    Notes: We have previously shown that neurite outgrowth from 6-day chick embryo dorsal root ganglia (DRG) in vitro was stimulated when nerve growth factor (NGF) and pulsed magnetic fields (PMF) are used in combination. 392 DRGs were studied in a field excited by a commercial PMF generator. We have now analyzed an additional 416 DRGs exposed to very similar PMF's produced by an arbitrary wave from generator and power amplifier. We reproduced our previous findings that combination of NGF and bursts of asymmetric, 220 μs-wide, 4.0 mT-peak pulses induced significantly (p〈0.05) greater outgrowth than NGF alone, that fields without NGF do not significantly alter outgrowth, and that, unlike NGF alone, 4.0 mT fields and NGF can induce asymmetric outgrowth. The asymmetry does not seem to have a preferred orientation with respect to the induced electric field. Analysis of the data for the entire 808 DRGs confirms these findings. Importantly, we find similar results for pulse bursts repeated at 15 or 25 Hz. © 1996 Wiley-Liss, Inc.

    Notes: The involvement of cAMP and calcium in the rise in alkaline phosphatase (AP) activity observed when confluent, serum-free primary cultures of neonatal mouse calvarial cells are treated with parathyroid hormone (PTH) has been studied. Synthetic bovine PTH [bPTH-(1-34)] increased cellular cAMP at concentrations (10-9 to 10-7 M) previously found to elevate AP activity. Other substances that increase cAMP in these cells (forskolin, prostaglandin E2, 8-bromoadenosine cAMP and 3-isobutyl-1-methylxanthine) also increased enzyme activity. By comparison, increasing the concentration of calcium in the culture medium from 1.8 to 3.8 or 5.8 mM lowered the magnitude of the maximal AP response. In addition, treatment of cultures with the divalent cation ionophore A23187 caused a significant decrease in AP activity. These results suggest that: (1) cAMP mediates the rise in the specific activity of AP in cultured neonatal mouse calvarial cells treated with bPTH-(1-34) and (2) the concentration of calcium in the environment significantly influences the responsivity of bone cells to the hormone.

    Notes: Using monoclonal antibodies, we have analysed the distribution of three recently described Qa antigenic determinants (Qa-m7, Qa-m8 and Qa-m9) on murine clonable hemopoietic progenitor cells and spleen colony-forming units (CFU-S). Cytotoxicity experiments showed that Qa-m7 was expressed on almost all the progenitor cells (colony-forming cells, CFC) of megakaryocytes (MEG-CFC), erythroid cells (E-CFC), B lymphocytes (BL-CFC), and mixed colonies (MIX-CFC) as well as day 13 CFU-S, and a major proportion of granulocyte-macrophage colony-forming cells (GM-CFC) and day 8 CFU-S. Experiments using four sources of granulocyte-macrophage colony-stimulating activity suggested differential expression of Qa-m7 on subpopulations of GM-CFC, those preferentially forming macrophage colonies having lowest Qa-m7 antigen density. Immune rosetting techniques demonstrated the selective expression of Qa-m8 on approximately 50% of MEG-CFC, MIX-CFC and day 13 CFU-S, a pattern similar to that of Qa-m2. In contrast, Qa-m9 was not detected on any of the primitive hemopoietic precursors assayed. The results demonstrate the complexity of the Qa antigenic system, and suggest a possible role for these antigens in hemopoietic differentiation.

    Notes: Three parameters involved in the production of new ribosomal RNA (rRNA) were measured in Tetrahymena thermophilia: (i) the rate of synthesis of the rRNA precursor, (ii) the rate of processing of the RNA precursor and rRNA intermediates and (iii) the efficiency of utilization of the rRNA precursor in producing mature ribosomal RNA. These parameters were measured in cells in exponential growth and in cells starved in a dilute salt solution. Growing cells synthesize rRNA 20 times faster and process rRNA precursors and intermediates 10 to 15 times more rapidly than do starved cells. Both utilize their rRNA precursors with an efficiency of one in converting them to mature rRNA.

    Notes: We have measured the turnover rate of ribosomal RNA in exponentially growing Tetrahymena thermophila cells, cells entering the plateau phase of growth, and nutrient-deprived (starved) cells. Ribosomal RNA is stable in cells in early log phase growth but it begins to turnover as the cells begin a deceleratory growth phase prior to entering a plateau state. Likewise, rRNA in cells transferred from early log phase growth to a starvation medium begins to be degraded immediately upon starvation. In both cases the degradation of rRNA exhibits biphasic kinetics. A rapid initial exponential degradation with a half time of nine and one-half hours lasting for six hours is followed by a slower exponential degradation with a half-life of 35 hours. When starved cells are transferred to fresh growth medium turnover of rRNA ceases. The evidence presented suggests that the alteration in degradation rate is a regulated process which is most likely independent of the cell cycle.
