    Notizen: Abstract Experiments on isolated frog neuromuscular junctions in voltage-clamped conditions demonstrated increases in the probability of spontaneous release of quanta of mediator, resulting in increases in the K+ ion concentration in the perfusion solution, and changes in the endplate miniature current amplitude induced by increased quantum generation were studied. Factors promoting the desensitization of the post-synaptic cholinergic membrane (inhibition of acetylcholinesterase and addition of proadifen) to levels greater than a certain (critical) frequency of endplate miniature current of the order of 50 spikes/sec were found to result in accumulation of activity and progressive reductions in the sensitivity of the post-synaptic membrane to the mediator acetylcholine.

    Notizen: Abstract The effect of desensitized cholinoceptors on the time course of end-plate currents was evaluated in frog skeletal muscle at a high (physiological) level of acetylcholine secretion in the presence of active acetylcholinesterase, with two-electrode recording of the membrane potential. When the number of cholinoceptors was small so that they did not appreciably affect the amplitude of end-plate currents or the parameters of onequantum responses (miniature currents), the decay of multiquantum currents was significantly accelerated. Moreover, the presence of cholinoceptors drastically reduced the ability of acetylcholinesterase inhibitors to prolong the decay of end-plate currents. It is suggested that desensitized cholinoceptors in a synapse with a physiological level of acetylcholine secretion and active acetylcholinesterase may bind free acetylcholine with high affinity and thus supplement the well-known physiological role of acetylcholinesterase in limiting the reactivation of postsynaptic membrane cholinoceptors

    Notizen: Abstract Bradykinin did not change the amplitude and duration of growth and decrease of individual currents of the terminal plate, but potentiated the depression of amplitudes of consecutive currents during stimulation of the motor nerve at a frequency of 10–60 Hz under conditions of two-electrode fixation of potential on a frog neuromuscular preparation. The origin of this effect is presynaptic, because it does not depend on the activity of acetylcholinesterase and is associated with a longer increase in the terminal plate currents which are synchronous with the amplitude depression. The deconvolution method revealed a slower kinetics of the mediator secretion under the effect of bradykinin. Bradykinin impairs the conduction of rhythmic pulse series through the synapse by augmenting the asynchronism of the transmitter quantum secretion from the motor nerve endings, which may underlie the development of muscular asthenia observed in inflammatory and allergic reactions.
