    Notes: Summary During the course of a systematic investigation of the renal corpuscles in various desert rodents (Meriones unguiculati, Meriones shawii, Psammomys obesus, and Dipodymis), a thickened Bowman's capsule was observed light microscopically in some kidneys of adult Meriones unguiculati (Mongolian gerbil). Electron microscopic studies show that this unusual finding may depend on the presence of one or two layers of typical smooth muscle cells adjacent to the outer surface of the basal lamina. In the kidneys of other species of desert rats examined, no pericapsular smooth muscle cells were observed.

    Notes: A novel highly sensitive method for the detection and quantification of proteinase activity is presented. The proteinases were forced electrophoretically into the substrate (sodium caseinate) containing agarose gel. The formation of enzyme-substrate complexes retarded the migration of the proteinases in the electric field and decreased the overall mobility of the enzymes. The proteinases were released as a result of the degradation of the substrate, and consequently the height of the obtained rocket shaped zones of hydrolysis were proportional to the concentration of proteinases in the samples. The electrophoretic method accelerated penetration of the substrate-containing gel by the proteinases and subsequently resulted in a 100-fold increase in sensitivity as compared to the radial diffusion method, depending entirely on diffusion.

    Notes: The development of rocket enzyme activity electrophoresis for the detection and quantification of various proteinases, lipases and pectinases is presented. Rocket enzyme activity electrophoresis is more sensitive than the radial diffusion assay and often enables distinction between qualitatively different enzymes present in the same samples, whereas the radial diffusion assay only provides information on the overall enzyme activity. However, calibration and optimization of the enzyme activity electrophoretic assay have to be performed for each new enzyme-substrate system to be analyzed. Some of the common pitfalls in the development of new enzyme activity electrophoretic assays are presented. Enzyme activity electrophoresis can be applied in combination with other electrophoretic assays. Particularly the combination of enzyme activity electrophoresis with various immunoelectrophoretic methods can provide detailed information on the enzymes studies.

    Notes: A thin-layer counter current line absorption immunoelectrophoresis (TL-CCLAIE) assay was developed for the analysis of antigens and antibodies to Aleutian disease virus. The TL-CCLAIE assay is a rapid and sensitive assay employing 0.4 mm gels cast in moulds in order to allow technicians not routinely engaged in electrophoretic work to perform the assay. Thin gels allow a higher voltage and thus higher electrophoretic mobility of the antigens, making it possible to perform the complete assay within 2 h.

    Notes: The development and application of a rocket immunoelectrophoretic and an enzyme activity electrophoretic assay for the quantitative and qualitative analysis of Geotrichum candidum lipase activity is presented. The sensitivities of the four assays were (in arbitrary units): enzyme activity electrophoresis, 1-0.5; rocket immunoelectrophoresis, 0.5-0.2; radial diffusion, 1; titrimetry, 1. The electrophoretic methods made it possible to distinguish between high and low molecular weight forms of the G. candidum lipases. The enzyme activity electrophoretic methods can be combined with other electrophoretic techniques, as demonstrated here with isoelectric focusing, and produce useful information on physico-chemical differences between different molecular forms of the lipase, e. g. forms with different pI.

    Description / Table of Contents: Jahn-Teller Effect and Crystal Structure Distortion of the Copper Fluoroperovskites NaCuF3 and RbCuF3The crystal structures of triclinic NaCuF3 (a = 539.1, b = 555.2, c = 792.8 pm, α = 90.66, β = 92.05, γ = 86.95°; P1, Z = 4) and of tetragonal RbCuF3, isotypic with KCuF3, (a = 602.3, c = 791.2 pm; I4/mcm, Z = 4), were determined by x-ray methods and refined to wR = 0.038 (2113 reflections) resp. 0.039 (257 reflections). The structures derive from orthorhombic (GdFeO)3 and cubic perovskite type, resp., by Jahn-Teller distortion. The CuF64- octahedra, differently elongated in the two compounds (average NaCuF3: Cu—F = 188.9/197.3/225.6(2) pm; RbCuF3: 188.3/197.8/237.6(3) pm), show the same pattern of antiferrodistortive ordering, but there are bent bridges Cu—F—Cu = 144.2…148.0(1)° in NaCuF3. Structural relations to the prototypes mentioned and to comparably Jahn-Teller distorted copper fluoro compounds are discussed.

Die Strukturen leiten sich vom orthorhombischen (GdFeO3-) bzw. kubischen Perowskit-Typ durch Jahn-Teller-Verzerrung ab. Die in beiden Verbindungen unterschiedlich gestreckten CuF64--Oktaeder (Mittelwert NaCuF3: Cu—F = 188,9/197,3/225,6(2) pm; RbCuF3: 188,3/197,8/237,6(3) pm) sind nach demselben Muster antiferrodistortiv geordnet, jedoch unter Winkelung Cu—F—Cu = 144,2…148,0(1)° in NaCuF3. Strukturelle Beziehungen zu den genannten Prototypen und zu vergleichbar Jahn-Teller-verzerrten Kupfer-Fluorverbindungen werden diskutiert.

    Notes: A cellulose-binding endoglucanase (endoglucanase A) of Cellulomonas sp. ATCC 21399 was purified to immunological homogeneity by affinity chromatography ob H3PO4-swollen cellulose. This method of purification turned out to be an easy and very gentle method for obtaining a high yield of cellulose-binding endoglucanase. The purified enzyme was immunologically homogeneous but appeared heterogeneous when analyzed by denaturing polyacrylamide gel electrophoresis. In addition to the cellulose-binding of endoglucanase A, the enzyme also had a strong affinity for Concanavaline A, indicating that the enzyme was glycosylated. Purified endoglucanase A showed an endo mode of action on carboxymethylcellulose. The enzyme could hydrolyze microcrystalline cellulose when acting alone, and the enzyme had a high specific activity on H3PO4-swollen cellulose.

    Notes: A novel electrophoretic method for detecting proteinase activity was used in order to study the column chromatographic separation of different acid, neutral and alkaline proteinases of a commercial crude proteinase preparation of Aspergillus oryzae (Rhozyme 41). Fractions obtained by gel filtration of Rhozyme 41 on Sephadex G-100 were analyzed by the electrophoretic method and the radial diffusion method at pH 4.6 and pH 8.6. The electrophoretic method made it possible to discriminate between various acid, neutral and alkaline proteinases due to differences in the morphology of the clearing zones, whereas no differences in the morphology of the clearing zones could be observed when using the radial diffusion method.
