    Notes: The optical stability of gossypol [1,1',6,6',7,7'-hexahydroxy-3,3'-dimetyl-5,5'-bis(1-methylethyl)-2,2'-binaphthalene-8,8'-dicarboxaldehyde], a natural product exhibiting profoundly enantiospecific antitumor and male antifertility action, was investigated by means of computational methods and thermal racemization experiments. The calculations on gossypol and several derivatives and model compounds were carried out using the MM2 force field; energies and geometries of minimum energy conformations, as well as structures along various inversion pathways, were calculated. According to the calculations, gossypol (the dialdehyde form) and its simple analogues are not thermally racemizable (energy barriers for rotational inversion above 50 kcal/mol). By contrast, the calculations suggest that the acetal tautomer of gossypol and its dehydration product (anhydrogossypol) are thermally racemizable, although the energy barriers are still relatively high (35-40 kcal/mol). Optically pure (+)-anhydrogossypol was prepared and characterized; its racemization became rapid only at high temperatures (180-200°C). When dehydration of gossypol was hindered (in aqueous solution), no racemization of gossypol was observed after prolonged heating at 90°C. © 1992 Wiley-Liss, Inc.

    Notes: Using crossed affinoimmunoelectrophoresis with free concanavalin A the glycandependent microheterogeneity of the immunoregulatory glycoprotein orosomucoid was examined in serum from 30 healthy individuals, 28 patients with benign chronic inflammation and 56 patients with cancer of various types and stages and before and after surgery. We found significant differences in glycan structure between the different patient groups. Thus the patients with the acute phase reaction of surgery differed from patients with chronic inflammation, and patients with testicular cancer differed from patients with colorectal cancer in having a greater proportion of biantennary glycan orosomucoid. Surgery was found to result in a temporary change in the relative proportions of the three microheterogeneity types of orosomucoid, lasting about one week.

    Notes: Studies of orosomucoid (α1-acid glycoprotein) in human serum have revealed that orosomucoid is a mixture of molecules with differences in the glycan chains. This microheterogeneity has been studied using crossed affinoimmuno-electrophoresis with the lectin concanavalin A which binds to biantennary glycans. The relative proportions of the three orosomucoid subtypes are altered in various pathological conditions independently of the total serum orosomucoid concentration. There are reproducible differences in microheterogeneity patterns between some pathological conditions: Acute tissue injury or inflammation results in a high proportion of orosomucoid with biantennary glycans. Conditions with increased estrogen levels are associated with a high proportion of orosomucoid with tri- or tetraantennary glycans and a low total serum orosomucoid concentration. Chronic inflammation also seems to be associated with a high proportion of orosomucoid with tri- or tetraantennary glycans but with a high total serum concentration of orosomucoid. Other diseases, such as cancer, can not be associated with any specific microhetero-geneity pattern. The microheterogeneity pattern in these conditions seems to be determined by disease activity and unspecific inflammation in surrounding tissues.

    Notes: Crossed immunoelectrophoresis of human serum revealed two heterogeneity types of ceruloplasmin with different electrophoretic migration. The two types both consisted of peptides with Mr 150 000, 100 000 and 45 000, which were interpreted as native ceruloplasmin and two hydrolytic fragments. The two types were different in copper content, and one type could reversibly be changed into the other. The glycan microheterogeneity of ceruloplasmin was analyzed by crossed affinommunoelectrophoresis with free Lens culinaris agglutinin (LCA) and wheat germ agglutinin (WGA). A third of the ceruloplasmin molecules, both high and low copper type, bound to LCA and two thirds to WGA. The heterogeneity and the microheterogeneity of ceruloplasmin in two groups of patient sera were compared to sera from healthy individuals. The ceruloplasmin type with respect to copper content was a much better factor than either glycan microheterogeneity or total serum concentration in discriminating between the three groups.

    Notes: Summary Cyanogen bromide (CNBr) peptides were prepared of the insoluble collagen of bovine dental cementum. Following chromatographic separation, the peptides were identified by their amino-acid composition. Type I collagen ([α1(I)]2α2) accounted for more than 90% of the organic matrix, while Type III collagen ([α1(III)]3) was present at a level of approximately 5%. Amino-acid analyses revealed that the CNBr peptides from α1(I) and α2 chains of cementum closely resembled the corresponding peptides from calf skin. The only systematic difference was a higher level of hydroxylation of prolyl and lysyl residues of the cementum peptides.

    Notes: An investigation has been made of transient heat transfer and water removal on an unfelted cylinder dryer. This investigation has included the development of a theory for describing conduction of heat in the drying material and an experimental testing of the adequacy of the theory.The theory describes the heat transfer and evaporation of water in terms of a second-order partial-differential equation and appropriate boundary conditions. Numerical solutions obtained on a digital computer are presented.The experimental work, performed on a specially constructed laboratory dryer, included measurements of temperatures at internal points in a drying sheet and also measurements of water removed during drying.Good agreement was found between theory and experiment, and the usefulness of the theory is demonstrated in the analysis of water removal in some drying experiments.Although the work was primarily concerned with a description of the paper drying process, the methods should apply equally well to the drying of other materials on heated cylinders.

    Notes: Early turbulence has for the first time been observed in the flow of very dilute polymer solutions in tubes larger than capillary size. Flow rate-wall shear stress measurements were conducted in 0.553 and 0.660 cm. diameter pipes. Polyethylene oxide samples of two molecular weights were employed, dissolved in water-glycerine mixtures with viscosities ranging from 0.0261 to 0.160 poise. The flow rate-wall shear stress relationship for these solutions corresponded to Poiseuille's Law below a well-defined onset wall shear stress, at which the Reynolds number was less than the transition value for Newtonian fluids. As the flow rate was increased from the onset condition, however, the wall shear stress became progessively larger than that predicted by the Poiseuille relationship. The onset wall shear stress for the phenomenon increased linearly with solvent viscosity. Its relationship to polymer concentration was dependent on the solvent viscosity. An explanation for early turbulence is suggested on the basis of these results, and the relationship of the phenomenon to turbulent flow drag reduction is discussed.

    Notes: A theoretical study of high-phase-velocity disturbances in the laminar pipe flow of drag-reducing polymer solutions is reported. These disturbances are found to be more stable in a polymer solution with a sufficiently small relaxation time than in a Newtonian fluid. The opposite effect is predicted when the relaxation time exceeds a critical value, however. These results are shown to be consistent with a hydrodynamic instability explanation of the early turbulence phenomenon.
