Notes: Lipstatin (1), a natural product, and tetrahydrolipstatin (2) are pancreatic lipase inhibitors. Non-stereoselective and partially stereoselective syntheses of 2 are used to establish the absolute configuration of tetrahydrolipstatin and lipstatin.

Notes: 8-Fluoro-6-(methoxymethoxy)quinoline (1) was synthesized, and the reactivity of 1 against organolithium compounds was studied under different reaction conditions. With BuLi, directed ortho-metalation (DoM) was accompanied by 1,2-addition to the C=N bond. 1,2-Addition was exclusively observed with t-BuLi. Selective ortho-metalation was achieved with MeLi (Table). Based on these findings, a short and high-yielding synthesis of the highly functionalized quinolines 12a-c was developed.

Notes: Marginalin [(E)-1 or (Z)-2] is rearranged by bases to the strongly fluorescent 5-hydroxy-2-(4′-hydroxyphenyl)-3-benzo[b]furancarboxylic acid (3) or to methyl 5-hydroxy-2-(4′-hydroxyphenyl)-7-methoxy-3-benzo[b]furancarboxylate (4).

Notes: A method has been developed for quantitative determination of pinaverium-bromide, a quaternary ammonium derivative with papaverine-like activity, in human serum. The method involves a chloroform extraction of serum spiked with N-(6,6-dimethyl bicyclo[3.1.1]2-heptenyl-ethoxyethyl)perhydro-1,4-oxazine as internal standard. After evaporation of the solvent, and reduction of the residue with Raney-Nickel, the internal standard and the reduced pinaverium-bromide are re-extracted from the reaction mixture with toluene and analysed isothermally on a fused silica column coated with OV-101. Although chemical ionization with methane revealed intense protonated molecular ions for both pinaverium-bromide and the internal standard, selectivity and sensitivity were significantly lower in comparison with electron impact ionization at 70 eV. Therefore, quantification was performed in the electron impact mode by single ion monitoring of the common fragment ion at m/z 100.2. A linear detector response was observed up to 160 ng ml-1. A within-run assay precision better than 2% CV (n=5) was found, and a detection limit of 1 ng pinaverium-bromide ml-1 of serum was attained.

Notes: The base-catalyzed reaction of p-hydroxybenzaldehyde (1) with (2,5-dihydroxyphenyl)acetic acid γ-lactone (2) directly gives Z-marginalin (3) with 60% yield. Comparisons of the physical properties with reported data of the substance isolated from the beetle Dytiscus marginalis and of a synthetic product previously obtained through a different method establish the E structure (4) for the natural marginalin.

Notes: New alkyl and allyl complexes 1-3 {1: [Cp′2Sm(C3H5)]n, Cp′ = Me3CC5H4; 2: [Me4C2(C5H4)2]Sm(C3H5)2Li(dme),dme = (CH3OCH2CH2OCH3); 3: Cp′2SmMe2Li(dioxane)} were synthesized from (Cp′2SmCl)2and from the magnesium derivative [Me4C2(C5H4)2]SmCl · MgCl2(THF)4 (4). The ansa anionic complex 2 exhibited good activity for the stereospecific 1,4-trans polymerization of isoprene, whereas the neutral derivative 1 was inactive. In the same way, the anionic complex [Cp′2SmMe2]Li(dioxane) (3) was found to be an ethylene polymerization catalyst of very short lifetime. The lack of reactivity of 1 is related to the associated structure of this coordinatively unsaturated complex: this fact was established by the formation of the carbene adduct Cp′2Sm(C3H5)[C(NiPr)2(CMe)2] (1′). Crystals of 1′ were isolated but this new compound undergoes a partial rearrangement into a tris-Cp′ species in solution. Similar behaviour is observed for the analogous complex Cp′2SmCl[C(NiPr)2(CMe)2] (1′′). The X-ray crystal structure revealed the formation of the adduct, as a toluene solvate, which exists in benzene solution in equilibrium with Cp′3Sm and Cp′SmCl2[C(NiPr)2(CMe)2]2. The catalytic behaviour of 2 is compared with that of other early lanthanide derivatives.

Description / Table of Contents: Measurement of the extracellular fluid volume (ECFV) in the human body is an useful means of recognizing pathological conditions. X-ray fluorescence spectrometry allows a simple and accurate determination of the difusion space of bromide ions, from which the ECFV value can be calculated. The method consists of the quantitative determination of stable bromine (80Br) at low concentration in biological fluids, using the Br Kα line. A simultaneous study run with a radioactive tracer (82Br) allowed us to check the validity of the procedure.

Description / Table of Contents: It can be interesting to determine bromine in a number of biological solutions, although the choice of method is made difficult by the occurrence of other elements. Yet, X-ray fluorescence spectrometric analysis appears suitable under certain conditions. The technique described here is based on the use of a non-dispersive X-ray fluorescence spectrometer, with 109Cd radioactive isotope as an excitation source. The factors studied are the system geometry, the volume of samples, the choice of cells and the reproducibility of measurements. Special conditions, as imposed by the collection of biological samples, are also considered.
