    Notes: Although widely used in experimental and industrial situations, genetically engineered plasmids containing the lac promoter from Escherichia coli are subject to catabolite repression when grown in glucose-containing media. Several methods of overcoming this problem have been investigated by studying the expression of the protein A gene from Staphylococcus aureus under the control of the Escherichia coli lac promoter. When glycerol is used as a sole carbon source, the plasmid is unstable and is rapidly lost from the culture. When the bacteria are grown in chemostats under glucose limitation, the plasmid is maintained, even at high dilution rates, and the expression of protein A is similar to that observed when glycerol was used. The balance between metabolic load and protein A expression seems to be maintained by reducing the gene dose to a tolerable level. Depending on the metabolic conditions prevailing in the culture, this is achieved, either by reducing the copy number of the plasmid or in extreme cases by removing the plasmid altogether.

    Description: In August of 1982, an internal NASA team was chartered by the Associate Administrators for the Office of Space Flight (OFS) and the Office of Aeronautics and Space Technology (OAST) to assess the status of the nation's liquid chemical space propulsion test facilities and their adequacy to support current, near-term, and long-range national program requirements. This paper describes the results of that assessment, including the government and industry test capabilities in exisitence at that time, facility deficiencies that existed relative to what was needed to meet the perceived future test requirements, an integrated facility implementation plan recommended by the team, and the state of the liquid rocket propulsion industry at the time the assessment was made.

    Description: Various highlights of NASA's recently held space transportation propulsion and avionics symposia and the participation in this symposia by industry, the university community, and government organizations are discussed. The potential contributions of the integrated control and health monitoring (ICHM) field are considered. The goals of advanced ICHM technologies are reliability, operability, performance, and affordability. It is proposed that conventional liquid rocket engine configurations, characterized as noninteracting, stand-alone, compactly packaged and usually gimballed sets of hardware, can profit through embracing advanced ICHM capabilities.
