    Description: Skeletal unloading in an animal hindlimb suspension model and microgravity experienced by astronauts or as a result of prolonged bed rest causes site-specific losses in bone mineral density of 1%-2% per month. This is accompanied by reductions in circulating levels of 1,25-(OH)(2)D(3), the active metabolite of vitamin D. 1,25-(OH)(2)D(3), the ligand for the vitamin D receptor (VDR), is important for calcium absorption and plays a role in differentiation of osteoblasts and osteoclasts. To examine the responses of cells to activators of the VDR in a simulated microgravity environment, we used slow-turning lateral vessels (STLVs) in a rotating cell culture system. We found that, similar to cells grown in microgravity, MG-63 cells grown in the STLVs produce less osteocalcin, alkaline phosphatase, and collagen Ialpha1 mRNA and are less responsive to 1,25-(OH)(2)D(3). In addition, expression of VDR was reduced. Moreover, growth in the STLV caused activation of the stress-activated protein kinase pathway (SAPK), a kinase that inhibits VDR activity. In contrast, the 1,25-(OH)(2)D(3) analog, EB1089, was able to compensate for some of the STLV-associated responses by reducing SAPK activity, elevating VDR levels, and increasing expression of osteocalcin and alkaline phosphatase. These studies suggest that, not only does simulated microgravity reduce differentiation of MG-63 cells, but the activity of the VDR, an important regulator of bone metabolism, is reduced. Use of potent, less calcemic analogs of 1,25-(OH)(2)D(3) may aid in overcoming this defect. Copyright 2002 Elsevier Science Inc.

    Description: Closely spaced (cm-scale) traverses through the K-T boundary at Stevns Klint (Denmark), Woodside Creek (New Zealand) and a new Southern Hemisphere site at Richards Bay (South Africa) were subjected to trace element and isotopic (C, O, Sr) investigation. Intercomparison between these data-sets, and correlation with the broad K-T database available in the literature, indicate that the chemistry of the boundary clays is not globally constant. Variations are more common than similarities, both of absolute concentrations, and interelement ratios. For example, the chondrite normalized platinum-group elements (PGE) patterns of Stevns Klint are not like those of Woodside Creek, with the Pt/Os ratios showing the biggest variation. These differences in PGE patterns are difficult to explain by secondary alteration of a layer that was originally chemically homogeneous, especially for elements of such dubious crustal mobility as Os and Ir. The data also show that enhanced PGE concentrations, with similar trends to those of the boundary layers, occur in the Cretaceous sediments below the actual boundary at Stevns Klint and all three the New Zealand localities. This confirms the observations of others that the geochemistry of the boundary layers apparently does not record a unique component. It is suggested that terrestrial processes, eg. an extended period of Late Cretaceous volcanism can offer a satisfactory explanation for the features of the K-T geochemical anomaly. Such models would probably be more consistent with the observed stepwise, or gradual, palaeontological changes across this boundary, than the instant catastrophe predicated by the impact theory.

    Description: A terrestrial upstream wave event which demonstrates multiple, ion cyclotron harmonic resonances between the interplanetary wave population and an observed proton beam is analyzed. The techniques and parameters employed in the data analysis are discussed, including the use of differential and band-pass filters. An upstream wave event demonstrating multiple harmonic waves is examined, and the instability analysis relevant to the ion beam observations thought to be responsible for that event is discussed. It is shown that an observed bi-Maxwellian ion beam is capable of generating right and left-hand polarized waves through ion cyclotron harmonic resonance.

    Description: It is shown that current or planned spacecraft measurements could contribute toward the input data required for studies of natural anthropogenic influences on the middle atmosphere and their effects on weather and climate. Emphasis is placed on the solar occultation sounding technique used in the SAGE I experiment on board the Atmospheric Explorer Mission B spacecraft. It is shown that the aerosol and ozone number densities cannot be recovered simultaneously without introducing some formula for the aerosol extinction, or assumptions on the form of the aerosol size distribution.

    Description: Mice with targeted mutations of CD18, the common beta2 subunit of CD11/CD18 integrins, have leukocytosis, impaired transendothelial neutrophil emigration, and reduced host defense to Streptococcus pneumoniae, a gram-positive extracellular bacterium. Previous studies using blocking monoclonal antibodies suggested roles for CD18 and CD11b in hepatic neutrophil recruitment and host innate response to Listeria monocytogenes, a gram-positive intracellular bacterium. We induced systemic listeriosis in CD18 knockout (CD18-ko) and wild-type (WT) mice by tail vein injection with Listeria. By 14 days postinjection (dpi), 8 of 10 WT mice died, compared with 2 of 10 CD18-ko mice (P 〈 0.01). Quantitative organ culture showed that numbers of Listeria organisms in livers and spleens were similar in both groups at 20 min postinfection. By 3, 5, and 7 dpi, however, numbers of Listeria organisms were significantly lower in livers and spleens of CD18-ko mice than in WT mice. Histopathology showed that following Listeria infection, CD18-ko mice had milder inflammatory and necrotizing lesions in both spleens and livers than did WT mice. Cytokine assays indicated that baseline interleukin-1beta and granulocyte colony-stimulating factor (G-CSF) levels were higher in CD18-ko mice than in WT mice and that CD18-ko splenocytes produced higher levels of interleukin-1beta and G-CSF than WT splenocytes under the same amount of Listeria stimulation. These findings show that CD18 is not an absolute requirement for antilisterial innate immunity or hepatic neutrophil recruitment. We propose that the absence of CD18 in the mice results in the priming of innate immunity, as evidenced by elevated cytokine expression, and neutrophilic leukocytosis, which augments antilisterial defense.
