Notes: 1′-Hydroxysafrole is a proximate carcinogenic metabolite of the naturally occurring hepatocarcinogen safrole. Comparison by high-performance liquid chromatography of the nucleoside adducts obtained from hepatic DNA of adult female mice treated with [2′,3′-3H]1′-hydroxysafrolc with those formed by reaction of deoxyribonucleo-sides with electrophilic derivatives of 1′-hydroxysafrole indicated that the four in vivo adducts studied were derived from an ester of 1′-hydroxysafrole. Three of the four adducts comigrated with products of the reaction of 1′-acetoxysafrole with deoxyguanosine, whereas the fourth adduct comigrated with the major reaction product of the ester with deoxyadenosine. Analysis of the three deoxyguanosine ad-ducts indicated that all three involve substitution on the 2-amino group of guanine. A sample of the major adduct prepared from deoxyguanylic acid has been charac-terized from its NMR spectrum as N2-(trans-isosafrol-3′-Y1)-deoxyguanosine, and the deoxyadenosine adduct has been similarly characterized as N6-(trans-isosafrol-3′-yl)-deoxyadenosine.Repair replication was measured in cultured human T98G cells exposed to 1′-acetoxysafrole using the combined 5-bromodcoxyuridine density label and radioiso-topic label metnod. At a concentration of 1 mM 1′-acetoxysafrole, the amount of repair synthesis approached maximum values only about 15% of those obtained af-ter saturating doses of ultraviolet light. Repair patch size distribution was found to be similar in cells treated with ultraviolet light or 1′-acetoxysafrole as determined by the density of repair-labeled DNA relative to that of parental DNA.

Notes: Evaluation of the number of anchorage-dependent cells growing on the surface of microbeads kept in suspension in a bioreactor is a tedious procedure fraught with many difficulties. Using the Coulter counter as a biomass probe, this article shows that the number of cells adhering to microbeads can be determined while the cells are still attached.

Notes: A limited number of measurements were made of the heat transfer coefficient from an electrically heated rod to water flowing in an annulus. Tests were performed at Reynolds numbers ranging from 5,000 to 22,000 (based on equivalent diameter), water temperatures of 70° and 125°F., and relatively high heat fluxes of 52,000 to 208,000 B.t.u./(hr.) (sq. ft.). The annulus dimensions were 0.625 in. I.D. and 0.840 in. O.D. The coefficients varied as the 0.8 power of the velocity; they were 20% higher than predicted by use of Colburn's equation for flow inside pipes with the equivalent diameter. Over the range of conditions studied, it was found that the thermal boundary layer was fully developed in 1 1/8 in. (Lh/De=5).

Notes: Heat transfer was measured for water flowing parallel to 5/8-in. rods in a triangular lattice (0.914-in. spacing). Velocity was varied from 5 to 20 ft./sec., temperature from 150° to 325°F., and heat flux from 50,000 to 200,000 B.t.u./(hr.)(sq. ft.). Reynolds numbers ranged from 70,000 to 700,000. Coefficients were 40% higher than predicted from the Colburn equation for flow in pipes by use of the equivalent diameter. The effect of heated length was studied. No variation in local coefficient around the rod periphery was found. Pressure drops were 65% higher than predicted by the Fanning equation (by use of De).

Notes: Instantaneous shadow photography is used to study the mixing of air flowing in a 2-in. square duct with a secondary stream of carbon dioxide entering sidewise from an orifice. Mixing is described in terms of four mixing parameters measured from the shadow photographs, penetration distance, bulk mixing distance, macromixing distance, and the quality of micromixing, as a functon of sidestream: mainstream mass velocity and volume flow ratios, main duct velocity, and the average orifice shroud velocity component parallel to the air flow in the main duct. The first three parameters are reported quantitatively as a function of the variables mentioned. The latter parameter is reported semiquantitatively by means of a scale of grading the micromixing which describes the condition of the schlieren at arbitrary duct lengths downstream from the entrance of the gases. Comparisons are made with previous results from studies of the secondary gas entering sidewise from a tube.Orifice discharge coefficients are correlated as a function of the orifice diameter, orifice Reynolds number, main duct velocity, and orifice shroud velocity. These are presented to aid in the description of the mixing and to be used in the design of similar mixing configurations.
