Notes: Six enzymes associated with the activities of the nucleus (thymidine kinase), mitochondria (succinate dehydrogenase), lysosomes (acid phosphatase), peroxisomes (catalase), (cytosol lactate dehydrogenase), and the intermembranal mitochondrial space (alkaline DNase) were assayed at 2 hr intervals over the division cycle of repetitively resynchronized HeLa cells. The results indicated a high degree of reproductibility for cells synchronized by the method of perpetual resynchronization and may be of direct use to those interested in subcellular organellogenesis.

Notes: Unbalanced growth has been studied in HeLa cell cultures maintained in perpetual division synchrony by periodic inhibition of DNA synthesis. Observation of eight independent cellular parameters indicates that once synchrony has been established the degree of unbalanced growth in later cycles is not as great as is observed with the conventional “double-block” technique. This significant diminution of unbalanced growth appears to be related to the significantly shorter periods of inhibition of DNA synthesis used in the resynchronization process.

Notes: A variety of diploid human fibroblast lines have been successfully grown to high densities (〉106 cell/ml) on recently developed microcarriers. Interferon induction using poly I·poly C and a superinduction procedure resulted in yields greater than 10,000 units/ml with one cell line. A direct comparison of microcarrier cultures to roller bottle cultures showed equivalent interferon yields on a per cell basis and some apparent differences relating to optimum inducer concentrations and kinetics of interferon accumulation.

Description / Table of Contents: The Use of Titanium Castings to Produce a Complex Shaped Intermediate Casing of RB 199.This paper describes the present position on a highly stressed casing and its functions in the RB 199-34 R engine. When the parts of the casing are made from solid forged titanium billets, up to 93% of the material used is machined away. For this reason, the most complicated parts of the casing were procured as castings and welded together by TIG welding. The paper describes the present quality standard of the titanium castings. The tests on the materials and their mechanical properties show that the castings are inferior to the forgings only with respect to their HCF behaviour and their elongation at rupture. Macroscopic flaws are not entirely inevitable but can be detected by X-ray tests. The future prospects for titanium castings are considered favourable.

Notes: Two new sample preparation procedures that produce solvent-free extracts especially suitable for use with high resolution GC are described. One is based on Soxhlet extraction in a pressurized container that permits the use of liquid carbon dioxide as the extractant; the other utilizes refluxing difluorodichloro-methane (Freon 12) to isolate entrained volatiles from gas streams.

Notes: The electronic properties of a variety of methylated isoalloxazine derivatives have been investigated with absorption and fluorescece techniques (Table 1). The spectral effects of methyl substitution at different positions in the isoalloxazine ring are interpreted in terms of different bond orders between the C-atoms of the methyl groups and of the aromatic ring. Methyl groups at C(6) or C(9) induce quenching of the fluorescence and shortening of the lifetime whereas a methyl group at C(7) has an opposite effect. The quenching effect might be due to strain in the molecular framework caused by the methyl groups. The lowest energy levels of the isoalloxazine cations are shifted to higher energy as compared to the neutral isoalloxazines. Apart from these shifts the trends in spectral behaviour upon methylation are similar to those observed for the neutral isoalloxazine (cf. Table 2). The fluorescence of the isoalloxazine cations is strongly quenched in auqeous solution and their lifetime much shortened. These observations indicated that there is substantial interaction between the charged fluorophore and the water dipoles and that this interaction produces efficient radiationless deactivation of the first excited singlet state.

Notes: Conformational transitions induced by pH changes in random copolymers of leucine and glutamic acid have been studied. Significant differences were observed in the potentiometric titration curves of copolymers with small (up to 4%) and large leucine contents. The helical stability of copolymers with small leucine content, determined from titration curves by the Zimm and Rice method, decreases slightly with an increase in the leucine content, whereas the helical stability of copolymers with large leucine content increases sharply with an increase of the leucine content. It is shown that copolymers with large leucine content aggregate in the region of transition into the helical state, but the increase of their helical state stability is not connected with intermolecular aggregation, as it was also observed for a nonaggregating fraction isolated from one of the copolymers by gel chromatography. A conclusion is made that the helix-coil equilibrium constant s for leucine does not itself exceed the s constant for uncharged polyglutamic acid. The stabilization of the helical state in copolymers with large leucine content is due to intramolecular aggregation of helices in these copolymers. The analysis of the leucine residue distribution between helical and nonhelical regions in globular proteins also gives no real arguments to ascribe special helix-forming properties to leucine.

Notes: Filaments of ultra-oriented high density polyethylene were solid-state extruded at an extrusion (draw) ratio of 26 and subsequently irradiated under vacuum by a cobalt-60 source at doses of 10,15, 20, 40 and 60 megarads (MRad). Several identically prepared but unirradiated strands were also tested. One set of samples at each dose was immersed in a silicone oil bath for one half hour at 128°C and one set was given no post-irradiation thermal treatment. Characterization of the resultant morphologies included differential scanning calorimetry (DSC), birefringence, thermomechanical analysis (TMA), and tensile testing. Results from DSC measurements indicate that initial radiation crosslinking only slightly disrupts the crystal lattice, but on subsequent melting and re crystallization, the chains are unable to recrystallize effectively in their former habit. In all cases, melting point and crystallinity decrease with increasing radiation dose. Birefringence and TMA results indicate that orientation is not disrupted by irradiation. For unannealed samples, Young's modulus increases slightly then levels off while tensile strength and elongation at break increase initially, then drop, For annealed irradiated samples, Young's modulus rises at first, then levels off at higher doses. Tensile strength and elongation at break increase significantly with increasing radiation treatment, then essentially reach constant values at highest doses.

Notes: A number of phosphoryl solvates and adducts of beryllium(II) tetrafluoroborate and nitrate is reported. The infrared and Raman spectra of these complexes have been investigated. The anion vibrations in both solvates and adducts are assigned and discussed. The beryllium-oxygen stretching vibrations of the solvates are assigned and compared to those reported for other complexes. An assignment for the more complicated adduct spectra is attempted.
