    Notes: Carbon dioxide, oxygen, Freon 114, and butane were stripped from deionized water, sodium chloride solutions, and freshly acidified seawater in a vacuum-spray chamber using three types of hydraulic nozzle. Purely physical desorption was found for each gas/liquid system.For both conical-spray and fan-jet nozzles, the bulk of the gas was shown to desorb from the thin liquid sheet issuing from the nozzle, that is, before droplet formation occurs. Behavior was explained semiquantitatively by a mathematical model of the diffusion in the sheet. This model was also shown to predict well the desorption from the laminar liquid sheets formed by jet impingement nozzles. For turbulent flow in jet impingement nozzles, the fit of the model was again semiquantitative.

    Notes: Data are presented for wall temperatures and heat transfer coefficients for solid-vapor mixtures of parahydrogen and nitrogen flowing in an electrically heated straight tube of length 40 times its diameter. These are interpreted by the application of flat plate, constant property boundary-layer theory to models in which the solid particle geometrical distribution takes on simple limiting forms. The observed enhancement of the heat transfer coefficient over that for gas alone traveling at the same velocity is qualitatively predicted as a function of a dimensionless heat flux, the Sterman parameter qw/ρvUλ.

    Notes: Experimental data on vacuum-spray stripping of sparingly soluble gases from aqueous solution using hydraulic nozzles were presented in part I of this communication. Mass transfer was shown to occur primarily from the thin liquid sheet issuing from the nozzle tip in semiquantitative accordance with an analytical model based on laminar flow behavior.Jet impingement hydraulic nozzles offer the advantages of simplicity and low operating cost in vacuum-spray stripping. An empirical model for the turbulent flow regime has been developed to facilitate incorporating jet impingement nozzles into a vacuum stripper design. The results of an economic analysis comparing other types of industrial vacuum strippers with the proposed design of a jet impingement vacuum stripper show the latter process to compare very favorably in terms of estimated equipment size and cost and in expected operating cost.

    Notes: Extension of the understanding of properties of films of metals produced on glass surfaces by vacuum evaporation has permitted the fabrication of film-resistance thermometers that with simple instrumentation accurately measure surface or average surface temperatures without altering the geography of that surface. Formerly unknown and unstabled related properties of such films have been classified and may be anticipated or eliminated by recommended experimental procedures. Films of several of the most chemically inert and refractory metals 300 to 3,000 Å. thick have been shown to attain accuracies as high as 0.01°C. for practical periods of time. Their use, which is described, is developing satisfactorily, and the technique and equipment for their preparation are relatively simple.

    Notes: Grouping of reactions around key metabolite branch points can facilitate the study of metabolic control of complex metabolic networks. This top-down Metabolic Control Analysis is exemplified through the introduction of group (flux, as well as concentration) control coefficients whose magnitudes provide a measure of the relative impact of each reaction group on the overall network flux, as well as on the overall network stability, following enzymatic amplification. In this article, we demonstrate the application of previously developed theory to the determination of group flux control coefficients. Experimental data for the changes in metabolic fluxes obtained in response to the introduction of six different environmental perturbations are used to determine the group flux control coefficients for three reaction groups formed around the phosphoenolpyruvate/pyruvate branch point. The consistency of the obtained group flux control coefficient estimates is systematically analyzed to ensure that all necessary conditions are satisfied. The magnitudes of the determined control coefficients suggest that the control of lysine production flux in Corynebacterium glutamicum cells at a growth base state resides within the lysine biosynthetic pathway that begins with the PEP/PYR carboxylation anaplorotic pathway. © 1998 John Wiley & Sons, Inc.     Notes: TB/C3 hybridoma cells were transected with either pEF-MClneopA or pEF bcl2-MClneopA vectors to produce a control cell line (TB/C3 pEF) and a cell line that overexpresses the “antiapoptotic” human bcl-2 protein (TB/C3 bcl2). Flow cytometry analysis of intracellular bcl-2 protein levels enabled near on-line monitoring of the stability of bcl-2 expression in the absence of drug selection. It was possible to maintain spontaneous selection of cells with the overexpression of bcl-2 protein during semicontinuous cultures at very low dilution rates, where cells were subjected to the selective conditions of nutrient limitation and high toxic metabolite concentrations. Interestingly, cells that overexpressed bcl-2 were adapted to suspension culture conditions significantly faster than control cells. Dual fluorescence staining with acridine orange and propidium iodide allowed for discrimination between viable, apoptotic, secondary necrotic, and necrotic cells, respectively. Compared with the usual trypan blue method of establishing culture viability, dual staining demonstrated that under stressful conditions a significant proportion of cells that excluded trypan blue were also undergoing cell death through apoptosis. In batch cultures the overexpression of bcl-2 more than doubled the membrane intact (MI) cell productive period (the integral of Ml cell density with respect to culture time) and increased the monoclonal antibody (mAb) production by approximately 40% when compared with the control cell line. The overexpression of bcl-2 protein also significantly extended the cell integrity and viability by the suppression of apoptosis in conditions of hypoxia, hyperoxia, glutamine deprivation, glucose deprivation, and serum limitation. The suppression of apoptosis in anaerobic conditions suggests that bcl-2 exerts its antiapoptotic activity by a mechanism that does not involve an oxidative reactive pathway. In conditions of excess thymidine, which suppressed cell proliferation, Ml cell density and specific mAb productivity were further enhanced by the overexpression of bcl-2, which suggests the possibility of accomplishing a controlled proliferation in immortalized cell lines without invoking cell death. Cell size and intracellular mAb were increased for TB/C3 bcl2 cells compared with TB/C3 pEF control cells when analyzed by flow cytometry. © 1997 John Wiley & Sons, Inc.     Notes: The transfection of murine hybridomas with the apoptosis suppressor gene bcl-2 has been reported to result in the extension of batch culture duration, leading to significant improvements in culture productivity. In the present study, the effect of deprivation, individually, of each amino acid found in culture medium was examined to characterize the chemical environment of the culture in terms of its propensity to induce apoptosis. When cells were deprived of each amino acid, individually for 48 h, the majority of cell deaths in each case occurred by apoptosis, with essential amino acids being clearly most effective. For nearly all the amino acids, the viability of the bcl-2 cell line cultures was greater than 70% after 48 h, representing a substantial improvement in viability over control cell line cultures. Time course studies revealed that the induction of death could be divided into two phases. Initially, following the deprivation of a single essential amino acid, there was a period of time during which all the control cell line cultures retained high viability. The duration of this phase varied from 15 h in the case of lysine deprivation, through to 40 h in the case methionine deprivation. In the second phase of deprivation, the cultures exhibited an abrupt and rapid collapse in viability. The time taken for the viability to fall to 50% was similar for each amino acid. In every case, the duration of both phases of the bcl-2 cultures was considerably extended. Specific utilization rates were increased during the control cultures relative to the bcl-2 cultures for both the growth phase (ranging between 2% and 57% higher than the bcl-2 cultures) and the death phase (ranging between 172% to 1900% higher than the bcl-2 culture). © 1998 John Wiley & Sons, Inc.     Notes: Experimental latices of synthetic cis-1,4-polyisoprene and films and cellular objects produced from them are described. The latices of the synthetic polymers with molecular weights of about 2,000,000 are stabilized by anionic surfactants and have weight median particle diameters of about 0.9 μ and log normal particle size distribution. Tensile properties and flow characteristics of unvulcanized latex films could be regulated by introduction of controlled, small, amounts of crosslinking into the latex. Procedures used for crosslink density determination on cis-polyisoprene latex at various stages of use are described. Vulcanized latex films having maximum tensile strengths (5500-6000 p.s.i. and 800-950% elongation) were obtained when the final crosslink density was in the range 1.0-1.6 × 10-4 moles of effective chains/cc. Use of the synthetic cis-polyisoprene latex in production of foam is described. Gelation with sodium silicofluoride at room temperature occurs within 2-10 minutes. Production of a controlled level of crosslinking in the latex prior to frothing and gelling was found to improve the uniformity of cell structure of foams and to increase their resistance to compression. Excessive crosslinking in the latex during maturation was shown to cause a loss of tensile strength in cured products presumably because the coalescence of latex particles during gelation is adversely affected. The properties of synthetic cis-1,4-polyisoprene latices and the physical properties of films and foams derived from them are shown in general to be very similar to those of natural rubber latex and its products.

    Notes: Conventional tangential flow filtration (TFF) has traditionally been limited to separation of solutes that differ by about ten-fold in size. Wide pore-size distributions, membrane fouling, and concentration polarization phenomena have commonly been cited as reasons for this limitation. The use of TFF in the biotechnology industry has therefore been restricted to cell-protein, virus-protein, and protein-buffer separations. A multi-disciplinary team with industrial and academic members was formed to overcome these limitations and enable protein-protein separations using High Performance TFF (HPTFF) systems. Pore-size distributions have been improved with the development of new membrane formulation and casting techniques. Membrane fouling has been controlled by operating in the transmembrane pressure-dependent regime of the filtrate flux curve and by carefully controlling fluid dynamic start-up conditions. Concentration polarization was exploited to enhance, rather than limit, the resolution of solutes. Concentration polarization has also been controlled by operating a co-current filtrate stream that maintains transmembrane pressure constant along the length of the TFF module. High yields and purification factors were obtained even with small differences in protein sieving. IgG-BSA and BSA monomer-oligomer mixtures have successfully been separated with these systems. HPTFF technology provides a competitive purification tool to complement chromatographic processing of proteins. © 1997 John Wiley & Sons, Inc. Biotechnol Bioeng 56: 71-82, 1997.
