    Description: Four chemical extenders in 7 different concentrations (potassium chloride, sodium chloride, glucose, sodium citrate, Ringer s solution, cow serum and milkfish (Chanos chanos) serum) were compared in the preservation of milkfish sperm. Results showed milkfish serum to be the most suitable of the various extenders tested. This may be attributed to suitable osmotic potential and/or presence of proteins which may have directly or indirectly influenced sperm viability. The effects of milkfish serum on the motility and fertilizing capacity of sperm at different durations of storage however need to be investigated.

    Notes: Polystyrene-supported aluminium silver chloride was prepared as a solid and selective adsorbent for ethylene by refluxing silver chloride, aluminium chloride, and macroreticular-type polystyrene resin in carbon disulfide, followed by removal of the solvent in vacuo. The adsorbent rapidly adsorbed ethylene about equimolar to aluminium silver chloride under 1 atm at 20°C. Almost all the ethylene adsorbed was released by subjecting the adsorbent to a reduced pressure 8 mm Hg at 20°C for 10 min. The adsorbent, however, showed no adsorption of carbon monoxide under 1 atm at 20°C.

    Notes: A procedure for enzyme purification by means of line immunoelectrophoresis compatible with subsequent sodium dodecyl sulfate-polyacrylamide gel electrophoresis of the purified enzyme and Coomassie Blue staining is described. Electrophoresis was carried out in an agarose gel backed to GelBond film. The enzyme activity was detected after immunoelectrophoresis on the wet gel and the immunoprecipitate by protein staining on the dried gel. The strip bearing the maximum amount of precipitate was excised and the dried agarose gel scratched off the GelBond support. The dried gel was extracted with the Laemmli sample buffer, heated at 100 °C for 10 min and submitted to sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Proteins were stained with Coomassie Blue. Difficulties due to the presence of the immunoglobulin G subunits were encountered. The validity of the procedure was checked with an α-amylase from barley purified by immunoaffinity chromatography. The procedure was used to compare the molecular weight of α-amylase isozymes identified in barley and in rice seeds. In barley, α-amylase I, the minor enzyme group appearing during germination, had a slightly higher molecular weight than α-amylase II, the main enzyme group. In germinating rice seeds, the minor enzyme group, antigen D, was found to have a molecular weight slightly lower than the one of the other rice α-amylases. A new α-amylase antigen identified in rice as the constituent with the lowest pI was called pre-A. Its molecular weight was found to be the same as the one of the main enzyme group, antigen (A + B).

    Notes: Sodium alkyl ether sulfate (AES), an analog of sodium dodecyl sulfate (SDS) was used in polyacrylamide gel electrophoresis (PAGE) for the partial decomposition of the photosynthetic reaction center (RC) of Rhodopseudomonas viridis. Unlike SDS, AES did not completely dissociate RC into its subunits but selectively detached H-subunit from RC to give RC(-H) without losing the spectroscopic nature of RC. For the denaturation of RC(-H), AES was found to be as mild as 3-[3-cholamidopropyl dimethylammonio]-1-propanesulfonate (CHAPS).

    Notes: ---In this paper we propose a new post-column detection method for polyols containing 1,2-diol, 1,3-diol, and saccharides. The polyols are oxidized in a photochemical reactor to yield oxalate with subsequent chemiluminescence detection using [Ru(III)(bpy)3]3+. A mixing solution of eluate and oxidizing reagent is delivered to a reaction coil, which is then irradiated with ultraviolet light to promote the oxidation reaction. The detection limits for 1,2-ethanediol (ethylene glycol) and 1,3-propanediol were 38 pmol and 23 pmol, respectively.

    Notes: The permeability (P) of cellulose nitrate (CN)/poly(ethylene glycol) (PEG) blend membranes for N2, O2, and CO2 has been measured as a function of film composition. The system CN/PEG-300 showed excellent miscibility, and films of the composition from 100/0 to 50/50 could be used for permeability measurements. P for each gas has been found to be almost constant or rather slightly lowered up to ca. 20 wt % PEG-300 content and then increased appreciably with increasing fraction of PEG. The increment of permeability was most remarkable for CO2, and hence the permselectivity for CO2 was considerably enhanced. Such a behavior of P has been found to be attributable to the plasticizing effect of PEG molecule lowering the glass transition temperature of the blend polymers. The effect of the molecular weight of PEG and that of closed voids generated in glassy blend membranes fabricated from acetone cast on gas permeabilities have been also discussed.

    Notes: Field desorption mass spectrometry (FDMS) was applied to the study of the initiation mechanism of thermal polymerization of styrenes. The FD mass spectrum was measured for a cooligomer of α,α,β-trideuterostyrene and p-methoxystyrene produced by thermal polymerization using ethylbenzene as a solvent, and this was compared with the spectrum of an oligomer of p-methoxystyrene. The above two FD mass spectra showed characteristic differences, depending upon the number of hydrogen and deuterium atoms and upon the presence or absence of the methoxy group, in the oligomers. Molecular species observed in the spectra were identified based on these considerations. As a result, we could distinguish the “true trimer” produced from three monomers from the “cross trimer” produced from fragments of five monomers. It was found that the “true trimer” was the major constituent in the trimer region, and that the “true trimer” was produced mainly by the cage reaction between initiating radicals.
