    Notizen: A strong positive nonlinear effect (asymmetric amplification) was found to take place during asymmetric hydrogenations using chiral atropisomeric diphosphane-ruthenium catalysts. As an example, at atmospheric pressure the use of 50% ee BINAP to prepare [(binap)Ru(Br)2] give rise to a hydrogenated product with 91% ee. The influence of temperature and hydrogen pressure on this effect are presented. These nonlinear effects can be explained on the basis of a hydrogenation mechanism in which diastereomeric dimers as pre-catalytic species are present.

    Notizen: The miscibility of polyetherimides (PEIs) with epoxy monomers based on diglycidylether of bisphenol-A (DGEBA), and with reactive mixtures based on stoichiometric amounts of DGEBA and an aromatic diamine (DA) {either 4,4′-diaminodiphenylsulfone (DDS) or 4,4′-methylenebis[3-chloro 2,6-diethylaniline] (MCDEA)}; was experimentally studied. Cloud-point curves (temperature vs. composition) are reported for PEI-DGEBA and PEI-DGEBA-DA initial mixtures. Cloud-point conversions are reported for the reactive mixtures, for various PEI amounts and polycondensation temperatures. A thermodynamic model based on the Flory-Huggins-Staverman approach, taking polydispersity of both components into account, was used to analyze the experimental information. A single relationship between the interaction parameter and temperature, χ(T), could fit experimental results of mixtures of two commercial PEIs with DGEBA. The addition of DDS led to a decrease in miscibility whereas MCDEA improved the initial miscibility. In both cases, the interaction parameter decreased with conversion, meaning that PEI was more compatible with oligomeric species than with the mixture of starting monomers. The phase separation process in initially miscible rubber- or thermoplastic-modified thermosetting polymers is the result of two factors: increase in the average molar size of the thermosetting oligomer (main driving force favoring demixing), and variation of the interaction parameter with conversion, which may act to increase or decrease the cloud-point conversion determined by the first factor. © 1996 John Wiley & Sons, Inc.

    Notizen: Selectivity for polychlorinated biphenyl (PCB) congeners was examined for a series of cyanobiphenyl stationary phases, and the results were compared to a nonpolar 5% phenyl methylpolysiloxane phase and a shape-selective smectic liquid crystalline phase. For all of the columns studied, the degree of ortho substitution of the congener was found to be a significant factor controlling chromatographic retention of PCBs. The retention pattern on six different cyanobiphenyl phases followed the same general pattern as on the liquid crystalline column, though the trends were more exaggerated on the liquid crystalline phase. While the general retention pattern was similar for all of the cyanobiphenyl phases studied, subtle differences were observed, resulting in significant selectivity differences. For isomeric planar PCB congeners, elution order could be correlated to length-to-breadth ratios. Among the cyanobiphenyl phases, the p,p- and the o,p,p-cyanobiphenyl phases had the greatest enhancement of retention for planar congeners. A shapeselective test mixture reflected the similarity of retention on the cyanobiphenyl phases to both the 5% phenyl methylpolysiloxane phase and the smectic liquid crystalline phase. © 1995 John Wiley & Sons, Inc.

    Notizen: Raman scattering is used to make a dynamical study of the disubstituted and perdeuterated diacetylene pTS-D in monomer crystal from, where an incommensurate phase exists from T1 = 155 K to T1 = 195 K. The low-frequency polarised Raman spectra in the temperature interval 9-300 K show that the phase transition is related to side-group rotations decoupled as from the diacetylene backbone, as in pTS-D polymer. A comparison with another disubstituted diacetylene DNP shows that it is very difficult to obtain symmetrical diacetylene polymers in a non-centrosymmetric structure.

    Notizen: A novel p,p-cyanobiphenyl stationary phase (p-cyanobiphenyl, p-allyloxy methylpolysiloxane) has been evaluated for the GC investigation of polychlorinated biphenyls (PCBs). Several PCB congeners which coelute on the phases typically used for PCB analysis (e.g. 5% phenyl methylpolysiloxane) are separated on the p,p-cyanobiphenyl phase, including the hexachlorobiphenyl congeners PCB 163, PCB 164, and PCB 138. In this work, a p,p-cyanobiphenyl stationary phase was used to measure selected PCB congeners in two Standard Reference Materials (SRMs) available from the National Institute of Standards and Technology (NIST), and two Certified Reference Materials (CRMs) available from the Community Bureau of Reference (BCR). The materials analyzed were SRM 1588, Organics in Cod Liver Oil; SRM 1945, Organics in Whale Blubber; CRM 349, Chlorobiphenyls in Cod Liver Oil; and CRM 350, Chlorobiphenyls in Mackerel Oil. Concentrations are reported for several PCB congeners which coelute on the 5% phenyl methylpolysiloxane, including PCB 163 and PCB 164.

    Notizen: The present study was undertaken to evaluate a new prototype mesh that consists of a knitted polyester structure treated with a fluoropolymer and impregnated with gelatin. The Fluoropassiv® mesh, as well as two controls, the Surgipro™ polypropylene mesh and the Gore-Tex® expanded polytetrafluoroethylene patch, were used for the repair of experimentally induced abdominal hernias in piglets and followed for scheduled implantation periods of 4, 15, and 60 days. At the sacrifice the mesh and surrounding tissue were excised for histological assessment of the healing sequence, for the identification of changes in hematologic and immunological characteristics, and for the measurement of the mechanical properties. After cleaning to remove the encroaching tissue, the explanted devices were monitored for biostability by infrared spectroscopy (FTIR) and differential scanning calorimetry (DSC). The present study has demonstrated that the Fluoropassiv® mesh provides adequate mechanical strength and compares favorably with the two controls. No exacerbated systemic or in situ hematologic or immunological reactions were observed with either the meshes or the patch material. Histological studies revealed that thick collagenous and vascularized tissue were well anchored to the three biomaterials as early as 15 days after implantation. The degree of tissue penetration differed depending on the device. Chemically, they proved stable over time. © 1996 John Wiley & Sons, Inc.

    Notizen: The surgical management of abdominal hernias requires prosthetic grafting in situations where the defect is too large or the surrounding tissue is not available for repair. Flat patches made of different biomaterials have been used in textile or microporous forms. The present work describes the results of an in vitro study comparing the morphological, mechanical, and chemical characteristics of a new textile prototype, Fluoropassiv®, made of polyester fibers treated with a fluoropolymer and impregnated with gelatin to those of seven existing commercial meshes and patches made from polypropylene, polyester, polytetrafluoroethylene (PTFE) yarns, and expanded microporous PTFE graft. The morphological study revealed a diversity of structures having a minimal relative porosity of 70%, high bursting, and suture retention strengths in comparison with natural muscular tissue. Elasticmoduli proved to depend more on the direction of the textile: the rigidity was higher for those materials having tight structure, like the Fluoropassiv® and the Surgipro™ meshes (〉30 MPa), whereas those with more open structures, such as the Marlex®, Trelex®, Lars®, Bard Teflon®, and Gore-Tex® structures, showed lower elastic modulus (10 mPa). In addition, chemical analyses confirmed no irregularities in the polymers used in all prostheses and demonstrated that the fluoropolymer coating of the Fluoropassiv® was uniformly distributed. The innovative aspects in the construction of the knitted fabric Fluoropassiv® appears to make it suitable for repairing hernias, and the inclusion of both continuous fluoropolymer surface treatment of polyester fibers and gelatin impregnation appears to improve the healing process. © 1996 John Wiley & Sons, Inc.

    Notizen: Polymerase chain reaction (PCR)-based DNA typing is routinely used in forensics for identity testing. Those assays that distinguish single nucleotide polymorphisms (SNPs) require other biochemical reactions in addition to PCR to identify the sequence polymorphisms. Low-stringency sequence-specific PCR (LSSP-PCR) is an example of a recent method that does not require additional biochemical treatments. The analysis of LSSP-PCR by capillary electrophoresis (CE) to discriminate the highly polymorphic mitochondrial DNA (mtDNA) D-loop region is described. The DNA from five individuals were amplified (first step) using sequence-specific primers to produce 1021 bp fragments containing the D-loop region. Each fragment was isolated by electroelution using CE and UV detection, and subjected to a second amplification (second step) using a single primer annealed under low stringency conditions. This generated a range or profile of PCR products for each sample, which were resolved and analyzed by CE with the intercalator TOTO-1 and laser-induced fluorescence (LIF) detection. The LSSP-PCR profiles were unique for each individual, indicating that this technique may be applicable for forensic identity testing.

    Notizen: Polymerase chain reaction (PCR) amplified alleles need to be isolated and purified before carrying out additional analysis to confirm sequence, number of repeats and microvariants within a short tandem repeat (STR) locus. Also, PCR amplification of tetranucleotide repeat loci, used in DNA typing assays, often result in heteroduplex formation, adding to the complexity of analysis. Sequencing reactions require single specific target DNA for reliable sequencing analysis. Alkylated poly(styrene-divinylbenzene) columns at elevated temperature and gradient elution conditions increase the efficiency of separation to allow for the purification of PCR products. Using the separation technique of ion-paring reverse-phase (IPRP) high performance liquid chromatography (HPLC), molecular biologists can separate and purify DNA fragments without alteration to the double-stranded DNA sequencing properties. In this study, the IP-RP chromatography technique has been demonstrated by separation of alleles of the short tandem repeat loci of TH01, vWA31, F13A01 and FES/FPS. Alleles differing in size range of 12 to 4 base pairs were separated by IPRP/HPLC and individual alleles were peak-captured, then cycle-sequenced. These HPLC fractions required no additional steps prior to cycle sequencing. Capillary electrophoresis (CE) was used to sequence the alleles. Furthermore, CE offers advantages over traditional slab methods via automation and higher applied voltages. Interestingly, unlike traditional gel electrophoresis, samples were introduced into the sieving matrix by electrokinetic injection, which allows for multiple injections from a single sample, a key feature for method development. Applied voltage was 320 V per centimeter using a nonderivatized fused silica capillary with an interior diameter of 50 μm and a total length of 47 centimeters. The total analysis time including capillary filling and pre-electrophoresis was less than 30 min for a 220-bp fragment. A sequencing rate of 530 bp/h was achieved using these conditions. By combining the techniques of HPLC separation and CE sequencing, the results confirmed the sequence and number of nucleotide repeats for each STR loci. An average sequencing efficiency of 97% was achieved. Additionally, this method defined the absence of a 9.3 microvariant for a TH01 heterozygous individual previously typed as a 9,9.3/10 using slab gel electrophoresis. The techniques described can be applied to other DNA purification and isolation problems.
