuclear areas of type 2 and 3 cells were intermediate to those of the type 1 and 4 cells. Type 3 and 4 neurons lacked neurofilaments. Four forms of Golgi apparatus were found: (1) large bent grains forming a network throughout the soma, (2) dispersed fine granular deposits, (3) fine or small granules, and (4) coarse bent deposits arranged confluently in the perinuclear zone. In some rat neurons, the concentration of acid phosphatase reaction products suggested a high enzymatic activity, whereas the chicken ganglion cells showed no such concentration. These findings are discussed and compared with the classifications of previous studies.

    Notes: To explore possible roles of calmodulin in Ca2+-induced ciliary reversal, we tested the effects of calmodulin antagonists on Triton-extracted models of Paramecium. In the extracted models prepared by the method of Naitoh and Kaneko [Science 176:523-524, 1972], the Ca2+ -induced ciliary reversal was not inhibited by calmodulin antagonists, trifluoperazine (TFP), or 5-chloro-l-naphthalenesulphone amide (W-7). However, in the presence of adenosine 3′,5′-cyclic mono-phosphate (cAMP), whose concentration is below the one that alters the ciliary direction, TFP inhibited ciliary reversal and the models swam forward at 10-5 M Ca2+. When the washing medium in the preparation of the extracted models was replaced with one containing MgCl2, the extracted model showed sensitivity to calmodulin antagonists without addition of cAMP; at 10-5 M Ca2+, 40 μM TFP or 100 μM W-7 inhibited the ciliary reversal and the models swam forward. Such effect of antagonists was abolished by an inhibitor of cAMP-dependent protein kinase. On the other hand, addition of cAMP enhanced the inhibitory effect of antagonists. These results suggest that calmodulin antagonists act to increase the extent of cAMP-dependent protein phosphorylation that inhibits the Ca2+ -induced ciliary reversal.

    Notes: Complex lipid biomarkers, including phosphatidylcholines, cerebrosides and sulfatides, are shown to be desorbed intact from rat brain myelin and rat liver microsomes by liquid secondary ion mass spectrometry, by plasma desorption and by laser desorption. Different polar lipids are favored by the different desorption techniques and as negative or positive ions. These selectivities support current theories about ionization for the different techniques.

    Notes: Steroid hormones and peptide growth factors promote growth and development of normal mammary tissues and some types of breast cancer. Ovarian steroids may influence mammary growth directly or indirectly. The epidermal growth factor (EGF) family of proteins may also regulate mammary growth. These two pathways may function independently of each other or they may act in concert, with steroids inducing transcription of genes that encode growth factors or growth factor receptors. We used a feline mammary adenocarcinoma cell line (K12) to address whether there was an interrelation between progesterone (PGN) and EGF-associated growth pathways. K12 cells responded to EGF by a dose-dependent increase in proliferation. PGN or promegestone (R5020, a synthetic progestagen) alone did not stimulate K12 growth, but when EGF and PGN, or EGF and R5020 were combined, they were synergistic. This synergistic response was abrogated by the PGN receptor antagonist RU486 or by antibodies that blocked binding of EGF to its receptor. K12 cells expressed characteristic double-affinity EGF receptors, as well as p185 (a functionally and structurally related protein, product of the neu gene) on their surface. PGN receptors were also found on intact cells and in cleared cytosols. Stimulation of K12 cells by PGN or by R5020 induced a two- to threefold increase in the number of high-affinity surface EGF receptors after 24 h. Stimulation of these cells by PGN also affected the relative levels of phosphorylation of the EGF receptors and p185 within minutes, but not of other cellular phosphoproteins. Our results show that PGN enhances the EGF-induced growth of K12 cells and suggest that this effect may be mediated at least partly via an increase in the number or function of high-affinity EGF receptors.

    Notes: Membrana granulosa was isolated from healthy large antral follicles of prepubertal or cyclic gilts stimulated with PMSG or PMSG and hCG. Ultrastructural observations revealed that pieces of pig membrana granulosa were associated with the basement membrane. The cattle cumulus-enclosed oocytes (COC) were placed in the rolled pieces of the pig membrana granulosa (PMG). After 8 and 24 hr of coculture with PMG from prepubertal gilts, only 16% and 21% of oocytes underwent GVBD, respectively. PMG from PMSG-stimulated cyclic gilts blocked the resumption of meiosis in all COC. The inhibitory effect of heterologous granulosa cells was fully reversible. When COC were initially incubated for 2 and 4 hr, subsequent culture in PMG prevented GVBD in 100% and 36% of oocytes, respectively. This suggests that functional contact between COC and PMG was established during the first 2 hr of coculture. To follow metabolic cooperation between PMG and COC, PMG was prelabeled with 3H-uridine and cocultured with COC. Autoradiography on semithin sections revealed the intensive passage of 3H-uridine from PMG into the cumulus layer and an oocyte. COC placed in PMG after GVBD (8 and 12 hr of an initial incubation) did not extrude the first polar body. PMG isolated from cyclic gilts after PMSG and hCG stimulation also inhibited GVBD of COC. Since nearly all COC placed in PMG isolated 10 and 12 hr after hCG remained in the GV stage after 24 hr of coculture, the hCG stimulation did not substantially diminish the meiosis inhibiting activity of PMG. During coculture, cattle cumulus cells were closely associated with the basement membrane, but no gap junctions were formed among heterologous granulosa cells. These results suggest that an inhibitory factor secreted by pig granulosa cells is not species specific and it can act in vitro without the mediation of gap junctions. © 1993 Wiley-Liss, Inc.

    Notes: The metrial gland develops in the uterus of many rodent species during normal pregnancy. It is a maternally-derived tissue that contains stromal and vascular elements plus a population of large cells, striking in their light microscopic appearance due to the presence of numerous cytoplasmic granules. These cells, which have become known in mice and rats as granulated metrial gland (GMG) cells, are derived from bone marrow precursors and recent work suggests they are a subset of lymphocytes belonging to the natural killer (NK) cell lineage. The functions of GMG cells during normal gestation have not been clearly defined. In vitro, GMG cells have been shown to produce cytokines and their cytokine profile is altered upon addition of medium containing the T cell growth factor interleukin-2 (IL-2). GMG cell granules contain the cytolytic protein perforin but GMG cells have a very limited capacity to kill in vitro unless they have been stimulated by IL-2 or interferon-gamma. Histological study of GMG cells has suggested they preferentially associate with fetal trophoblast. Since trophoblast appears resistant to immune lysis, except by IL-2-activated effector lymphocytes, and because resorbing murine embryos become infiltrated by lytic cells of the NK cell lineage, it is important to establish whether GMG cells are activated by pregnancy-associated events to play a major lytic role in vivo. © 1993 Wiley-Liss, Inc.

    Notes: Bis(triphenylphosphine)iminium salts of a series of high-nuclear and anionic ruthenium carbonyl cluster complexes were characterized by use of the negative-ion fast atom bombardment mass spectrometry. With a m-nitrobenzyl alcohol matrix, all of the monoanionic cluster complexes gave relatively intense intact anion peaks [A]- and all of the dianionic cluster complexes examined gave protonated monoanion peaks [HA]-. In some cases weak adduct ions were observed, which arose from the reaction of the cluster anions with the matrix during the bombardment process. The predominant fragmentation process of intact ions was found to be sequential loss of CO ligands and no loss of methyl, allyl or iodo ligand was observed. However, dissociation of bromo ligand from [Ru6C(CO)16Br]- and fragmentation of acetyl groups in [Ru5C(CO)13I2(COMe)]- and 6C(CO)16(COMe)- were noted. Degradation of cluster metal frameworks of [HRu10C(CO)24]- and [Ru10C(CO)24]2- into [HRu6C(CO)16]- was observed. These fragmentation and degradation patterns are comparable to their reactions in solution.

    Notes: A cell line was established in vitro from a spontaneous myeloid leukemia of SL strain mice. This cell line was cytologically identified as myeloblast, and its normal differentiation appeared to be completely blocked in mass culture. When the line cells were seeded in soft agar with a conditioned medium from normal cells, either macrophages or neutrophil granulocytes appeared from a single clone. The rate of formation of colonies containing differentiated cells always increased with an increase in the concentration of conditioned medium. The conditioned medium from this line cell was not as effective as was that from normal cells in inducing differentiation.

    Notes: In a myeloid leukemia cell line, the inducibilities of the Fc receptor, phagocytosis and cell motility were compared. Thymidine analogues such as BUdR, BCdR and IUdR blocked the induction of phagocytosis and motility but not induction of the Fc receptor. This BUdR susceptibility in the induction of phagocytosis and motility was lost in a BUdR resistant line which was isolated for its growth capability in a high concentration of BUdR. Actinomycin D and puromycin brought about a marked decrease in the inducibility of phagocytosis but not in that of the Fc receptor.This led us to the following conclusion: There is a genetic control in the inducibility of phagocytosis and motility in this cell line, and the incorporation of BUdR into cellular DNA results in the DNA becoming unresponsive to a differentiation-stimulating factor. In contrast, gene activation does not seem to be necessary for induction of the Fc receptor.The order of induction of several differentiation markers was also discussed.
