    Notes: Sequence information is obtained by in-source fragmentation of electrospray-generated peptide ions. The complete sequences of leucine enkephalin, angiotensin I, and renin substrate are determined from their fragment ions and consume only low picomole amounts of material. The method involves collision-induced fragmentation of singly and multiply charged molecular ions in the electrospray ion source and does not involve tandem mass spectrometric instrumentation. The charge on the fragment ions is determined by the spacing of the isotope peaks.

    Notes: The (+)- and (-)-enantiomers of 3-isopropyl 5-(4-methylphenethyl) 1,4-dihydro-2,6-dimethyl-4-(2-pyridyl)-3,5-pyridinedicarboxylate were synthesized using an efficient highly enantioselective (ee ≥ 96%) variant of the Hantzsch dihydropyridine synthesis. The key step in this procedure involved the asymmetric Michael addition of a metalated chiral aminocrotonate, derived from D-valine or L-valine, respectively, to the Knoevenagel acceptor (Z)-2-isopropoxycarbonyl-1-(2-pyridyl)-but-1-en-3-one. Both enantiomers exhibited a dual cardioselective partial calcium channel agonist (positive inotropic)/smooth muscle selective calcium channel antagonist effect. The relative in vitro smooth muscle calcium channel antagonist activities of the (-):(+) enantiomers was 26:1. In contrast, the (+)-enantiomer exhibited a greater in vitro positive inotropic effect on guinea pig left atrium where the contractile force was maximally increased by 14.8% at a concentration of 1.63 × 10-8 M. © 1994 Wiley-Liss, Inc.

    Notes: The miscibilities of ternary copolymer blends prepared from poly(styrene-stat-acrylonitrile), poly(styrene-stat-methyl methacrylate), and poly(methyl methacrylate-stat-acrylonitrile) were predicted by calculating the interaction parameter, χblend, for various blend combinations, from the corresponding binary segmental interaction parameters estimated from previous work. Binodal and spinodal curves were calculated using the Flory-Huggins theory and it was observed that the most accurate estimate of the boundary between miscible and immiscible blends was given by the spinodal. It has also been demonstrated that in some of the ternary blends with fixed copolymer compositions the miscibility of the blend can be altered by changing the ratio of the three components in the mixture. Conditions for miscibility in this ternary system, and possibly a general feature of all such systems, are (a) that at least two of the binary interaction parameters χij are less than the critical value χcrit, while the third should not be too much larger, that is, one of the copolymers may act as a compatibilizer for the other two copolymers, (b) that the difference Δχ = /χ12 - χ13/ is small. © 1992 John Wiley & Sons, Inc.

    Notes: We have used transmission (TEM) and scanning electron microscopy (SEM) and leakage of lactate dehydrogenase (LDH; EC 1.1.1.27) to evaluate two published procedures which use filipin to render isolated rat hepatocytes permeable to ionic substrates. Cells treated by the procedure of Jorgenson and Nordlie6 retained less than 10 per cent of their LDH. TEM revealed severe damage to the internal structure of these cells, which included swelling, disintegration and extensive vesicularization of the endoplasmic reticulum (ER). Hepatocytes treated with filipin by the procedure of Gankema et al.13 retained 65-75 per cent of their LDH and displayed incomplete but highly variable permeability to Trypan blue. SEM revealed the loss of microvilli, other signs of swelling, and the presence of large lesions in the plasma membrane. TEM revealed signs of cell swelling, but the nuclei and the mitochondria were only moderately altered. The rough ER was not swollen, but significant fragmentation was evident and characteristic stacks of lamellar ER were never seen.We conclude that useful information about the functions of the ER in situ cannot be obtained from studies of filipin-treated cell. Our results indicate that retention of LDH is not a sufficient criterion of preservation of cell morphology and that staining with Trypan blue may significantly underestimate the permeability of cells to small ionic metabolites.

    Notes: An improved internal reflection infrared spectroscopy (ATR) technique (1) has been found to be effective for measuring the relative concentrations of polyester and polystyrene (resin) and calcium carbonate filler on sheet molding compound (SMC) surfaces. The technique has been used to determine the effect of molding conditions on the surface compositions of three commercial SMC materials. The surface compositions of two of the materials, of the same formulation but obtained from different sources, were the same and were unaffected by molding conditions. The surface of the third material (or a different formulation) was found to have substantially less resin than the first two materials. The surface composition of the third material varied with molding conditions, the greatest uniformity being obtained with high molding temperatures and pressures. This study has shown that the ATR technique is suitable for determining the relative surface compositions of SMC formulations. This method will be used to correlate the SMC surface composition to SMC properties, such as surface appearance, paintability, and adhesive bond durability.

    Notes: CI mass spectra of the five isomeric vicinal d2-decanes have been recorded using methane and d4-methane as reagent gases. In contrast to earlier suggestions, we find that a large fraction of the alkyl fragment ions from n-decane are formed by elimination of olefins from the abundant [M - 1] ion. Only the C9 and C8 fragment ions are produced completely by a one-step reaction between the decanes and the methane reagent ions. Isotope exchange does not occur between the hydrocarbon and the reagent ions derived from d4-methane but extensive scrambling of the deuterium label in the d2-decanes does take place in the [M - 1] ion.
