    Beschreibung: The most abundant mRNA post-transcriptional modification is N(6)-methyladenosine (m(6)A), which has broad roles in RNA biology. In mammalian cells, the asymmetric distribution of m(6)A along mRNAs results in relatively less methylation in the 5' untranslated region (5'UTR) compared to other regions. However, whether and how 5'UTR methylation is regulated is poorly understood. Despite the crucial role of the 5'UTR in translation initiation, very little is known about whether m(6)A modification influences mRNA translation. Here we show that in response to heat shock stress, certain adenosines within the 5'UTR of newly transcribed mRNAs are preferentially methylated. We find that the dynamic 5'UTR methylation is a result of stress-induced nuclear localization of YTHDF2, a well-characterized m(6)A 'reader'. Upon heat shock stress, the nuclear YTHDF2 preserves 5'UTR methylation of stress-induced transcripts by limiting the m(6)A 'eraser' FTO from demethylation. Remarkably, the increased 5'UTR methylation in the form of m(6)A promotes cap-independent translation initiation, providing a mechanism for selective mRNA translation under heat shock stress. Using Hsp70 mRNA as an example, we demonstrate that a single m(6)A modification site in the 5'UTR enables translation initiation independent of the 5' end N(7)-methylguanosine cap.     Notizen: In industrial recycling of waste plastics the on-line sorting of different kinds of consumer plastics often represents the key to the successful commercial operation. The principle and a limited feasibility study of a novel wavelength independent photoacoustic sensor applied to this problem are presented.     Notizen: The photodegradation of polyesters prepared by the reaction of o-phthalic, adipic, and “dimer” acids with ethylene glycol has been studied. The polyesters softened at temperatures in the range of 25-50°C. Polymer films were irradiated in air by means of a medium pressure mercury lamp and the extent of formation of COOH groups determined by infrared spectroscopy. The films were also irradiated in vacuo and in air by means of a high-pressure mercury lamp, and the volatile products, consisting mainly of carbon monoxide and carbon dioxide, were determined by gas chromatography.The results are interpreted in terms of simultaneous Norrish type I and type II decompositions of the polyesters. The type I process accounts for the formation of carbon monoxide and carbon dioxide in the absence of air. The enhanced yield of carbon dioxide in irradiations performed in air is ascribed to the formation of hydroperoxide which undergoes further photolysis. The type II process accounts for the formation of COOH groups. The photodegradation of these glassy polyesters parallels that of crystalline poly(ethylene terephthalate) and is relatively unaffected by changes in composition of the aromatic polyester.

    Notizen: The photochemistry of p-fluoranil in dioxane was studied by electron spin resonance (ESR) and the ESR-rotating sector technique. The transient photoradical is identified as the p-tetrafluorobenzosemiquinone neutral radical with a hyperfine splitting of 1.1 gauss for the hydroxy proton and the fluorine hyperfine splittings of 3.8 and 14.1 gauss for the meta and ortho fluorines, respectively. The radicals decayed by self-disproportionation with a second-order rate constant at room temperature of approximately 3.2 × 109M-1s-1. The activation energy of the decay process is found to be about 2.4 ± 0.4 kcal/mole.
