    Notes: Summary Dictyosome-like structures (DLS) of guinea pig spermatocytes, when prefixed in mixtures of glutaraldehyde and tannic acid, exhibited laminated figures with a repeating periodicity of about 4.5 nm in the spaces between DLS saccules or in association with the surfaces of the DLS saccules. These laminated figures were similar to those figures derived from saturated lipids in other tissues. Alternatively, spaces between saccules were collapsed leaving only thin, electron-dense material separating adjacent saccules. These changes were not observed when the DLS were prefixed in glutaraldehyde before exposure to tannic acid. The presence of laminated figures following fixation with tannic acid and osmium tetroxide suggests that saturated lipids are present in, or associated with, the intersaccular regions of the DLS. The distribution of laminated figures in other membrane structures was not affected by post fixation with tannic acid nor were laminated figures comparable to those of the DLS observed between cisternae of the Golgi apparatus. These results support previous conclusions that DLS are distinct from Golgi apparatus and are a unique component of the germ cell cytoplasm.

    Notes: A method is described for eliminating a form of contamination called embedding pepper that appears in, or on, thin sections following poststaining in lead citrate. The pepper is present most often in the matrixes of mitochondria, peroxisomes, and red blood cells. Formation of pepper can be prevented by treating the section with 0.5 N HCl or 1.0% ethylenediaminete-traacetic acid (EDTA) for 1-5 min before staining in uranyl acetate or lead citrate.

    Notes: Ease of cutting thin sections with glass knives is markedly improved if the embedding resin contains a surfactant such as lecithin. With lecithin, it is possible to cut 50-100 thin sections from the same place on the knife edge even after facing off the block with 1-2-μm-thick sections. Image quality is similar to that of the unmodified resin if the resin formula is optimized. If not, some chatter or a “mottled” appearance of the tissue image may be present. Lecithin does not significantly affect sectioning with a diamond knife or the appearance of the section in the microscope. The increased ease of sectioning with glass presumably will be translated to diamond knives in the form of an increased useful life of the cutting edge.

    Notes: Low-angle chromium metal shadowing was used to compare the surface roughness of ultrathin sections cut with diamond, sapphire, and glass knives. Surface roughness was less on diamond-cut and sapphire-cut sections than on glass-cut sections, and was greater over tissue than resin, and greatest over red blood cells and mitochondria. Surface roughness contributed to image density variation and thereby lowered contrast and informational content of the sectioned tissue.
