Notes: Protamine sulfate blocked 125I-PDGF binding to its specific physiological receptor on Swiss mouse 3T3 cells. Reduced 125I-PDGF binding in the presence of protamine sulfate correlated directly with a protamine sulfate dose-dependent decrease in the PDGF-dependent incorporation of [3H]-thymidine into 3T3 cells and a decreased PDGF-stimulated tyrosine-specific protein kinase activity in isolated membrane preparations of 3T3 cells. Protamine sulfate blocked 125I-PDGF binding to simian sarcoma virus transformed cells (SSV-NIH 3T3 and SSV-NPl cells) and to nontransformed cells in a manner qualitatively identical to unlabelled PDGF. In contrast, protamine sulfate enhanced the specific binding of 125I-EGF by increasing the apparent number of EGF receptors on the cell surface. The increase in 125I-EGF receptor binding was not prevented by cycloheximide nor by actinomycin D. Protamine sulfate did not affect 125I-EGF binding to membranes from 3T3 cells or the EGF-stimulated 3T3 cell membrane tyrbsinc specific protein kinase activity, suggesting that protamine sulfate may have exposed a population of cryptic EGF receptors otherwise not accessible. Protamine sulfate was fractionated into four active fractions by Sephadex G-50 gel filtration columns; the half maximum inhibition concentration of 125I-PDGF binding to 3T3 cells of protamines I and II (MW ∼ 11,000 daltons and 7,000 daltons, respectively) is ∼ 0.4 μM. Protamine II (MW ∼ 4,800 daltons) was equally active (half maximum inhibition concentration ∼ 0.4 μM); protamine IV (MW ∼ 3,300 daltons) was substantially less active (half maximum inhibition concentration ∼ 2.8 μM).These investigations have extended previous observations that protamine sulfate is a potent inhibitor of PDGF binding and establish that protamine sulfate blocks PDGF binding at the physiological receptor, preventing PDGF initiated biological activities. Protamine sulfate can be used as a reagent to separate the influence of PDGF and EGF on cells with high specificity and has been used to demonstrate that the receptors on simian sarcoma virus transformed 3T3 cells qualitatively respond identically to protamine sulfate as to unlabelled PDGF and are likely identical to those on nontransformed 3T3 cells.

Notes: Trimethylsilyl-di-n-butylamine has been used to amidate methyl n-propionate and polymethylmethacrylate (PMMA) in THF. Trimethylsilyl (N-phenyl-p-phenyleneamine) amine was also prepared and used to amidate PMMMA and poly(ethylene-co-propylene)-graft-methylmethacrylate (OCP-graft-MMA) in THF and n-hexane, respectively. Amidations were performed between 55 and 65°C. When trimethylsilyl-di-n-butylamine was reacted with methyl propionate, less than 10% of the corresponding amide was generated because of a competitive aldol condensation. PMMA was smoothly amidated in excess of 60% using either trimethylsilyl(N-phenyl-p-phenyleneamine)amine or trimethylsilyl-di-n-butylamine without detectable competitive side reactions. OCP-graft-MMA was amidated in excess of 90% using a 5 M excess of trimethylsilyl(N-phenyl-p-phenyleneamine)amine in n-hexane.

Notes: Two new acyltetramic acids related to streptolydigin have been isolated from fermentations of Streptomyces lydicus. The principal members of this complex were resolved by TLC on silica gel. However, the methods of detection, permanganate spray or bioautography, were not suitable for both crude fermentation broths and purified extracts. Gas chromatography is unsuitable for the detection of either underivatized or silylated streptolydigins. High performance liquid chromatography (HPLC) particularly on triethylaminoethyl cellulose is rapid and sensitive and is the method of choice for the analysis of both crude and purified samples. Using high performance liquid chromatography, two components were detected in the complex, which are not observed using any of the other chromatographic procedures.

Notes: Quantitative methods of evaluating ingrowth into porous coated implants have traditionally involved destructive mechanical testing. In this study, it was hypothesized that the response to a vibrational stimulus could estimate the shear strength at the bone-implant interface as measured by pushout. A derived model predicts that the square of the frequency at resonance is linearly related to this interfacial strength. It was further hypothesized that the level of vibration required for a meaningful response would not be destructive. This study was done using nonloaded, porous coated cylindrical implants which were press fit in the distal femora of dogs. Femora were harvested at 2, 4, and 6 weeks postimplantation. The implants were sectioned into 4-mm slices and then evaluated by vibration, pushout, and light microscopy. Vibrational response analysis was conducted with broad band random excitation using Fast Fourier transform signal analysis. Pushout values were obtained using a materials testing machine. The resulting correlation was statistically significant (F = 201, r2 = 0.90, p 〈 10-6). Light microscopic evaluation of vibrated samples showed no difference at the bone-implant interface when compared to controls.

Notes: This new coupling agent shows promise for use in reinforcing hydrocarbon type polymers, such as polyolefins and polystyrene, with glass fibers and inexpensive mineral fillers. Sulfonyl azide derivatives readily form durable bonds to many thermoplastics at normal extrusion and molding temperatures, without the use of peroxide or sulfur-type curing agents. Alkoxy silanes, attached to the sulfonyl azide, hydrolyze in the known manner forming bonds to inorganic fillers. Using commercial compounding and injection molding procedures, 40 wt percent of mineral fillers containing 1 percentofan azidosilane coupling agent were incorporated in polypropylene. Very substantial improvements in tensile, flexural and heat distortion properties were obtained with little loss in impact strength. An initial evaluation in a filled polystyrene showed significant, but less dramatic, improvements.

Notes: The complex and conspicuous basket-like structure attached to the third segment of each maxillary palp of Melittomma sericeum males is densely covered with tactile hairs on its outer or convex surface and with thinwalled chemoreceptors on its inner or concave surface. In a living male the structure is highly mobile and is extended laterally and ventrally. It evidently serves to detect odors produced by the female.
