    Beschreibung: Artemisinin resistance in Plasmodium falciparum threatens global efforts to control and eliminate malaria. Polymorphisms in the kelch domain-carrying protein K13 are associated with artemisinin resistance, but the underlying molecular mechanisms are unknown. We analyzed the in vivo transcriptomes of 1043 P. falciparum isolates from patients with acute malaria and found that artemisinin resistance is associated with increased expression of unfolded protein response (UPR) pathways involving the major PROSC and TRiC chaperone complexes. Artemisinin-resistant parasites also exhibit decelerated progression through the first part of the asexual intraerythrocytic development cycle. These findings suggest that artemisinin-resistant parasites remain in a state of decelerated development at the young ring stage, whereas their up-regulated UPR pathways mitigate protein damage caused by artemisinin.     Beschreibung: Charlotte, De Soto, and Hardee counties are east-southeast ofTampa in west-central peninsular Florida, figure 1. In order toplan the future water-resource development of the area, informationabout the water resources is needed. To meet this need, the WaterResources Division of the U.S. Geological Survey, in cooperationwith the Peace River Basin Board of the Southwest Florida WaterManagement District as part of the statewide cooperative programwith the Division of Geology, Florida Board of Conservation, begana continuing hydrologic data collection program in July, 1963, asan initial step in the investigation and evaluation of the groundwaterresources of Hardee and De Soto counties. A similar hydrologicdata program commenced in Charlotte County in July, 1964.Previous work in Hardee and De Soto counties included aone year reconnaissance by the Division of Water Resources andConservation, Florida Board of Conservation, which concluded inJune, 1963, and resulted in a hydrologic report (Woodard, 1964).As an outgrowth of the hydrologic data program, a Map Seriesreport portraying the chemical character of water in the Floridanaquifer in the southern Peace River basin was prepared in 1967(Kaufman and Dion).The data contained herein constitute the basis for the MapSeries report. Additional selected data, including records of wellsand chemical analyses,, on the ground-water resources of the threecounty area are also included and are published to make the dataavailable.(Document has 28 pages.)

    Notizen: Steviol (ent-13-hydroxykaur-16-en-19-oic acid), the aglycone of various plant-derived glycoside sweeteners consumed by human populations, is known to be mutagenic toward Salmonella typhimurium strain TM677 when metabolically activated using a 9000 × g supernatant fraction derived from the liver of Aroclor 1254-pretreated rats. Mass spectral analysis of this diterpenoid and some analogs revealed characteristic patterns reflecting differential stereochemistry at the C/D rings and variations in the nature of the substituents present. Such information has been used to help identify several in vitro metabolites of steviol in conditions known to produce a mutagenic response, when analyzed by human populations, is known to be mutagenic toward Salmonella typhimurium strain TM677 when metabolically be allylic oxidation and epoxidation. 15-Oxosteviol, a product of oxidation of the major steviol metabolite, 15α-hydroxysteviol, was found to be a direct-acting mutagen.

    Notizen: This article describes a method that assists in structure determination of cyclic sulfides mixtures by GC/MS. The method involves the use of a gas-phase hydrodesulfurization microreactor (AP-56 catalyst, reaction temperture 300-315°C, hydrogen as a carrier and reagent gas) located between the capillary column (OV-17) and the mass spectrometer. The presence of a microreactor in such a position does not essentially affect the chromatographic separation achieved by using the capillary column. Combined analysis of the mass spectra registered for the original sulfides and the hydrodesulfurization products permits the structure determination of each effluent to be made more reliably. This is illustrated by the investigation of a reaction mixture containing the structural and geometric isomers of methyl-substituted thiabicyclononanes.

    Notizen: A new, simple procedure to deactivate fused silica capillaries without hydrothermal treatment is proposed. Based on high temperature reactions of octamethylcyclotetrasiloxane (D4) in the presence of oxygen, this procedure results, upon coating with dimethylsilicone (SE-30), in columns which give almost symmetric peaks for as little as 150 pg of critical compounds, such as decylamine, thereby demonstrating very high inertness. A mechanism of the deactivation procedure, which, in contrast to previously developed procedures, is based on thermal reactions under aerobic conditions, is proposed.
